The complete mitochondrial genome of Oenopia sauzeti (Coleoptera: Coccinellidae).

ABSTRACT: Oenopia sauzeti Mulsant are predators of various aphids, such as Myzus persicae, Rhopalosiphum maidis, and Sitobion avenae. We sequenced the mitochondrial genome (mitogenome) of O. sauzeti. Consistent with other mitogenomes of Coleoptera, the circular mitogenome of O. sauzeti consists of 17,630 bp, including 13 PCG genes, 2 rRNA genes, and 22 tRNA genes. The mitogenome differs in 2 long non-coding regions of 1285 bp and 1843 bp. The 22 tRNA genes products are folded into the typical cloverleaf secondary structure, except trnS1, which lacks the dihydrouridine (DHU) arm. The base composition is AT-biased (80.1%). The phylogenetic tree confirms monophyly at the genus level within the Coccinellinae and supports O. sauzeti as a sister group to Aiolocaria hexaspilota.

Characterization of the complete mitochondrial genome and phylogenetic analysis of Epiverta chelonia (Coleoptera: Coccinellidae).

Project description:Epiverta chelonia (Mader 1933; Coleoptera: Coccinellidae) is an important economically and scientifically valuable insect. In this study, the first complete mitochondrial genome of E. chelonia was sequenced and characterized using next-generation sequencing techniques. The circular mitogenome of E. chelonia consists of 17,347 bp including 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and a control region (D-loop). The base composition was AT-biased (75.77%). Bayesian Inference and Maximum likelihood phylogenetic trees strongly supported the monophyly of Coccinellinae. Also, E. chelonia was supported as the sister group of Subcoccinella vigintiquatuorpunctata, within Epilachninae. Thus, the E. chelonia mitochondrial genome will be a fundamental resource for understanding the molecular phylogenetic relationships of the species-rich family Coccinellidae of Coleoptera.

Phylogenetic relationship and characterization of the complete mitochondrial genome of Cheilomenes sexmaculata (Coleoptera: Coccinellidae).

Project description:Cheilomenes sexmaculata is a common natural enemy for aphid and psyllid in agricultural systems in South China. In this study, we sequenced and analyzed the complete mitochondrial genome (mitogenome) of C. sexmaculata. This mitogenome was 17,297 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs) and two ribosomal RNA unit genes (rRNAs). Gene order was conserved and identical to most other previously sequenced Coccinellidae. All PCGs of C. sexmaculata have the conventional start codon for invertebrate mitochondrial PCGs (ATN), with the exception of cox1 (AAT) and nad3 (TTG). Except for seven genes (cox1, cox2, cox3, nad3, nad5, nad4 and nad6) end with the incomplete stop codon T-, all other PCGs terminated with the stop codon TAA or TAG. The whole mitogenome exhibited heavy AT nucleotide bias (78.0%). Phylogenetic analysis positioned C. sexmaculata in a well-supported clade with Aiolocaria hexaspilota. The relationships (Sticholotidinae + (Coccinellinae + (Scymninae + Epilachninae))) were supported in Coccinellidae, and Halyziini was paraphyletic to Coccinellini within Coccinellinae.

Characterization of the complete mitochondrial genome and phylogenetic analysis of Menochilus sexmaculata (Fabricius, 1781) (Coleoptera: Coccinellidae).

Project description:We sequenced the complete mitochondrial genome of Menochilus sexmaculata (Fabricius, 1781) and compared it with that of other insects. The mitogenome of M. sexmaculata is a circular molecule of 16,663 bp with 75.00% AT content, containing 13 protein-coding genes (PCGs), 22 tRNAs, 2 rRNAs, and one non-coding control region. All of the PCGs use the typical ATN as the initiation codon, with the exception of cox1 and nad3 which begin with AAT and TTG, respectively. Cox1, cox2, cox3, nad3, nad4, nad5 and nad6 employ a single T as a termination signal, while others have the typical termination codons (TAA or TAG). All the 22 typical animal tRNA genes are found in M. sexmaculata mitogenome, and most of the tRNAs could be folded into the classic cloverleaf secondary structure. Phylogenetic tree based on 13 PCGs suggested that M. sexmaculata is closely related to Anatis ocellata and Calvia championorum, and clustered within Coccinellidae.

Comparative and phylogenomic analyses of mitochondrial genomes in Coccinellidae (Coleoptera: Coccinelloidea).

