Project description:Controlling the six legs of an insect walking in an unpredictable environment is a challenging task, as many degrees of freedom have to be coordinated. Solutions proposed to deal with this task are usually based on the highly influential concept that (sensory-modulated) central pattern generators (CPG) are required to control the rhythmic movements of walking legs. Here, we investigate a different view. To this end, we introduce a sensor based controller operating on artificial neurons, being applied to a (simulated) insectoid robot required to exploit the "loop through the world" allowing for simplification of neural computation. We show that such a decentralized solution leads to adaptive behavior when facing uncertain environments which we demonstrate for a broad range of behaviors never dealt with in a single system by earlier approaches. This includes the ability to produce footfall patterns such as velocity dependent "tripod", "tetrapod", "pentapod" as well as various stable intermediate patterns as observed in stick insects and in Drosophila. These patterns are found to be stable against disturbances and when starting from various leg configurations. Our neuronal architecture easily allows for starting or interrupting a walk, all being difficult for CPG controlled solutions. Furthermore, negotiation of curves and walking on a treadmill with various treatments of individual legs is possible as well as backward walking and performing short steps. This approach can as well account for the neurophysiological results usually interpreted to support the idea that CPGs form the basis of walking, although our approach is not relying on explicit CPG-like structures. Application of CPGs may however be required for very fast walking. Our neuronal structure allows to pinpoint specific neurons known from various insect studies. Interestingly, specific common properties observed in both insects and crustaceans suggest a significance of our controller beyond the realm of insects.

Project description:Down syndrome (DS) is the most common genetic cause of intellectual disability and a major biomedical concern. Despite a general consensus that protein homeostasis is essential for normal brain function, little is known about its role in DS. Here, we show that the integrated stress response (ISR)—a conserved signaling network that maintains proteostasis by controlling protein synthesis—is activated in the brains of DS mice and individuals with DS, leading to reduced translation. Genetic and pharmacological suppression of the ISR, either by inhibiting the ISR-initiating double-stranded RNA-activated protein kinase (PKR) or boosting the function of the eIF2•eIF2B complex, which becomes limiting upon ISR activation, de-repressed translation, reversed enhanced inhibitory synaptic transmission, and rescued the deficits in synaptic plasticity and memory in DS mice. Our findings unveil a crucial role for the ISR in DS pathophysiology and suggest tuning of the ISR as a promising therapeutic intervention for DS.

Project description:Simple motor tics are sudden, nonrhythmic jerk like movements that are often preceded by premonitory urge and can be voluntarily suppressed. Some clinical characteristic of tics such as variability of movement, distractibility and fluctuating course can mimic functional movement disorders. Here we report a case of motor tics where the physiological approach in addition to careful clinical assessments is helpful to support the diagnosis of tic.

Project description:BackgroundIn Parkinson's Disease (PD), upper limb tremor during walking (TW) is observed and clinical observations suggest it may represent a variant of rest tremor. However, its neurophysiological characteristics remain unexplored.ObjectivesThis study compared the neurophysiological features of TW with other PD tremors and tested whether TW arises from reduced ipsilateral arm swing.MethodsInertial measurement units were used to measure frequency and amplitude of tremors and arm swing during walking in 25 PD patients.ResultsTW shared a similar frequency with rest and re-emergent tremor (RET) but showed significantly greater amplitude. A positive correlation was observed between the amplitude and frequency of TW with those of rest and RET on the same side. TW distribution was unrelated to reduced arm swing during walking, suggesting TW is not due to decreased ipsilateral arm movement.ConclusionsThese findings suggest that walking may act as a provocation maneuver, triggering rest tremor.

Project description:In many animals the ability to navigate over long distances is an important prerequisite for foraging. For example, it is widely accepted that desert ants and honey bees, but also mammals, use path integration for finding the way back to their home site. It is however a matter of a long standing debate whether animals in addition are able to acquire and use so called cognitive maps. Such a 'map', a global spatial representation of the foraging area, is generally assumed to allow the animal to find shortcuts between two sites although the direct connection has never been travelled before. Using the artificial neural network approach, here we develop an artificial memory system which is based on path integration and various landmark guidance mechanisms (a bank of individual and independent landmark-defined memory elements). Activation of the individual memory elements depends on a separate motivation network and an, in part, asymmetrical lateral inhibition network. The information concerning the absolute position of the agent is present, but resides in a separate memory that can only be used by the path integration subsystem to control the behaviour, but cannot be used for computational purposes with other memory elements of the system. Thus, in this simulation there is no neural basis of a cognitive map. Nevertheless, an agent controlled by this network is able to accomplish various navigational tasks known from ants and bees and often discussed as being dependent on a cognitive map. For example, map-like behaviour as observed in honey bees arises as an emergent property from a decentralized system. This behaviour thus can be explained without referring to the assumption that a cognitive map, a coherent representation of foraging space, must exist. We hypothesize that the proposed network essentially resides in the mushroom bodies of the insect brain.

