Thermally driven fission of protocells.

ABSTRACT: We propose a simple mechanism for the self-replication of protocells. Our main hypothesis is that the amphiphilic molecules composing the membrane bilayer are synthesized inside the protocell through exothermic chemical reactions. The slow increase of the inner temperature forces the hottest molecules to move from the inner leaflet to the outer leaflet of the bilayer. Because of this outward translocation flow, the outer leaflet grows faster than the inner leaflet. This differential growth increases the mean curvature and amplifies any local shrinking of the protocell until it splits in two. The proposed model, based on mere laws of physics, is a step in the study of the origin of life, as well as a clue for a better understanding of cell proliferation in cancer.

Competition between model protocells driven by an encapsulated catalyst.

Project description:The advent of Darwinian evolution required the emergence of molecular mechanisms for the heritable variation of fitness. One model for such a system involves competing protocell populations, each consisting of a replicating genetic polymer within a replicating vesicle. In this model, each genetic polymer imparts a selective advantage to its protocell by, for example, coding for a catalyst that generates a useful metabolite. Here, we report a partial model of such nascent evolutionary traits in a system that consists of fatty-acid vesicles containing a dipeptide catalyst, which catalyses the formation of a second dipeptide. The newly formed dipeptide binds to vesicle membranes, which imparts enhanced affinity for fatty acids and thus promotes vesicle growth. The catalysed dipeptide synthesis proceeds with higher efficiency in vesicles than in free solution, which further enhances fitness. Our observations suggest that, in a replicating protocell with an RNA genome, ribozyme-catalysed peptide synthesis might have been sufficient to initiate Darwinian evolution.

Non-equilibrium conditions inside rock pores drive fission, maintenance and selection of coacervate protocells.

Project description:Key requirements for the first cells on Earth include the ability to compartmentalize and evolve. Compartmentalization spatially localizes biomolecules from a dilute pool and an evolving cell, which, as it grows and divides, permits mixing and propagation of information to daughter cells. Complex coacervate microdroplets are excellent candidates as primordial cells with the ability to partition and concentrate molecules into their core and support primitive and complex biochemical reactions. However, the evolution of coacervate protocells by fusion, growth and fission has not yet been demonstrated. In this work, a primordial environment initiated the evolution of coacervate-based protocells. Gas bubbles inside heated rock pores perturb the coacervate protocell distribution and drive the growth, fusion, division and selection of coacervate microdroplets. Our findings provide a compelling scenario for the evolution of membrane-free coacervate microdroplets on the early Earth, induced by common gas bubbles within heated rock pores.

Applicability of Taylor's hypothesis in thermally driven turbulence.

Project description:In this paper, we show that, in the presence of large-scale circulation (LSC), Taylor's hypothesis can be invoked to deduce the energy spectrum in thermal convection using real-space probes, a popular experimental tool. We perform numerical simulation of turbulent convection in a cube and observe that the velocity field follows Kolmogorov's spectrum (k-5/3). We also record the velocity time series using real-space probes near the lateral walls. The corresponding frequency spectrum exhibits Kolmogorov's spectrum (f-5/3), thus validating Taylor's hypothesis with the steady LSC playing the role of a mean velocity field. The aforementioned findings based on real-space probes provide valuable inputs for experimental measurements used for studying the spectrum of convective turbulence.

Chirality-Induced Magnetoresistance Due to Thermally Driven Spin Polarization.

Project description:Chirality-induced current-perpendicular-to-plane magnetoresistance (CPP-MR) originates from current-induced spin polarization in molecules. The current-induced spin polarization is widely recognized as a fundamental principle of chiral-induced spin selectivity (CISS). In this study, we investigate chirality-induced current-in-plane magnetoresistance (CIP-MR) in a chiral molecule/ferromagnetic metal bilayer at room temperature. In contrast to CPP-MR, CIP-MR observed in the present study requires no bias charge current through the molecule. The temperature dependence of CIP-MR suggests that thermally driven spontaneous spin polarization in chiral molecules is the key to the observed MR. The novel MR is consistent with recent CISS-related studies, that is, chiral molecules in contact with a metallic surface possess a finite spin polarization.

Robust rotation of rotor in a thermally driven nanomotor.

Project description:In the fabrication of a thermally driven rotary nanomotor with the dimension of a few nanometers, fabrication and control precision may have great influence on rotor's stability of rotational frequency (SRF). To investigate effects of uncertainty of some major factors including temperature, tube length, axial distance between tubes, diameter of tubes and the inward radial deviation (IRD) of atoms in stators on the frequency's stability, theoretical analysis integrating with numerical experiments are carried out. From the results obtained via molecular dynamics simulation, some key points are illustrated for future fabrication of the thermal driven rotary nanomotor.

Thermally Driven Continuous Rolling of a Thick-Walled Cylindrical Rod.

