A quadratic model captures the human V1 response to variations in chromatic direction and contrast.

ABSTRACT: An important goal for vision science is to develop quantitative models of the representation of visual signals at post-receptoral sites. To this end, we develop the quadratic color model (QCM) and examine its ability to account for the BOLD fMRI response in human V1 to spatially uniform, temporal chromatic modulations that systematically vary in chromatic direction and contrast. We find that the QCM explains the same, cross-validated variance as a conventional general linear model, with far fewer free parameters. The QCM generalizes to allow prediction of V1 responses to a large range of modulations. We replicate the results for each subject and find good agreement across both replications and subjects. We find that within the LM cone contrast plane, V1 is most sensitive to L-M contrast modulations and least sensitive to L+M contrast modulations. Within V1, we observe little to no change in chromatic sensitivity as a function of eccentricity.

Coding of chromatic spatial contrast by macaque V1 neurons.

Project description:Color perception relies on comparisons between adjacent lights, but how the brain performs these comparisons is poorly understood. To elucidate the underlying mechanisms, we recorded spiking responses of individual V1 neurons in macaque monkeys to pairs of stimuli within the classical receptive field (RF). We estimated the spatial-chromatic RF of each neuron and then presented customized colored edges using a closed-loop technique. We found that many double-opponent (DO) cells, which have spatially and chromatically opponent RFs, responded to chromatic contrast as a weighted sum, akin to how other V1 neurons responded to luminance contrast. Yet other neurons integrated chromatic signals nonlinearly, confirming that linear signal integration is not an obligate property of V1 neurons. The functional similarity of cone-opponent DO cells and cone non-opponent simple cells suggests that these two groups may share a common underlying circuitry, promotes the construction of image-computable models for full-color image representation, and sheds new light on V1 complex cells.

Contrast response in a comprehensive network model of macaque V1.

Project description:The response to contrast is one of the most important functions of the macaque primary visual cortex, V1, but up to now there has not been an adequate theory for it. To fill this gap in our understanding of cortical function, we built and analyzed a new large-scale, biologically constrained model of the input layer, 4Cα, of macaque V1. We called the new model CSY2. We challenged CSY2 with a three-parameter family of visual stimuli that varied in contrast, orientation, and spatial frequency. CSY2 accurately simulated experimental data and made many new predictions. It accounted for 1) the shapes of firing-rate-versus-contrast functions, 2) orientation and spatial frequency tuning versus contrast, and 3) the approximate contrast-invariance of cortical activity maps. Post-analysis revealed that the mechanisms that were needed to produce the successful simulations of contrast response included strong recurrent excitation and inhibition that find dynamic equilibria across the cortical surface, dynamic feedback between L6 and L4, and synaptic dynamics like inhibitory synaptic depression.

Primate area V1: largest response gain for receptive fields in the straight-ahead direction.

Project description:Although neuronal responses in behaving monkeys are typically studied while the monkey fixates straight ahead, it is known that eye position modulates responses of visual neurons. The modulation has been found to enhance neuronal responses when the receptive field is placed in the straight-ahead position for neurons receiving input from the peripheral but not the central retina. We studied the effect of eye position on the responses of V1 complex cells receiving input from the central retina (1.1-5.7° eccentricity) while minimizing the effect of fixational eye movements. Contrast response functions were obtained separately with drifting light and dark bars. Data were fit with the Naka-Rushton equation: r(c)=Rmax×c/(c+c50)+s, where r(c) is mean spike rate at contrast c, Rmax is the maximum response, c50 is the contrast that elicits half of Rmax, and s is the spontaneous activity. Contrast sensitivity as measured by c50 was not affected by eye position. For dark bars, there was a statistically significant decline in the normalized Rmax with increasing deviation from straight ahead. Data for bright bars showed a similar trend with a less rapid decline. Our results indicate that neurons representing the central retina show a bias for the straight-ahead position resulting from modulation of the response gain without an accompanying modulation of contrast sensitivity. The modulation is especially obvious for dark stimuli, which might be useful for directing attention to hazardous situations such as dark holes or shadows concealing important objects (Supplement 1: Video Abstract, Supplemental digital content 1, http://links.lww.com/WNR/A295).

