Evaluation of exotic oat (Avena sativa L.) varieties for forage and grain yield in response to different levels of nitrogen and phosphorous.

ABSTRACT: A field experiment was conducted during the Rabi season 2017-2018 (October-March) at the University of Agriculture, Peshawar research farm to examine the influence of different nitrogen (N) and phosphorus (P) levels on two different oat varieties: Australian and Ukrainian. The treatments included control and three levels of nitrogen and phosphorus at 30, 60, and 90 kg ha^-1. The treatments were arranged in randomized complete block design (RCBD) and replicated three times. The findings showed that the oat varieties were significantly different from one another in yield and yield parameters. The Australian variety recorded higher emergence (49 plants m^-2), days to emergence (15 days), days to flowering (122 days), days to maturity (145 days), plant height (142.7 cm), number of leaves (6.03 leaves plant^-1), number of tillers (92.2 tillers m^-1), biological yield (8,179.2 kg ha^-1), and grain yield (3,725.6 kg ha^-1) than the Ukrainian variety. Similarly, different N and P levels, the maximum days to emergence, days to flowering, and days to maturity were recorded in a control plot. The application of 105 kg N + 90 kg P ha^-1 was statistically similar to the application of 105 kg N + 60 kg P ha^-1. Maximum emergence (60 plants m^-2), number of leaves (7.0 leaves plant^-1), plant height (118.6 cm), number of tillers m^-1 (102.6), biological yield (9,687.5 kg ha^-1), and grain yield (4,416.7 kg ha^-1) were determined in Australian variety. Based on the findings of this study, the Australian variety performed better in terms of yield and yield components and the application of N and P fertilizers at the rate of 105 kg N + 60 kg P ha^-1 produced the best results in both oat varieties.

Project description:Calcareous soil contains many problems such as the lack of sources of major and minor elements that are useful for plant growth and development. Plant extracts and nanoparticles are very popular as biostimulants in plant production. Here, the effect of aqueous, non-aqueous and alcoholic oat extracts on the growth, biochemical response of oats leaves and grains grown in experimental fields under new reclamation lands were studied. Moreover, different oat extracts were a pathway through the copper-dependent metal–organic framework (MOFs) to separate bioactive molecules from extracts such as salicylic acid, anthraquinone, and triacylglycerol. Additionally, the separated molecules incorporated in Cu-BTC MOFs and oats extracts missed active molecules were spray applied on oat plants. The results showed that the treated plants showed stimulatory responses in growth and physiology. The treatments improved plant growth and biomass, enhanced total protein, water-soluble carbohydrates, free phenolic compounds content in oat leaves, photosynthesis, and chlorophyll contents. The treatments also improved the level of vitamins E and K, phenolic compounds, and avenanthramides C in the oat grains. Moreover, the treatments showed an improvement in the yield of oats (grain and straw) using water and alcoholic oat extracts in which the active molecules were missed. Our findings demonstrate that Cu-BTC and oats extracts can act as a biostimulant to enhance the biological and chemical properties of oats and increase the yield in calcareous soils. The cytotoxicity study of oats (produced from AE, c@Cu-BTC, and AE-c treatments) was conducted using Vero Cell lines. The anticancer activities of different oat grains were carried out using MCF 7cell lines. The results show that the grains produced from the AE, c@Cu-BTC, and AE-c treatments possessed 94.3, 72.3, and 100% activity towards the cancer cell line. Removal of growth inhibitors from spray solutions increases grain yield and anticancer activity. Project description:BackgroundDeficiency of available forms of phosphorus is common in most soils and causes reduction of crop plants growth and yield. Recently, model plants responses to phosphate (Pi) deficiency have been intensively studied. However, acclimation mechanisms of cereals like oat (Avena sativa L.), to low Pi stress remains not fully understood. Oat plants have been usually cultured on poor soils, with a low nutrient content, but their responses to such conditions are not well known, therefore the main goal of the study was to investigate the mechanisms that enable oat plants to grow under low Pi conditions.MethodsFour oat cultivars (A. sativa, cv. Arab, Krezus, Rajtar and Szakal) were grown for three weeks in a nutrient media with various P sources: inorganic-KH2PO4 (control), organic-phytate (PA) and with no phosphate (-P). The effects of Pi deficiency on the level of P, oat growth parameters, intensity of photosynthesis, plant productivity, root exudation ability, localization, activity and isoforms of acid phosphatases, enzymes involved in Pi mobilization, were estimated. In addition, the effect of mycorrhization on plant growth was also observed.ResultsAll studied oat cultivars grown on Pi-deficient media had significantly decreased Pi content in the tissues. Pi deficiency caused inhibition of shoot growth, but generally it did not affect root elongation; root diameter was decreased, root/shoot ratios increased, whereas PA plants showed a similar growth to control. Photosynthesis rate and productivity parameters decreased under low Pi nutrition, however, sugar content generally increased. Studied oat cultivars did not respond to low Pi via increased exudation of carboxylates from the roots, as pH changes in the growth media were not observed. Pi starvation significantly increased the activity of extracellular and intracellular acid phosphatases (APases) in comparison to the control plants. Three major APase isoforms were detected in oat tissues and the isoform pattern was similar in all studied conditions, usually with a higher level of one of the isoforms under Pi starvation. Generally no significant effects of mycorrhizal colonization on growth of oat cultivars were observed.DiscussionWe postulated that acid phosphatases played the most important role in oat cultivars acclimation to Pi deficiency, especially extracellular enzymes involved in Pi acquisition from soil organic P esters. These APases are mainly located in the epidermis of young roots, and may be released to the rhizosphere. On the other hand, intracellular APases could be involved in fast Pi remobilization from internal sources. Our study showed that oat, in contrast to other plants, can use phytates as the sole source of P. The studied oat cultivars demonstrated similar acclimation mechanisms to Pi deficiency, however, depending on stress level, they can use different pools of acid phosphatases.

