Engineering proteinaceous colloidosomes as enzyme carriers for efficient and recyclable Pickering interfacial biocatalysis.

ABSTRACT: Despite Pickering interfacial biocatalysis being a popular topic in biphasic biocatalysis, the development of water-in-oil (w/o) emulsion systems stabilized by single particles remains a challenge. For the first time, hydrophobized proteinaceous colloidosomes with magnetic-responsiveness are developed to function as both an enzyme carrier and emulsifier, achieving a breakthrough in protein-based w/o Pickering bioconversion. Enzyme-loaded protein colloidosomes are synthesized by a facile and mild method via emulsion templating. This system exhibits superior catalytic activity to other systems at the oil-water interface. Besides, feasible enzyme recovery and reusability ensure that this novel system can be employed as an efficient and eco-friendly recyclable platform.

Temperature-responsive Pickering high internal phase emulsions for recyclable efficient interfacial biocatalysis.

Project description:The field of biocatalysis is expanding owing to the increasing demand for efficient low-cost green chemical processes. However, a feasible strategy for achieving product separation, enzyme recovery, and high catalytic efficiency in biocatalysis remains elusive. Herein, we present thermoresponsive Pickering high internal phase emulsions (HIPEs) as controllable scaffolds for efficient biocatalysis; these HIPEs demonstrate a transition between emulsification and demulsification depending on temperature. Ultra-high-surface-area Pickering HIPEs were stabilized by Candida antarctica lipase B immobilized on starch particles modified with butyl glycidyl ether and glycidyl trimethyl ammonium chloride, thus simplifying the separation and reuse processes and significantly improving the catalytic efficiency. In addition, the switching temperature can be precisely tuned by adjusting the degree of substitution of the modified starches to meet the temperature demands of various enzymes. We believe that this system provides a green platform for various interfacial biocatalytic processes of industrial interest.

CO<sub>2</sub>-Switchable Hierarchically Porous Zirconium-Based MOF-Stabilized Pickering Emulsions for Recyclable Efficient Interfacial Catalysis.

Project description:Stimuli-responsive Pickering emulsions are recently being progressively utilized as advanced catalyzed systems for green and sustainable chemical conversion. Hierarchically porous metal-organic frameworks (H-MOFs) are regarded as promising candidates for the fabrication of Pickering emulsions because of the features of tunable porosity, high specific surface area and structure diversity. However, CO2-switchable Pickering emulsions formed by hierarchically porous zirconium-based MOFs have never been seen. In this work, a novel kind of the amine-functionalized hierarchically porous UiO-66-(OH)2 (H-UiO-66-(OH)2) has been developed using a post-synthetic modification of H-UiO-66-(OH)2 by (3-aminopropyl)trimethoxysilane (APTMS), 3-(2-aminoethylamino)propyltrimethoxysilane (AEAPTMS) and 3-[2-(2-aminoethylamino)ethylamino]propyl-trimethoxysilane (AEAEAPTMS), and employed as emulsifiers for the construction of Pickering emulsions. It was found that the functionalized H-UiO-66-(OH)2 could stabilize a mixture of toluene and water to give an emulsion even at 0.25 wt % content. Interestingly, the formed Pickering emulsions could be reversibly transformed between demulsification and re-emulsification with alternate addition or removal of CO2. Spectral investigation indicated that the mechanism of the switching is attributed to the reaction of CO2 with amino silane on the MOF and the generation of hydrophilic salts, leading to a reduction in MOF wettability. Based on this strategy, a highly efficient and controlled Knoevenagel condensation reaction has been gained by using the emulsion as a mini-reactor and the emulsifier as a catalyst, and the coupling of catalysis reaction, product isolation and MOF recyclability has become accessible for a sustainable chemical process.

CO<sub>2</sub>-responsive Pickering emulsions stabilized by soft protein particles for interfacial biocatalysis.

Project description:Pickering emulsions are emulsions stabilized by colloidal particles and serve as an excellent platform for biphasic enzymatic catalysis. However, developing simple and green strategies to avoid enzyme denaturation, facilitate product separation, and achieve the recovery of enzyme and colloidal particle stabilizers is still a challenge. This study aimed to report an efficient and sustainable biocatalysis system via a robust CO2/N2-responsive Pickering oil-in-water (o/w) emulsion stabilized solely by pure sodium caseinate (NaCas), which was made naturally in a scalable manner. The NaCas-stabilized emulsion displayed a much higher reaction efficiency compared with conventional CO2/N2-responsive Pickering emulsions stabilized by solid particles with functional groups from polymers or surfactants introduced to tailor responsiveness, reflected by the fact that most enzymes were transferred and enriched at the oil-water interface. More importantly, the demulsification, product separation, and recycling of the NaCas emulsifier as well as the enzyme could be facilely achieved by alternatively bubbling CO2/N2 more than 30 times. Moreover, the recycled enzyme still maintained its catalytic activity, with a conversion yield of more than 90% after each cycle, which was not found in any of the previously reported CO2-responsive systems. This responsive system worked well for many different types of oils and was the first to report on a protein-based CO2/N2-responsive emulsion, holding great promise for the development of more sustainable, green chemical conversion processes for the food, pharmaceutical, and biomedical industries.