Standing Variations Modeling Captures Inter-Individual Heterogeneity in a Deterministic Model of Prostate Cancer Response to Combination Therapy.

Project description:Sipuleucel-T (Provenge) is the first live cell vaccine approved for advanced, hormonally refractive prostate cancer. However, survival benefit is modest and the optimal combination or schedule of sipuleucel-T with androgen depletion remains unknown. We employ a nonlinear dynamical systems approach to modeling the response of hormonally refractive prostate cancer to sipuleucel-T. Our mechanistic model incorporates the immune response to the cancer elicited by vaccination, and the effect of androgen depletion therapy. Because only a fraction of patients benefit from sipuleucel-T treatment, inter-individual heterogeneity is clearly crucial. Therefore, we introduce our novel approach, Standing Variations Modeling, which exploits inestimability of model parameters to capture heterogeneity in a deterministic model. We use data from mouse xenograft experiments to infer distributions on parameters critical to tumor growth and to the resultant immune response. Sampling model parameters from these distributions allows us to represent heterogeneity, both at the level of the tumor cells and the individual (mouse) being treated. Our model simulations explain the limited success of sipuleucel-T observed in practice, and predict an optimal combination regime that maximizes predicted efficacy. This approach will generalize to a range of emerging cancer immunotherapies.

Chromatic detection from cone photoreceptors to V1 neurons to behavior in rhesus monkeys.

Project description:Chromatic sensitivity cannot exceed limits set by noise in the cone photoreceptors. To determine how close neurophysiological and psychophysical chromatic sensitivity come to these limits, we developed a parameter-free model of stimulus encoding in the cone outer segments, and we compared the sensitivity of the model to the psychophysical sensitivity of monkeys performing a detection task and to the sensitivity of individual V1 neurons. Modeled cones had a temporal impulse response and a noise power spectrum that were derived from in vitro recordings of macaque cones, and V1 recordings were made during performance of the detection task. The sensitivity of the simulated cone mosaic, the V1 neurons, and the monkeys were tightly yoked for low-spatiotemporal-frequency isoluminant modulations, indicating high-fidelity signal transmission for this class of stimuli. Under the conditions of our experiments and the assumptions for our model, the signal-to-noise ratio for these stimuli dropped by a factor of ?3 between the cones and perception. Populations of weakly correlated V1 neurons narrowly exceeded the monkeys' chromatic sensitivity but fell well short of the cones' chromatic sensitivity, suggesting that most of the behavior-limiting noise lies between the cone outer segments and the output of V1. The sensitivity gap between the cones and behavior for achromatic stimuli was larger than for chromatic stimuli, indicating greater postreceptoral noise. The cone mosaic model provides a means to compare visual sensitivity across disparate stimuli and to identify sources of noise that limit visual sensitivity.

Contrast-dependence of surround suppression in Macaque V1: experimental testing of a recurrent network model.

Project description:Neuronal responses in primary visual cortex (V1) to optimally oriented high-contrast stimuli in the receptive field (RF) center are suppressed by stimuli in the RF surround, but can be facilitated when the RF center is stimulated at low contrast. The neural circuits and mechanisms for surround modulation are still unknown. We previously proposed that topdown feedback connections mediate suppression from the "far" surround, while "near' surround suppression is mediated primarily by horizontal connections. We implemented this idea in a recurrent network model of V1. A model assumption needed to account for the contrast-dependent sign of surround modulation is a response asymmetry between excitation and inhibition; accordingly, inhibition, but not excitation, is silent for weak visual inputs to the RF center, and surround stimulation can evoke facilitation. A prediction stemming from this same assumption is that surround suppression is weaker for low than for high contrast stimuli in the RF center. Previous studies are inconsistent with this prediction. Using single unit recordings in macaque V1, we confirm this model's prediction. Model simulations demonstrate that our results can be reconciled with those from previous studies. We also performed a systematic comparison of the experimentally measured surround suppression strength with predictions of the model operated in different parameter regimes. We find that the original model, with strong horizontal and no feedback excitation of local inhibitory neurons, can only partially account quantitatively for the experimentally measured suppression. Strong direct feedback excitation of V1 inhibitory neurons is necessary to account for the experimentally measured surround suppression strength.

Spatio-chromatic contrast sensitivity under mesopic and photopic light levels.