Project description:Self-sustained motion can take advantage of direct energy extraction from a steady external environment to maintain its own motion, and has potential applications in energy harvesting, robotic motion, and transportation. Recent experiments have found that a thermally responsive rod can perform self-sustained rolling on a flat hot plate with an angular velocity determined by the competition between the thermal driving moment and the friction moment. A rod with a hollow cross section tends to greatly reduce the frictional resistance, while promising improvements in thermal conversion efficiency. In this paper, through deriving the equilibrium equations for steady-state self-sustained rolling of the thick-walled cylindrical rod, estimating the temperature field on the rod cross-section, and solving the analytical solution of the thermally induced driving moment, the dynamic behavior of the thermally driven self-sustained rolling of the thick-walled cylindrical rod is theoretically investigated. In addition, we investigate in detail the effects of radius ratio, heat transfer coefficient, heat flux, contact angle, thermal expansion coefficient, and sliding friction coefficient on the angular velocity of the self-sustained rolling of the thick-walled cylindrical rod to obtain the optimal ratio of internal and external radius. The results are instructive for the application of thick-walled cylindrical rods in the fields of waste heat harvesters and soft robotics.

Engineering Polymersome Protocells.

Project description:The field of biomimicry is embracing the construction of complex assemblies that imitate both biological structure and function. Advancements in the design of these mimetics have generated a growing vision for creating an artificial or proto- cell. Polymersomes are vesicles that can be made from synthetic, biological or hybrid polymers and can be used as a model template to build cell-like structures. In this perspective, we discuss various areas where polymersomes have been used to mimic cell functions as well as areas in which the synthetic flexibility of polymersomes would make them ideal candidates for a biomembrane mimetic. Designing a polymersome that comprehensively displays the behaviors discussed herein has the potential to lead to the development of an autonomous, responsive particle that resembles the intelligence of a biological cell.

Thermally Driven Membrane Phase Transitions Enable Content Reshuffling in Primitive Cells.

Project description:Self-assembling single-chain amphiphiles available in the prebiotic environment likely played a fundamental role in the advent of primitive cell cycles. However, the instability of prebiotic fatty acid-based membranes to temperature and pH seems to suggest that primitive cells could only host prebiotically relevant processes in a narrow range of nonfluctuating environmental conditions. Here we propose that membrane phase transitions, driven by environmental fluctuations, enabled the generation of daughter protocells with reshuffled content. A reversible membrane-to-oil phase transition accounts for the dissolution of fatty acid-based vesicles at high temperatures and the concomitant release of protocellular content. At low temperatures, fatty acid bilayers reassemble and encapsulate reshuffled material in a new cohort of protocells. Notably, we find that our disassembly/reassembly cycle drives the emergence of functional RNA-containing primitive cells from parent nonfunctional compartments. Thus, by exploiting the intrinsic instability of prebiotic fatty acid vesicles, our results point at an environmentally driven tunable prebiotic process, which supports the release and reshuffling of oligonucleotides and membrane components, potentially leading to a new generation of protocells with superior traits. In the absence of protocellular transport machinery, the environmentally driven disassembly/assembly cycle proposed herein would have plausibly supported protocellular content reshuffling transmitted to primitive cell progeny, hinting at a potential mechanism important to initiate Darwinian evolution of early life forms.

Geometric catalysis of membrane fission driven by flexible dynamin rings.

Project description:Biological membrane fission requires protein-driven stress. The guanosine triphosphatase (GTPase) dynamin builds up membrane stress by polymerizing into a helical collar that constricts the neck of budding vesicles. How this curvature stress mediates nonleaky membrane remodeling is actively debated. Using lipid nanotubes as substrates to directly measure geometric intermediates of the fission pathway, we found that GTP hydrolysis limits dynamin polymerization into short, metastable collars that are optimal for fission. Collars as short as two rungs translated radial constriction to reversible hemifission via membrane wedging of the pleckstrin homology domains (PHDs) of dynamin. Modeling revealed that tilting of the PHDs to conform with membrane deformations creates the low-energy pathway for hemifission. This local coordination of dynamin and lipids suggests how membranes can be remodeled in cells.

Selection for replicases in protocells.

Project description:We consider a world of nucleotide sequences and protocells. The sequences have the property of spontaneous self-replication. Some sequences - so-called replicases - have enzymatic activity in the sense of enhancing the replication rate of all (or almost all) sequences. In a well-mixed medium, natural selection would not favor such replicases because their presence equally benefits sequences with or without replicase activity. Here we show that protocells can select for replicases. We assume that sequences replicate within protocells and that protocells undergo spontaneous division. This leads to particular population structures which can augment the abundance of replicases. We explore various assumptions regarding replicase activity and protocell division. We calculate the error threshold that is compatible with selecting for replicases.