pH-Responsive Pickering emulsion stabilized by polymer-coated silica nanoaggregates and applied to recyclable interfacial catalysis.

Project description:We first synthesized a diblock copolymer poly[tert-butyl methacrylate]-b-poly[3-(trimethoxysilyl)propyl methacrylate] (PtBMA-b-PTMSPMA) through reversible addition-fragmentation chain transfer (RAFT) living radical polymerization and grafted it onto fumed silica by converting the PTMSPMA segment to silanol and the PtBMA segment to polymethylacrylic acid (PMAA) in the presence of trifluoroacetic acid in order to obtain PMAA brush-coated silica nanoaggregates P-Si. TEM, DLS, FTIR, and TGA results confirmed the successful modification of the starting materials. The nanoaggregates flocculated and stabilized a toluene-in-water Pickering emulsion at low pH, while the nanoaggregates were well dispersed in water and broke the emulsion under both neutral and basic conditions. Alternatively, the addition of acid/base induced emulsification/demulsification cycles that were sustained for several cycles. Moreover, when the P-Si was mixed with Rh-loaded silica, Rh-Si, the mixture had the same pH-responsive Pickering emulsion behavior as the single P-Si. This Pickering emulsion system can be used in the biphasic interfacial catalytic hydrogenation of olefins and had excellent yields under a hydrogen atmosphere. The yield of Pickering emulsion catalysis rapidly reached more than 99% in 3 h, while that of the demulsified mixture failed to reach 20% in 4 h, which verified the promotion of catalysis by the Pickering emulsion. Base-induced demulsification can be used to separate the products and recycle the catalyst. This pH-responsive Pickering emulsion catalytic system was capable of several cycles of reuse, and there was no significant decrease in catalytic efficiency even after eight cycles.

Selective Colorimetric "Turn-On" Probe for Efficient Engineering of Iminium Biocatalysis.

Project description:The efficient engineering of iminium biocatalysis has drawn considerable attention, with many applications in pharmaceutical synthesis. Here, we report a tailor-made iminium-activated colorimetric "turn-on" probe, specifically designed as a prescreening tool to facilitate engineering of iminium biocatalysis. Upon complexation of the probe with the catalytic Pro-1 residue of the model enzyme 4-oxalocrotonate tautomerase (4-OT), a brightly colored merocyanine-dye-type structure is formed. 4-OT mutants that formed this brightly colored species upon incubation with the probe proved to have a substantial activity for the iminium-based Michael-type addition of nitromethane to cinnamaldehyde, whereas mutants that showed no staining by the probe exhibited no or very low-level "Michaelase" activity. This system was exploited in a solid-phase prescreening assay termed as activated iminium colony staining (AICS) to enrich libraries for active mutants. AICS prescreening reduced the screening effort up to 20-fold. After two rounds of directed evolution, two artificial Michaelases were identified with up to 39-fold improvement in the activity for the addition of nitromethane to cinnamaldehyde, yielding the target γ-nitroaldehyde product with excellent isolated yield (up to 95%) and enantiopurity (up to >99% ee). The colorimetric activation of the turn-on probe could be extended to the class I aldolase 2-deoxy-d-ribose 5-phosphate aldolase, implicating a broader application of AICS in engineering iminium biocatalysis.

Sub-5 nm porous nanocrystals: interfacial site-directed growth on graphene for efficient biocatalysis.

Project description:The direct production of macromolecular scale (sub-5 nm) porous nanocrystals with high surface area has been a considerable challenge over the past two decades. Here we report an interfacial site-directed capping agent-free growth method to directly produce porous ultrasmall (sub-5 nm), fully crystalline, macromolecular scale nanocrystals. The porous sub-5 nm Prussian blue nanocrystals exhibit uniform sizes (∼4 ± 1 nm), high surface area (∼855 m2 g-1), fast electron transfer (rate constant of ∼9.73 s-1), and outstanding sustained catalytic activity (more than 450 days). The nanocrystal-based biointerfaces enable unprecedented sub-nanomolar level recognition of hydrogen peroxide (∼0.5 nM limit of detection). This method also paves the way towards the creation of ultrasmall porous nanocrystals for efficient biocatalysis.