Project description:Contrast sensitivity functions (CSFs) characterize the sensitivity of the human visual system at different spatial scales, but little is known as to how contrast sensitivity for achromatic and chromatic stimuli changes from a mesopic to a highly photopic range reflecting outdoor illumination levels. The purpose of our study was to further characterize the CSF by measuring both achromatic and chromatic sensitivities for background luminance levels from 0.02 cd/m2 to 7,000 cd/m2. Stimuli consisted of Gabor patches of different spatial frequencies and angular sizes, varying from 0.125 to 6 cpd, which were displayed on a custom high dynamic range (HDR) display with luminance levels up to 15,000 cd/m2. Contrast sensitivity was measured in three directions in color space, an achromatic direction, an isoluminant "red-green" direction, and an S-cone isolating "yellow-violet" direction, selected to isolate the luminance, L/M-cone opponent, and S-cone opponent pathways, respectively, of the early postreceptoral processing stages. Within each session, observers were fully adapted to the fixed background luminance (0.02, 2, 20, 200, 2,000, or 7,000 cd/m2). Our main finding is that the background luminance has a differential effect on achromatic contrast sensitivity compared to chromatic contrast sensitivity. The achromatic contrast sensitivity increases with higher background luminance up to 200 cd/m2 and then shows a sharp decline when background luminance is increased further. In contrast, the chromatic sensitivity curves do not show a significant sensitivity drop at higher luminance levels. We present a computational luminance-dependent model that predicts the CSF for achromatic and chromatic stimuli of arbitrary size.

The butterfly Papilio xuthus detects visual motion using chromatic contrast.

Project description:Many insects' motion vision is achromatic and thus dependent on brightness rather than on colour contrast. We investigate whether this is true of the butterfly Papilio xuthus, an animal noted for its complex retinal organization, by measuring head movements of restrained animals in response to moving two-colour patterns. Responses were never eliminated across a range of relative colour intensities, indicating that motion can be detected through chromatic contrast in the absence of luminance contrast. Furthermore, we identify an interaction between colour and contrast polarity in sensitivity to achromatic patterns, suggesting that ON and OFF contrasts are processed by two channels with different spectral sensitivities. We propose a model of the motion detection process in the retina/lamina based on these observations.

Stimulus-contrast-induced biases in activation order reveal interaction between V1/V2 and human MT+.

Project description:The luminance contrast of a visual stimulus is known to modulate the response properties of areas V1 and the human MT complex (hMT+), but has not been shown to modulate interactions between these two areas. We examined the direction of information transfer between V1/V2 and hMT+ at different stimulus contrasts by measuring magnetoencephalographic (MEG) responses to moving and stationary stimuli presented centrally or peripherally. To determine the direction of information flow, the different response latencies among stimuli and hemispheres in V1/V2 was compared with those of hMT+. At high contrast, responses to stimulus motion and position began in V1/V2, and were followed in hMT+ with a delay between 34 and 55 ms. However, at low contrast, lateralized responses in hMT+ came first, with those in V1/V2 lagging with a delay of 27 ms. Also, at high contrast, stationary stimuli produced greater responses than motion stimuli in V1/V2, while the reverse was true in hMT+, whose response lagged behind the initial response in V1/V2. The same activation order was found using Mutual Information Analysis of the response variances for each condition. Here, the response variances in hMT+ mimicked and trailed those of V1/V2 at high contrast, whereas the reverse was true at low contrast. Such consistent interactions found using two different methodologies strongly supports a processing link between these two areas. The results also suggest that feedback from hMT+ for low-contrast stimuli compensates for unresolved processing in V1/V2 when the input of a visual image is weak.

Naive stem cell blastocyst model captures human embryo lineage segregation.

Project description:Human naive pluripotent cells can differentiate into extraembryonic trophectoderm and hypoblast. Here we describe a human embryo model (blastoid) generated by self-organization. Brief induction of trophectoderm leads to formation of blastocyst-like structures within 3 days. Blastoids are composed of three tissue layers displaying exclusive lineage markers, mimicking the natural blastocyst. Single-cell transcriptome analyses confirm segregation of trophectoderm, hypoblast, and epiblast with high fidelity to the human embryo. This versatile and scalable system provides a robust experimental model for human embryo research.
