Development of the first axillary in vitro shoot multiplication protocol for coconut palms.

ABSTRACT: The coconut palm or "tree of life" is one of nature's most useful plants and the demand for its fruit is increasing. However, coconut production is threatened by ageing plantations, pests and diseases. Currently, the palm is exclusively propagated via seeds, limiting the amount of planting material. A novel micropropagation method is presented, based on axillary shoot formation. Apical meristems of in vitro coconut seedlings are cultured onto Y3 medium containing 1 µM TDZ. This induces the apical meristem to proliferate through axillary shoots in ~ 27% of the initiated explants. These axillary shoots are seen as white clumps of proliferating tissue and can be multiplied at a large scale or regenerated into rooted in vitro plantlets. This innovative micropropagation method will enable the production of disease-free, high quality in vitro plantlets, which will solve the worldwide scarcity of coconut planting material.

The origin, initiation and development of axillary shoot meristems in Lotus japonicus.

Project description:Background and aimsLotus japonicus 'Gifu' develops multiple axillary shoots in the cotyledonary node region throughout the growth of the plant. The origin, initiation and development of these axillary meristems were investigated.MethodsMorphological, histological and mRNA in situ analyses were done to characterize the ontogeny of cotyledonary axillary shoot meristems in Lotus. Morphological characterization of a putative Lotus shoot branching mutant (super-accessory branches) sac, is presented.Key resultsBy using expression of an L. japonicus STM-like gene as a marker for meristematic tissues, it was demonstrated that groups of cells maintained in the meristematic state at the cotyledonary axil region coincide with the sites where additional axillary meristems (accessory meristems) form. A Lotus shoot branching mutant, sac, is a putative Lotus branching mutant characterized by increased proliferation of accessory shoots in all leaf axils including the cotyledons.ConclusionIn Lotus, axillary shoot meristems continually develop at the cotyledonary node region throughout the growth of the plant. These cotyledonary primary and accessory axillaries arise from the position of a meristematic zone of tissue at the cotyledonary node axil region.

Profiles of Cytokinins Metabolic Genes and Endogenous Cytokinins Dynamics during Shoot Multiplication In Vitro of Phalaenopsis.

Project description:Shoot multiplication induced by exogenous cytokinins (CKs) has been commonly used in Phalaenopsis micropropagation for commercial production. Despite this, mechanisms of CKs action on shoot multiplication remain unclear in Phalaenopsis. In this study, we first identified key CKs metabolic genes, including six isopentenyltransferase (PaIPTs), six cytokinin riboside 5' monophosphate phosphoribohydrolase (PaLOGs), and six cytokinin dehydrogenase (PaCKXs), from the Phalaenopsis genome. Then, we investigated expression profiles of these CKs metabolic genes and endogenous CKs dynamics in shoot proliferation by thidiazuron (TDZ) treatments (an artificial plant growth regulator with strong cytokinin-like activity). Our data showed that these CKs metabolic genes have organ-specific expression patterns. The shoot proliferation in vitro was effectively promoted with increased TDZ concentrations. Following TDZ treatments, the highly expressed CKs metabolic genes in micropropagated shoots were PaIPT1, PaLOG2, and PaCKX4. By 30 days of culture, TDZ treatments significantly induced CK-ribosides levels in micropropagated shoots, such as tZR and iPR (2000-fold and 200-fold, respectively) as compared to the controls, whereas cZR showed only a 10-fold increase. Overexpression of PaIPT1 and PaLOG2 by agroinfiltration assays resulted in increased CK-ribosides levels in tobacco leaves, while overexpression of PaCKX4 resulted in decreased CK-ribosides levels. These findings suggest de novo biosynthesis of CKs induced by TDZ, primarily in elevation of tZR and iPR levels. Our results provide a better understanding of CKs metabolism in Phalaenopsis micropropagation.

Interaction between Growth Regulators Controls In Vitro Shoot Multiplication in Paulownia and Selection of NaCl-Tolerant Variants.

