ABSTRACT: Light mainly affects animal reproductive performance through the hypothalamus- pituitary-gonadal axis, but the specific regulating mechanism is not yet clear in duck. To reveal the effects of light on the laying performance of ducks and its possible regulatory mechanisms, Shanma ducks at 52 weeks age were divided into three group treated with different photoperiods of 16 hr (control group), 24 hr (long-photoperiod group, LP), and 8 hr (short-photoperiod group, SP). Laying performance, endocrine-related hormones and gene expression of three groups were compared. The results showed that laying performance was greatest in the LP group; including laying rate, average egg weight and feed-egg ratio. Compared to the SP group, GnIH plasma concentration was decreased in the LP group, whilst FSH was increased in the LP group. GnIHR gene expression in the pituitary and large yellow follicles were downregulated in the LP group. The expression of Mel-a in large white follicles, and Mel-b and Mel-c in the hypothalamus were also downregulated in the LP group. Altogether these results suggest that extended photoperiods may promote the laying performance of ducks by inhibiting the secretion of GnIH and the expression of GnIHR and melatonin receptor genes.

Project description:We investigated the effect of photoperiod on performance, ovarian morphology, reproductive hormones levels, and their receptors mRNA expressions in laying ducks. After adaption, 300 252-day-old Jinding laying ducks were randomly allocated to 5 groups, receiving 12L:12D, 14L:10D, 16L:8D, 18L:6D, or 20L:4D, respectively. Each treatment had 6 replicates of 10 birds each. The feeding trial lasted 8 wk. Egg production, egg mass, and ADFI increased linearly and quadratically with increasing photoperiods (P < 0.05), and the higher values of them occurred in photoperiods ≥ 16 h, compared with 12L:12D (P > 0.05). Initial and bare stroma weight increased quadratically, while total large white follicle (LWF) number and weight increased linearly and quadratically, with increasing photoperiods (P < 0.05). The higher values of them occurred in 16L:8D and 18L:6D treatments as well as the higher total LWF weight also occurred in 20L:4D, compared with 12L:12D (P > 0.05). Besides, 16.93 and 16.93 h were the optimal photoperiods for bare stroma (follicles ≥ 2 mm in diameter removed) weight and total LWF weight, respectively, calculated from reliable regression equations (R2 ≥ 0.5071). Compared with 12L:12D, the higher levels of estradiol, progesterone, follicle-stimulating hormone (FSH) as well as the higher expressions of estrogen, luteinizing hormone (LH) and progesterone receptors were observed in ≥16 h photoperiods (P < 0.05), while the higher LH level and FSH receptor expression only occurred in 16L:8D and 18L:6D (P < 0.05). In the hypothalamus, higher mRNA expression of gonadotropin-releasing hormone occurred in 16L:8D and 18L:6D groups (P < 0.05). Meanwhile, gonadotropin-inhibitory hormone and prolactin increased in 20-hour photoperiod (P < 0.05), and the latter may be due to theup-regulation of vasoactive intestinal peptide expression (P < 0.05). To sum up, an appropriate photoperiod could improve the performance and reproductive organ and ovarian follicles development through reproductive hormones and their receptors, and 16.56 to 10.93 h is an adequate photoperiod for laying ducks.

Project description:BackgroundThis study aimed to investigate whether the combination of Macleaya cordata extract (MCE) and Bacillus could improve the laying performance and health of laying hens better.MethodsA total of 360 29-week-old Jingbai laying hens were randomly divided into 4 treatments: control group (basal diet), MCE group (basal diet + MCE), Probiotics Bacillus Compound (PBC) group (basal diet + compound Bacillus), MCE + PBC group (basal diet + MCE + compound Bacillus). The feeding experiment lasted for 42 d.ResultsThe results showed that the laying rate and the average daily egg mass in the MCE + PBC group were significantly higher than those in the control group (P < 0.05) and better than the MCE and PBC group. Combination of MCE and Bacillus significantly increased the content of follicle-stimulating hormone (FSH) in the serum and up-regulated the expression of related hormone receptor gene (estrogen receptor-β, FSHR and luteinizing hormone/choriogonadotropin receptor) in the ovary of laying hens (P < 0.05). In the MCE + PBC group, the mRNA expressions of zonula occluden-1, Occludin and mucin-2 in jejunum was increased and the intestinal epithelial barrier detected by transmission electron microscopy was enhanced compared with the control group (P < 0.05). In addition, compared with the control group, combination of MCE and Bacillus significantly increased the total antioxidant capacity and catalase activity (P < 0.05), and down-regulated the mRNA expressions of inflammation-related genes (interleukin-1β and tumor necrosis factor-α) as well as apoptosis-related genes (Caspase 3, Caspase 8 and P53) (P < 0.05). The concentration of acetic acid and butyric acid in the cecum content of laying hens in the MCE + PBC group was significantly increased compared with the control group (P < 0.05).ConclusionsCollectively, dietary supplementation of 600 μg/kg MCE and 5 × 108 CFU/kg compound Bacillus can improve laying performance by improving microbiota to enhance antioxidant capacity and intestinal barrier, regulate reproductive hormones and the concentration of cecal short-chain fatty acids of laying hens, and the combined effect of MCE and Bacillus is better than that of single supplementation.

