Compromised placental function or premature loss has been linked to diverse neurodevelopmental disorders. Here we show that placenta allopregnanolone (ALLO), a progesterone-derived GABA-A receptor (GABA<sub>A</sub>R) modulator, reduction alters neurodevelopment in a sex-linked manner.
