Four Unrecorded Aspergillus Species from the Rhizosphere Soil in South Korea.

ABSTRACT: The genus Aspergillus is commonly isolated from various marine and terrestrial environments; however, only a few species have been studied in rhizosphere soil. As part of the Korean indigenous fungal excavation project, we investigated fungal diversity from rhizosphere soil, focusing on Aspergillus species. A total of 13 strains were isolated from the rhizosphere soil of three different plants. Based on phylogenetic analysis of β-tubulin and calmodulin and morphological characteristics, we identified five Aspergillus species. A. calidoustus and A. pseudodeflectus were commonly isolated from the rhizosphere soil. Four species were confirmed as unrecorded species in Korea: A. calidoustus, A. dimorphicus, A. germanicus, and A. pseudodeflecuts. The detailed morphological descriptions of these unrecorded species are provided.

Project description:BackgroundUlleungdo harbours a unique ecosystem owing to its isolation from the mainland alongside its maritime climate. The island, formed via volcanic activity, is the largest island in the East Sea of Korea and retains a primeval forest. The ecosystems are being destroyed owing to increasing human activity on the island. Therefore, through the investigation of the insect fauna of Ulleungdo, we tried to provide information that can be the basis for understanding the island ecology of Ulleungdo. This survey was conducted four times between April and October in 2020 at Seonginbong.New informationThe findings of the survey regarding insect fauna at Seonginbong, Ulleungdo included 10 orders, 105 families, 216 genera and 212 species, of which 12 families, two subfamilies, 13 genera and 74 species were previously unrecorded. The data have been registered in the Global Biodiversity Information Facility (GBIF; www.GBIF.org).

Isolation and Characterization of Four Unrecorded Mucor Species in Korea.

Project description:During an investigation of fungi of the order Mucorales from freshwater and sediment samples in Korea, we isolated six strains, NNIBRFG6649, NNIBRFG6255, NNIBRFG1498, CNUFC-YJ13, CNUFC-YR7, and NNIBRFG2739. The morphology and phylogeny of these strains were analyzed. Based on the morphological characteristics and molecular data from internal transcribed spacer (ITS) region, the isolates NNIBRFG6649 and NNIBRFG6255 were identified as Mucor abundans, and M. aligarensis, respectively. The isolates NNIBRFG1498 and CNUFC-YJ13 were identified as M. moelleri, whereas the isolates CNUFC-YR7 and NNIBRFG2739 were identified as M. heterogamus. To the best of our knowledge, M. abundans, M. aligarensis, M. moelleri, and M. heterogamus have not yet been reported in Korea.

Five Previously Unrecorded Fungal Species Isolated from Marine Plastic Wastes in South Korea.

Project description:Plastic wastes have a negative impact on marine environments; however, they can be used as carbon sources and habitats by certain microbes. Microbes in the marine plastisphere can migrate worldwide through the ocean and cause serious environmental problems when they encounter suitable environments. Therefore, efforts to investigate the microbes inhabiting the marine plastisphere are increasing. In the present study, fungal strains were isolated from plastic wastes buried in Korean sea sands and mudflats and identified using molecular and morphological analyses. Five species were identified that were previously unrecorded from South Korea: Cladosporium funiculosum, Neosetophoma poaceicola, Neosetophoma rosigena, Parasarocladium gamsii, and Trichoderma fomiticola. Their molecular phylogenies and morphological characteristics are described in this study.

Four unrecorded wood decay fungi from seoul in Korea.

Project description:During the investigation of Korean indigenous fungi from Seoul, three genera-Fuscoporia, Porostereum, and Trametopsis, and four species-Fuscoporia senex, Phlebia acerina, Porostereum spadiceum, and Trametopsis cervina were found. Their morphological characteristics were examined and their identification was confirmed by molecular analysis based on internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA region sequences. These fungi are new to Korea and registered here with descriptions.

Characterization of Two Species of Acremonium (Unrecorded in Korea) from Soil Samples: A. variecolor and A. persicinum.

Project description:During a survey of fungal diversity of the order Hypocreales in Korea, two Acremonium isolates, CNUFC-1YSRS2-4 and CNUFC-GSNPF3-1, were isolated from soils collected on a bank of the Yeongsan River, Naju, and in a forest on the Mt. Daegak located on Sinsi Island, Gunsan, South Korea, respectively. Based on the morphological characteristics and sequence analysis of the internal transcribed spacer and D1/D2 domains of 28S ribosomal DNA, the isolates CNUFC-1YSRS2-4 and CNUFC-GSNPF3-1 were identified as A. variecolor and A. persicinum, respectively. These 2 species represent novel Hypocreales isolates in Korea.

Seventeen Unrecorded Species from Gayasan National Park in Korea.

Project description:Macrofungi play important roles in forest ecology as wood decayers, symbionts, and pathogens of living trees. For the effective forest management, it is imperative to have a comprehensive overview of macrofungi diversity in specific areas. As a part of the National Institute of Biological Resources projects for discovering indigenous fungi in Korea, we collected macrofungi in Gayasan National Park from 2017 to 2018. These specimens were identified based on morphological characteristics and sequence analysis of internal transcribed spacer (ITS) or the nuclear large subunit rRNA (LSU) region. We discovered 17 macrofungi new to Korea: Butyrea japonica, Ceriporia nanlingensis, Coltricia weii, Coltriciella subglobosa, Crepidotus crocophyllus, Cylindrobasidium laeve, Fulvoderma scaurum, Laetiporus cremeiporus, Lentinellus castoreus, Leucogyrophana mollusca, Marasmius insolitus, Nidularia deformis, Phaeophlebiopsis peniophoroides, Phanerochaete angustocystidiata, Phlebiopsis pilatii, Postia coeruleivirens, and Tengioboletus fujianensis. We described their detailed morphological characteristics.