Project description:A 12-wk study was conducted to investigate the effects of a phytogenic premix (PP) inclusion level on production performance, and the expression of genes relevant for detoxification (aryl hydrocarbon receptor; AhR) and antioxidant capacity (Nuclear factor erythroid 2-related factor 2; Nrf2) in the ovaries of laying hens. The PP consisted of bioactive substances derived from ginger, lemon balm, oregano, and thyme substances (Anco FIT-Poultry). Depending on PP inclusion level (i.e., 0, 500, 750, 1,000, and 1,500 mg/kg diet) in the basal diet, 385 laying hens Hy-Line Brown, 20-wk-old were assigned into 5 treatments: CON, P500, P750, P1000, and P1500, with 7 replicates of 11 hens with ad libitum access to feed and water. Performance parameters were closely monitored on a weekly basis and analyzed in the following 3 experimental periods: 1 to 4 wk, 5 to 8 wk, and 9 to 12 wk of treatment administration (i.e., 21-24, 25-28, and 29-32 wk of layers age, respectively). At the end of the 8th and 12th wk of the experiment (i.e., 28 and 32 wk of layers age), a layer from each replicate was selected, euthanized, the ovaries sampled and stored deep frozen until gene expression analysis. Data were analyzed by ANOVA and means compared using Tukey's honest significant difference test. Polynomial contrasts tested the linear and quadratic effect of PP inclusion levels. Results revealed that PP inclusion, improved (P < 0.05) laying rate and egg mass, compared to CON. Increasing PP inclusion level enhanced laying rate and egg mass, linearly and quadratically and peaked at P1000 (P < 0.05). In the ovaries, the AhR pathway genes assessed were down-regulated (P < 0.05) mainly at P1000 and P750 treatments. In addition, PP related cytoprotective potential was demonstrated via beneficial changes seen for the majority of the Nrf2-pathway genes assessed with the P1000 displaying most significant differences from CON. Conclusively, new data highlighted beneficial cytoprotective effects of PP inclusion on layer ovaries and documented further layer performance, with the inclusion level of 1000 mg PP/kg diet being the most prominent.

Project description:BackgroundSex hormones play important roles in the estrus return of post-weaning sows. Previous studies have demonstrated a complex and bi-directional regulation between sex hormones and gut microbiota. However, the extent to which the gut microbiota affects estrus return of post-weaning sows is largely unknown.ResultsIn this study, we first screened 207 fecal samples from well-phenotyped sows by 16S rRNA gene sequencing and identified significant associations between microbes and estrus return of post-weaning sows. Using metagenomic sequencing data from 85 fecal samples, we identified 37 bacterial species that were significantly associated with estrus return. Normally returning sows were characterized by increased abundances of L. reuteri and P. copri and decreased abundances of B. fragilis, S. suis, and B. pseudolongum. The changes in gut microbial composition significantly altered the functional capacity of steroid hormone biosynthesis in the gut microbiome. The results were confirmed in a validation cohort. Significant changes in sex steroid hormones and related compounds were found between normal and non-return sows via metabolome analysis. An integrated analysis of differential bacterial species, metagenome, and fecal metabolome provided evidence that normal return-associated bacterial species L. reuteri and Prevotella spp. participated in the degradation of pregnenolone, progesterone, and testosterone, thereby promoting estrogen biosynthesis. Furthermore, the microbial metabolites related to sow energy and nutrient supply or metabolic disorders also showed relationships with sow estrus return.ConclusionsAn integrated analysis of differentially abundant bacterial species, metagenome, and fecal metabolome revealed the involvement of L. reuteri and Prevotella spp. in sow estrus return. These findings provide deep insight into the role of gut microbiota in the estrus return of post-weaning sows and the complex cross-talk between gut microbiota and sex hormones, suggesting that the manipulation of the gut microbiota could be an effective strategy to improve sow estrus return after weaning.