Project description:The interaction between cytokinin, auxin and GA controlled in vitro shoot multiplication in paulownia was influenced by a medium water potential (Ψ) modulation, where it was modulated using different textures or strengths of MS medium, media of different types (MS, WPM, SH and B5) or NaCl incorporation. The interaction between 2 mg/L BAP and 0.1 mg/L NAA expressed the highest shoot number on each media type, but it was better with media of lower water potential (MS and WPM), and MS medium was the best. Ψ of full-strength semisolid MS medium expressed the highest shoot multiplication. The opposite was detected when Ψ of MS medium was changed using half- or double-strength MS. Ψ of full-strength MS medium in semisolid form resulted in a valuable interaction between 2 mg/L BAP, 0.1 mg/L NAA and 0.1 mg/L GA, leading to efficient shoot formation, and it was associated with an increase in internode length and decrease in stem diameter, which facilitated obtaining synseeds with a high ability to convert. High genetic variation was recorded under long-term culture (14 subcultures). Polymorphism using the ISSR technique was higher than that of RAPD. A further increase in polymorphism was detected when NaCl was used, where five salt-tolerant lines were selected. Some salt-tolerant-selected lines showed one or more amplification products of a specific molecular weight that did not appear in the control. For example, with OPA-07 and OPG-02 RAPD primers, all the salt-tolerant-selected lines showed the appearance of amplification fragments (610 bp and 300 bp, respectively) that were not detected in control.

Successful In Vitro Shoot Multiplication of <i>Quercus robur</i> L. Trees Aged up to 800 Years.

Project description:The conservation of the genetic resources of old trees is crucial to their ecological role but is extremely difficult, especially for oak species (Quercus spp.) displaying recalcitrance in seed and vegetative propagation methods. Our study aimed to assess the regenerative potential of Quercus robur trees of different ages (up to 800 years) during micropropagation. We also aimed to determine how in vitro conditions can influence in vitro regeneration responses. Lignified branches collected from 67 selected trees were cultivated ex vitro in culture pots at 25 °C to obtain epicormic shoots (explant sources). The explants were cultivated on an agar medium supplemented with 0.8 mg L-1 6-benzylaminopurine (BAP) for at least 21 months. In a second experiment, two different shoot multiplication conditions (temporary immersion-RITA® bioreactor and agar medium) and two culture medium formulations (Woody Plant Medium and modified Quoirin and Lepoivre medium) were tested. The results showed that the mean length of the epicormic shoots obtained in a pot culture was a function of donor age and was similar among the group of younger trees (ca. 20-200 years), and varied between older trees (ca. 300-800 years). The efficiency of in vitro shoot multiplication strictly depended on the genotype. A sustainable in vitro culture (defined as survival after 6 months) was only possible for half of the tested old donor trees, even when they survived the first month of in vitro growth. A continuous monthly increase in the number of in vitro cultured shoots was reported in younger oaks and in some old oaks. We found a significant effect of the culture system and the macro- and micronutrient composition on in vitro shoot growth. This is the first report demonstrating that the in vitro culture can be successfully applied to the propagation of even 800-year-old pedunculate oak trees.

LED light improves shoot multiplication, steviol glycosides and phenolic compounds biosynthesis in Stevia rebaudiana Bertoni in vitro culture.

Project description:Light-emitting diode (LED) lamps are efficient elicitors of secondary metabolites. To investigate the influence of LED light on steviol glycosides (SGs) and phenolic compounds biosynthesis, stevia shoots were cultured under the following LED lights: white-WL, blue-B, red-R, 70% red and 30% blue-RB, 50% UV, 35% red and 15% blue-RBUV, 50% green, 35% red and 15% blue-RBG, 50% yellow, 35% red and 15% blue-RBY, 50% far-red, 35% red and 15% blue-RBFR and white fluorescent light (WFl, control). RBG light stimulated shoots' biomass production. RBFR had a beneficial impact on stevioside biosynthesis (1.62 mg/g dry weight, DW), while RBUV favoured the production of rebaudioside A (3.15 mg/g DW). Neochlorogenic, chlorogenic, caffeic, 4-feruloylquinic, isochlorogenic A, rosmarinic acids and the flavonoid quercitrin were identified in the obtained material. A stimulatory effect of RBFR and RBUV on the biosynthesis of phenolic compounds was noted. LED light also influenced stomata appearance, stomata density, photosynthetic pigments, soluble sugar content and antioxidant enzyme activities in stevia shoots. This is the first report to provide evidence of the stimulating effect of LED light on biomass yield, SGs production and phenolic compounds in stevia shoot cultures.