Penicillium from Rhizosphere Soil in Terrestrial and Coastal Environments in South Korea.

Project description:Penicillium, the most common genus plays an important ecological role in various terrestrial and marine environments. However, only a few species have been reported from rhizosphere soil. As part of a project to excavate Korean indigenous fungi, we investigated rhizosphere soil of six plants in the forest (terrestrial habitat) and sand dunes (coastal habitat) and focused on discovering Penicillium species. A total of 64 strains were isolated and identified as 26 Penicillium species in nine sections based on morphological characteristics and the sequence analysis of β-tubulin and calmodulin. Although this is a small-scale study in a limited rhizosphere soil, eight unrecorded species and four potential new species have been identified. In addition, most Penicillium species from rhizosphere soil were unique to each plant. Penicillium halotolerans, P. scabrosum, P. samsonianum, P. jejuense, and P. janczewskii were commonly isolated from rhizosphere soil. Eight Penicillium species, P. aurantioviolaceum, P. bissettii, P. cairnsense, P. halotolerans, P. kananaskense, P. ortum, P. radiatolobatum, and P. verhagenii were recorded for the first time in Korea. Here, we provide the detailed morphological description of these unrecorded species.

Four new species of isopods (Crustacea, Isopoda) from South Korea.

Project description:Four new isopods, Amakusanthura intermedia sp. nov., Apanthura laevipedata sp. nov., Idarcturus trispinosus sp. nov., and Neastacilla paralongipectus sp. nov., are reported from the sublittoral zones in Korean waters. Amakusanthura intermedia sp. nov. differs from its congeners by the following features: the uropodal exopod is sinuous distally and with pointed apex; the maxillipedal endite is present and reaching to the distal end of fused palp articles I and II; and the propodal palm of pereopod I is stepped. Apanthura laevipedata sp. nov. can be distinguishable from its related species by the following characteristics: the eye is lacking; the propodal palm of pereopod I is not stepped; and the uropodal exopod is not sinuous. Idarcturus trispinosus sp. nov. is diagnosed by the following features: the cephalon has three dorsal spines and a pair of lateral spines; pereonite IV has two pairs of small dorsolateral spines, four pairs of dorsal spines, and one posterior spine; and the pleotelson has three pairs of wings laterally. Neastacilla paralongipectus sp. nov. can be distinguished by the following characteristics: the body is smooth and lacking dorsal spines or tubercles; pereonite IV is approximately 5.4× longer than pereonites II and III together; and the pleotelson has two pairs of lateral wings. In this paper, detailed descriptions and illustrations of the four species are presented. A key to the genera of the family Arcturidae and keys to the species of the four genera are also provided.

Re-Identification of Aspergillus Subgenus Circumdati Strains in Korea Led to the Discovery of Three Unrecorded Species.

Project description:Aspergillus is one of the largest and diverse genera of fungi with huge economical, biotechnological, and social significance. Taxonomically, Aspergillus is divided into six subgenera comprising 27 sections. In this study, 235 strains of Aspergillus subgenus Circumdati (section: Candidi, Circumdati, Flavi, Flavipedes, Nigri, and Terrei) preserved at the Korean Agricultural Culture Collection (KACC) were analyzed and re-identified using a combined dataset of partial β-tubulin (BenA), Calmodulin (CaM) gene sequences and morphological data. We confirmed nineteen species to be priorly reported in Korea (A. neotritici, A. terreus, A. floccosus, A. allahabadii, A. steynii, A. westerdijkiae, A. ochraceus, A. ostianus, A. sclerotiorum, A. luchuensis, A. tubingensis, A. niger, A. welwitschiae, A. japonicus, A. nomius, A. tamarii, A. parasiticus, A. flavi, and A. oryzae). Among the studied strains, three species (A. subalbidus, A. iizukae, and A. uvarum), previously unreported or not officially documented, were discovered in Korea, to the best of our knowledge. We have given a detailed description of the characteristic features of the three species, which remain uncharted in Korea.

Two Previously Unrecorded Fungal Species Isolated from Muui Island in Korea.

Project description:Fungi are cosmopolitan and they occupy diverse niches as consumers, producers, and decomposers. They play critical roles in the environment by enabling nutrient cycling and generating a plethora of secondary metabolites. This study aimed to identify and characterize fungal strains isolated from diverse sources on Muui Island, Republic of Korea. In 2023, a total of 86 fungal strains were collected and examined. Investigation of the morphological features and phylogenetic analyses of multiple barcode loci identified one putative novel species and two species previously unrecorded in the Republic of Korea: Colletotrichum sp., Colletotrichum guizhouense, and Fusarium brachygibbosum. This study provides a comprehensive description of their molecular phylogenies and morphological characteristics. These findings will contribute to the existing knowledge about fungal species in the Republic of Korea and future research on the fungal diversity on Muui Island.