Project description:Animal, and insect walking (locomotion) in particular, have attracted much attention from scientists over many years up to now. The investigations included behavioral, electrophysiological experiments, as well as modeling studies. Despite the large amount of material collected, there are left many unanswered questions as to how walking and related activities are generated, maintained, and controlled. It is obvious that for them to take place, precise coordination within muscle groups of one leg and between the legs is required: intra- and interleg coordination. The nature, the details, and the interactions of these coordination mechanisms are not entirely clear. To help uncover them, we made use of modeling techniques, and succeeded in developing a six-leg model of stick-insect walking. Our main goal was to prove that the same model can mimic a variety of walking-related behavioral modes, as well as the most common coordination patterns of walking just by changing the values of a few input or internal variables. As a result, the model can reproduce the basic coordination patterns of walking: tetrapod and tripod and the transition between them. It can also mimic stop and restart, change from forward-to-backward walking and back. Finally, it can exhibit so-called search movements of the front legs both while walking or standing still. The mechanisms of the model that enable it to produce the aforementioned behavioral modes can hint at and prove helpful in uncovering further details of the biological mechanisms underlying walking.

Project description:Multi-legged animals such as myriapods can locomote on unstructured rough terrain using their flexible bodies and legs. This highly adaptive locomotion emerges through the dynamic interactions between an animal's nervous system, its flexible body, and the environment. Previous studies have primarily focused on either adaptive leg control or the passive compliance of the body parts and have shown how each enhanced adaptability to complex terrains in multi-legged locomotion. However, the essential mechanism considering both the adaptive locomotor circuits and bodily flexibility remains unclear. In this study, we focused on centipedes and aimed to understand the well-balanced coupling between the two abovementioned mechanisms for rough terrain walking by building a neuromechanical model based on behavioral findings. In the behavioral experiment, we observed a centipede walking when part of the terrain was temporarily removed and thereafter restored. We found that the ground contact sense of each leg was essential for generating rhythmic leg motions and also for establishing adaptive footfall patterns between adjacent legs. Based on this finding, we proposed decentralized control mechanisms using ground contact sense and implemented them into a physical centipede model with flexible bodies and legs. In the simulations, our model self-organized the typical gait on flat terrain and adaptive walking during gap crossing, which were similar to centipedes. Furthermore, we demonstrated that the locomotor performance deteriorated on rough terrain when adaptive leg control was removed or when the body was rigid, which indicates that both the adaptive leg control and the flexible body are essential for adaptive locomotion. Thus, our model is expected to capture the possible essential mechanisms underlying adaptive centipede walking and pave the way for designing multi-legged robots with high adaptability to irregular terrain.

Project description:Quadrupeds exhibit versatile and adaptive running by exploiting the flying phase during the stride cycle. Various interlimb coordination mechanisms focusing on mechanical loads during the stance phase have been proposed to understand the underlying control mechanism, and various gait patterns have been reproduced. However, the essential control mechanism required to achieve both steady running patterns and non-steady behaviours, such as jumping and landing, remains unclear. Therefore, we focus on the vertical motions of the body parts and propose a new decentralized interlimb coordination mechanism. The simulation results demonstrate that the robot can generate efficient and various running patterns in response to the morphology of the body. Furthermore, the proposed model allows the robot to smoothly change its behaviour between steady running and non-steady landing depending on the situation. These results suggest that the steady and non-steady behaviours in quadruped adaptive running may share a common simple control mechanism based on the mechanical loads and vertical velocities of the body parts.

Project description:Pain is a negative affective state arising from tissue damage or inflammation. Because pain is aversive and its relief is innately rewarding, animals may learn to avoid a context in which pain is experienced and prefer one where pain relief occurs. It is generally accepted that vertebrate animals experience pain; however, there is currently inconclusive evidence that the affective component of pain occurs in any invertebrate. Here, we show that octopuses, the most neurologically complex invertebrates, exhibit cognitive and spontaneous behaviors indicative of affective pain experience. In conditioned place preference assays, octopuses avoided contexts in which pain was experienced, preferred a location in which they experienced relief from pain, and showed no conditioned preference in the absence of pain. Injection site grooming occurred in all animals receiving acetic acid injections, but this was abolished by local anesthesia. Thus, octopuses are likely to experience the affective component of pain.