Then, main stems were decapitated, and axillary shoots were collected at 24 hours after decapitation (named as growing shoot). Total RNA was prepared from either control or growing shoots and used for the microarray analysis. Coconut Lethal Yellowing Diseases: A Phytoplasma Threat to Palms of Global Economic and Social Significance.

Project description:The recent discovery of Bogia coconut syndrome in Papua New Guinea (PNG) is the first report of a lethal yellowing disease (LYD) in Oceania. Numerous outbreaks of LYDs of coconut have been recorded in the Caribbean and Africa since the late Nineteenth century and have caused the death of millions of palms across several continents during the Twentieth century. Despite the severity of economic losses, it was only in the 1970s that the causes of LYDs were identified as phytoplasmas, a group of insect-transmitted bacteria associated with diseases in many other economically important crop species. Since the development of polymerase chain reaction (PCR) technology, knowledge of LYDs epidemiology, ecology and vectors has grown rapidly. There is no economically viable treatment for LYDs and vector-based management is hampered by the fact that vectors have been positively identified in very few cases despite many attempted transmission trials. Some varieties and hybrids of coconut palm are known to be less susceptible to LYD but none are completely resistant. Optimal and current management of LYD is through strict quarantine, prompt detection and destruction of symptomatic palms, and replanting with less susceptible varieties or crop species. Advances in technology such as loop mediated isothermal amplification (LAMP) for detection and tracking of phytoplasma DNA in plants and insects, remote sensing for identifying symptomatic palms, and the advent of clustered regularly interspaced short palindromic repeats (CRISPR)-based tools for gene editing and plant breeding are likely to allow rapid progress in taxonomy as well as understanding and managing LYD phytoplasma pathosystems.

In vitro shoot multiplication of Rumex vesicarius L., and quantification of ascorbic acid and major phenolics from its leaf derived callus.

Project description:An optimised method for enhanced in vitro shoot multiplication of Rumex vesicarius (Polygonaceae)-a branched succulent herb-was achieved. The in vitro seed pre-treatment with 4% urea was able to show 95% seed germination on MS medium within 2 weeks of culturing. In vitro shoot bud induction from shoot tip explants was best in the presence of 2 mg L-1 kinetin on Murashige and Skoog medium (MS medium), wherein 8.3 shoots/explants were observed. Shoot elongation was found to be high (3.75?±?0.5 cm) in 2 mg L-1 BA comprising medium. The transfer of micro-shoots on to MS medium comprising 1.5 mg L-1 IBA and 1% activated charcoal (w/v) supported efficiently in vitro rooting (2.5-4.0 cm) in 3-4 weeks. Upon hardening, 70% rooted plants survived under greenhouse conditions. Though friable callus was produced on MS medium-containing 2 mg L-1 BA followed by 3 mg L-1 BA, no organogenesis was noticed. The ascorbic acid content of 78.62?±?0.25 mg 100 g-1 FW was recorded in callus cultures grown on medium supplemented with 2 mg L-1 BA, and it is 1.74-fold more compared to normal ex vitro leaves of the same age. In vitro raised plant leaf showed 1.98-fold more ascorbic acid (89.42?±?0.18 mg 100 g-1 FW) to that of ex vitro leaves. The total phenolic content was found to be 60 mg in callus as compared to 610 mg (per 100 g GAE FW) of ex vitro leaves. The major phenolic compounds quantified were synergic, chlorogenic, ferulic, and generic acids, respectively. This optimised protocol will facilitate to pursue scale-up studies for in vitro ascorbic acid production and also to further investigate the kinetics of biosynthetic pathway genes involved.
