The digestive and reproductive tract microbiotas and their association with body weight in laying hens.

ABSTRACT: Body weight at the onset of egg production is a major factor influencing hen productivity, as suitable body weight is crucial to laying performance in laying hens. To better understand the association between body weight and microbial community membership and structure in different sites of the digestive and reproductive tracts in chickens, we performed 16S rRNA sequencing surveys and focused on how the microbiota may interact to influence body weight. Our results demonstrated that the microbial community and structure of the digestive and reproductive tracts differed between low and high body weight groups. In particular, we found that the species Pseudomonas viridiflava was negatively associated with body weight in the 3 digestive tract sites, while Bacteroides salanitronis was negatively associated with body weight in the 3 reproductive tract sites; and further in-depth studies are needed to explore their function. These findings will help extend our understanding of the influence of the bird digestive and reproductive tract microbiotas on body weight trait and provide future directions regarding the control of body weight in the production of laying hens.

Project description:Filarial worms are parasitic nematodes that cause devastating diseases such as lymphatic filariasis (LF) and onchocerciasis. Filariae are nematodes with complex anatomy including fully developed digestive tracts and reproductive organs. To better understand the basic biology of filarial parasites and to provide insights into drug targets and vaccine design, we conducted a proteomic analysis of different anatomic fractions of Brugia malayi, a causative agent of LF. Approximately 500 adult female B. malayi worms were dissected, and three anatomical fractions (body wall, digestive tract, and reproductive tract) were obtained. Proteins from each anatomical fraction were extracted, desalted, trypsinized, and analyzed by microcapillary reverse-phase liquid chromatography-tandem-mass spectrometry. In total, we identified 4,785 B. malayi proteins. While 1,894 were identified in all three anatomic fractions, 396 were positively identified only within the digestive tract, 114 only within the body wall, and 1,011 only within the reproductive tract. Gene set enrichment analysis revealed a bias for transporters to be present within the digestive tract, suggesting that the intestine of adult filariae is functional and important for nutrient uptake or waste removal. As expected, the body wall exhibited increased frequencies of cytoskeletal proteins, and the reproductive tract had increased frequencies of proteins involved in nuclear regulation and transcription. In assessing for possible vaccine candidates, we focused on proteins sequestered within the digestive tract, as these could possibly represent "hidden antigens" with low risk of prior allergic sensitization. We identified 106 proteins that are enriched in the digestive tract and are predicted to localize to the surface of cells in the the digestive tract. It is possible that some of these proteins are on the luminal surface and may be accessible by antibodies ingested by the worm. A subset of 27 of these proteins appear especially promising vaccine candidates as they contain significant non-cytoplasmic domains, only 1-2 transmembrane domains, and a high degree of homology to W. bancrofti and/or O. volvulus.

Project description:This study investigated the effects of multi-enzyme supplementation on various aspects of laying hens, including laying performance, egg quality, intestinal health and digestive function. In total, 384 Jingfen No.6 laying hens at 65-week-age were randomly assigned to four distinct dietary treatments: a basal diet (CON), CON supplemented with 150 g/t multi-enzyme (T1), CON with 300 g/t multi-enzyme (T2), and 600 g/t multi-enzyme (T3). A significant linear (P = 0.044) and quadratic (P = 0.014) increase was observed in the laying rate, while the feed/egg ratio exhibited a linear (P = 0.001) and quadratic (P < 0.001) decrease with increasing multi-enzyme supplementation. Additionally, linear (P < 0.05) and quadratic (P < 0.05) increases were observed in yolk rate and haugh unit with increasing levels of multi-enzyme supplementation. The trypsin activity in the duodenum and crude protein digestibility showed linear (P < 0.05) and quadratic (P < 0.05) increase with the addition of multi-enzyme. Furthermore, lipase and amylase activities in the duodenum increased quadratically (P = 0.041) and linearly (P = 0.040), respectively. Both jejunal and ileal digesta viscosities showed linear (P < 0.05) and quadratic (P < 0.05) decrease with the increasing addition of multi-enzyme. Moreover, multi-enzyme supplementation significantly increased (P < 0.05) the number of goblet cells in the intestinal of the treatment groups. The mRNA expression of Occludin-1, mucin 2 (MUC-2), large neutral amino acids transporter small subunit 1 (LAT-1) in the jejunum were significantly increased (P < 0.05) in the treatment groups (T1, T2 and T3) compared to the CON group. Additionally, the mRNA expression of solute carrier family 6-member 19 (B0AT-1) and large neutral amino acids transporter small subunit 4 (LAT-4) were significantly evaluated (P < 0.05) in the T2 and T3 groups, respectively. In conclusion, multi-enzyme supplementation enhanced digestive enzyme activities and intestinal barrier function, reduced intestinal digesta viscosity, and regulated mRNA expression of intestinal amino acid and lipid transporter genes, thereby improving crude protein digestibility and positively affecting laying performance and egg quality in hens.

Project description:This study was aimed to isolate sinapine-degrading bacteria from the intestinal tract of laying hens and to identify the predominant bacteria. Thirty-week old healthy laying hens were killed, and the chyme in the digestive tract was inoculated into modified Czapek medium containing sinapin and cultivated at 37 °C for 10 days. The optical density (OD) values of the bacterial solutions at different cultivating times were detected by a spectrophotometric method. The predominant strains were identified by 16S rRNA gene analysis. We extracted the extracellular products of the predominant strains to determine the total protein using the Coomassie brilliant blue method, and to determine the activities of some extracellular enzymes using the agar plate diffusion method. Nine strains were isolated from the lower intestinal tract of laying hens. Among the 9 strains, 5 were from the ileum, 2 were from the ceca and 2 were from the jejunum. We could not isolate any strains from the upper intestinal tract, such as the stomach and duodenum. Eight of those 9 isolated strains were gram negative and one was gram positive. Strains YD-1 and YD-2 were better than other strains in their abilities to degrade sinapine. Strains YD-1 and YD-2 were identified as Escherichia coli and Klebsiella spp., respectively, by the 16S rRNA sequence analysis. The total protein level of the extracellular products was 1.213 g/L for YD-1 and 1.990 g/L for YD-2. Both extracellular products of YD-1 and YD-2 had the activities of protease, amylase and urease. This study confirmed that the primary site of sinapine degradation is in the lower intestinal tract of laying hens. The sinapine-degrading strains are mainly gram negative. Strains YD-1 and YD-2 are predominant in degrading sinapine and they belong to E. coli and Klebsiella spp., respectively. Both extracellular products of YD-1 and YD-2 contain protease, amylase and urease. Strain YD-2 is better than strain YD-1 in its ability to degrade sinapine.