Project description:BackgroundOat is an important crop in North America and northern Europe. In Scandinavia, yields are limited by the fact that oat cannot be used as a winter crop. In order to develop such a crop, more knowledge about mechanisms of cold tolerance in oat is required.ResultsFrom an oat cDNA library 9792 single-pass EST sequences were obtained. The library was prepared from pooled RNA samples isolated from leaves of four-week old Avena sativa (oat) plants incubated at +4 degrees C for 4, 8, 16 and 32 hours. Exclusion of sequences shorter than 100 bp resulted in 8508 high-quality ESTs with a mean length of 710.7 bp. Clustering and assembly identified a set of 2800 different transcripts denoted the Avena sativa cold induced UniGene set (AsCIUniGene set). Taking advantage of various tools and databases, putative functions were assigned to 1620 (58%) of these genes. Of the remaining 1180 unclassified sequences, 427 appeared to be oat-specific since they lacked any significant sequence similarity (Blast E values > 10(-10)) to any sequence available in the public databases. Of the 2800 UniGene sequences, 398 displayed significant homology (BlastX E values < or = 10(-10)) to genes previously reported to be involved in cold stress related processes. 107 novel oat transcription factors were also identified, out of which 51 were similar to genes previously shown to be cold induced. The CBF transcription factors have a major role in regulating cold acclimation. Four oat CBF sequences were found, belonging to the monocot cluster of DREB family ERF/AP2 domain proteins. Finally in the total EST sequence data (5.3 Mbp) approximately 400 potential SSRs were found, a frequency similar to what has previously been identified in Arabidopsis ESTs.ConclusionThe AsCIUniGene set will now be used to fabricate an oat biochip, to perform various expression studies with different oat cultivars incubated at varying temperatures, to generate molecular markers and provide tools for various genetic transformation experiments in oat. This will lead to a better understanding of the cellular biology of this important crop and will open up new ways to improve its agronomical properties.