Project description:The Coccinellidae are one of the most familiar beetle families, the ladybirds. Despite the great ecological and economic significance, the phylogenetic relationships of Coccinellidae remain poorly understood. One of the reasons is that the sequenced mitogenomes available for this family are very limited. We sequenced complete or nearly complete mitogenomes from seven species of the tribe Coccinellini with next-generation sequencing. All species have the same gene content and gene order as the putatively ancestral insect mitogenome. A large intergenic spacer region (> 890 bp) was found located between trnI and trnQ. The potential for using secondary structures of the large and small ribosomal subunits for phylogenetic reconstruction was predicted. The phylogenetic relationships were explored through comparative analyses across more than 30 coccinellid species. We performed phylogenetic analyses with both concatenation methods (Maximum Likelihood and Bayesian Inference) and multispecies coalescent method (ASTRAL). Phylogenetic results strongly supported the monophyly of Coccinellidae. Within Coccinellidae, the Epilachnini and the Coccinellini including Halyziini were monophyletic, while the Scymnini and Coccidulini were non-monophyletic.

Next-generation sequencing yields mitochondrial genome of Coccidophilus cariba Gordon (Coleoptera: Coccinellidae) from museum specimen.

Project description:We assembled and annotated the first complete mitochondrial genome of a species from the subfamily Microweiseinae, Coccidophilus cariba Gordon, a predator of scale insect pest. The circular mitogenome consists of 15,343 bp in length, including 13 PCGs, 22 tRNA, and 2 rRNA genes, and exhibits the typical ladybird mitogenome structure. A phylogenetic analysis with the published mitogenomes of other 12 ladybirds is presented, which confirms the position of Microweiseinae as a sister group of Coccinellinae. The C. cariba mitogenome could be a useful source to future studies in the relationships inside the subfamily and the genus.

Project description:The complete mitochondrial DNA genome of the Oberea diversipes was reported. The genome was a circular molecule of 15,499 base pairs (bp), with 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and an estimated 830 bp A + T-rich control region. The total base composition of the mitogenome was A-T bias, with 40.9% for A, 37.0% for T, 13.4% for C, and 8.7% for G. The molecular data presented here would be useful for further study of O. diversipes.

Complete mitochondrial genome of Cyllorhynchites ursulus (Coleoptera: Rhynchitidae).

Project description:The complete mitochondrial genome of Cylloryhunchites ursulus was sequenced for the first time from its whole body using the next-generation sequencing method. This mitogenome was 14,508 base pairs in length, containing 13 protein-coding genes, 20 transfer RNA genes, and two ribosomal RNA genes. Its overall A, C, G, and T contents were 37.8, 17.8, 11.1, and 33.4%, respectively. Its A + T content (71.2%) was higher than G + C content. The phylogenetic result showed that although C. ursulus is a species of family Rhynchitidae, its phylogenetic position is closely related to family Attlabidae.

Complete mitochondrial genome of Niphades castanea (Coleoptera: Curculionidae).

Project description:Niphades castanea Chao is an important insect pest on many plants which belong to genus Castanea. The complete mitochondrial genome of N. castanea was sequenced and analyzed. The phylogenetic relationships between N. castanea and other 41 species in the family Curculionidae were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that N. castanea is closely related to Hylobitelus xiaoi, which is in accordance with the traditional morphological classification.

First Complete Mitochondrial Genome of Melyridae (Coleoptera, Cleroidea): Genome Description and Phylogenetic Implications.

Project description:To explore the characteristics of the mitogenome of Melyridae and reveal phylogenetic relationships, the mitogenome of Cordylepherus sp. was sequenced and annotated. This is the first time a complete mitochondrial genome has beengenerated in this family. Consistent with previous observations of Cleroidea species, the mitogenome of Cordylepherus sp. is highly conserved in gene size, organization and codon usage, and secondary structures oftRNAs. All protein-coding genes(PCGs) initiate withthe standard start codon ATN,except ND1, which starts with TTG, and terminate with the complete stop codons of TAA and TAG, or incomplete forms, TA- and T-. MosttRNAs have the typical clover-leaf structure, except trnS1 (Ser, AGN), whose dihydrouridine (DHU) armis reduced. In the A+T-rich region, three types of tandem repeat sequence units are found, including a 115 bp sequence tandemly repeated twice, a 16 bp sequence tandemly repeated three timeswith a partial third repeat and a 10 bp sequence tandemly repeated seven times. Phylogenetic analyses based on 13 protein-coding genes by both Bayesian inference (BI) and maximum likelihood (ML) methods suggest that Melyridae sensu lato is polyphyletic, and Dasytinae and Malchiinae are supported as independent families.