Magnetic MOF microreactors for recyclable size-selective biocatalysis.

Project description:In this contribution we report a synthetic strategy for the encapsulation of functional biomolecules within MOF-based microcapsules. We employ an agarose hydrogel droplet Pickering-stabilised by UiO-66 and magnetite nanoparticles as a template around which to deposit a hierarchically structured ZIF-8 shell. The resulting microcapsules are robust, highly microporous and readily attracted to a magnet, where the hydrogel core provides a facile means to encapsulate enzymes for recyclable size-selective biocatalysis.

Pickering Interfacial Catalysis-Knoevenagel Condensation in Magnesium Oxide-Stabilized Pickering Emulsion.

Project description:In the present study, a novel catalytic route for the Knoevenagel condensation reaction has been developed by Pickering interfacial catalysis using magnesium oxide (MgO) as both an emulsion stabilizer and a base catalyst. MgO was prepared by the precipitation method using sodium hydroxide or ammonium hydroxide as the precipitating agent and calcined at different temperatures. The calcined samples were characterized by XRD, SEM, TEM, AFM, BET, and DLS techniques. The catalytic application of the emulsions stabilized by MgO was investigated for the Knoevenagel condensation reaction of benzaldehyde and its derivatives with malononitrile. All of the reactions were carried out at an ambient temperature (30 °C) under static conditions without stirring. Both the emulsion-stabilizing ability and the catalytic activity of MgO were found to be affected by the method of preparation, calcination temperature, and the nature of the oil phase. It was observed that the method of preparation varied the texture and morphology of MgO and thus the stability and droplet size of the emulsion formed. This was further reflected in the catalytic activity. The highest yield (87%) of the condensation product was obtained with MgO prepared by precipitation using a strong base (NaOH) and further calcined at 400 °C. The developed catalytic system offers several green chemistry advantages such as reusable solid-base catalyst and use of a single material as both emulsion stabilizer and catalyst. Room-temperature reaction under static conditions is an additional advantage of the developed catalytic system.

Engineering enzyme conformation within liquid-solid hybrid microreactors for enhanced continuous-flow biocatalysis.

Project description:The artificial engineering of an enzyme's structural conformation and dynamic properties to promote its catalytic activity and stability outside cellular environments is highly pursued in industrial biotechnology. Here, we describe an elegant strategy of combining the rationally designed liquid-solid hybrid microreactor with a tailor-made polyethylene glycol functional ionic liquid (PEG-IL) microenvironment to exercise a high level of control over the configuration of enzymes for practical continuous-flow biocatalysis. As exemplified by a lipase driven kinetic resolution reaction, the obtained system exhibits a 2.70 to 30.35-fold activity enhancement compared to their batch or traditional IL-based counterparts. Also, our results demonstrate that the thermal stability of encapsulated lipase can be significantly strengthened in the presence of PEG groups, showcasing a long-term continuous-flow stability even up to 1000 h at evaluated temperature of 60 oC. Through systematic experiment and molecular dynamics simulation studies, the conformational changes of the active site cavity in the modified lipases are correlated with enzymatic properties alteration, and the pronounced effects of PEG-groups in stabilizing enzyme's secondary structures by delaying unfolding at elevated temperatures are identified. We believe that this study will guide the design of high-performance enzymatic systems, promoting their utilization in real-world biocatalysis applications.

Nanoscale interfacial engineering enables highly stable and efficient perovskite photovoltaics.

Project description:We present a facile molecular-level interface engineering strategy to augment the long-term operational and thermal stability of perovskite solar cells (PSCs) by tailoring the interface between the perovskite and hole transporting layer (HTL) with a multifunctional ligand 2,5-thiophenedicarboxylic acid. The solar cells exhibited high operational stability (maximum powering point tracking at one sun illumination) with a stabilized T S80 (the time over which the device efficiency reduces to 80% after initial burn-in) of ≈5950 h at 40 °C and a stabilized power conversion efficiency (PCE) over 23%. The origin of high device stability and performance is correlated to the nano/sub-nanoscale molecular level interactions between ligand and perovskite layer, which is further corroborated by comprehensive multiscale characterization. These results provide insights into the modulation of the grain boundaries, local density of states, surface bandgap, and interfacial recombination. Chemical analysis of aged devices showed that molecular passivation suppresses interfacial ion diffusion and inhibits the photoinduced I2 release that irreversibly degrades the perovskite. The interfacial engineering strategies enabled by multifunctional ligands can expedite the path towards stable PSCs.
