The mitochondrial genome of Nemalecium lighti (Hydrozoa, Leptothecata).

ABSTRACT: The hydrozoan species Nemalecium lighti (Hargitt, 1924) is widely distributed in tropical marine waters around the world. Here we report the complete linear mitochondrial genome of N. lighti from Sint Eustatius (Lesser Antilles). The mitochondrial genome with a length of 14,320 bp encodes for 13 protein-coding genes, two tRNA genes, and two rRNA genes. Gene arrangement differs from that found in other species of the same taxonomic order and a phylogenetic analysis shows that based on mitochondrial genes, N. lighti clusters outside of the Leptothecata, rendering the order paraphyletic.

Towards a phylogenetic classification of Leptothecata (Cnidaria, Hydrozoa).

Project description:Leptothecata are hydrozoans whose hydranths are covered by perisarc and gonophores and whose medusae bear gonads on their radial canals. They develop complex polypoid colonies and exhibit considerable morphological variation among species with respect to growth, defensive structures and mode of development. For instance, several lineages within this order have lost the medusa stage. Depending on the author, traditional taxonomy in hydrozoans may be either polyp- or medusa-oriented. Therefore, the absence of the latter stage in some lineages may lead to very different classification schemes. Molecular data have proved useful in elucidating this taxonomic challenge. We analyzed a super matrix of new and published rRNA gene sequences (16S, 18S and 28S), employing newly proposed methods to measure branch support and improve phylogenetic signal. Our analysis recovered new clades not recognized by traditional taxonomy and corroborated some recently proposed taxa. We offer a thorough taxonomic revision of the Leptothecata, erecting new orders, suborders, infraorders and families. We also discuss the origination and diversification dynamics of the group from a macroevolutionary perspective.

Complete mitochondrial genome of the hydrozoan jellyfish Blackfordia virginica Mayer, 1910 (Cnidaria; Hydrozoa; Leptothecata) with phylogenetic analysis.

Project description:In this study, we analyzed the complete mitochondrial genome of the hydrozoan jellyfish Blackfordia virginica. The genome was a linear form (15,109 bp long, 73.6% AT), including 13 protein-coding genes (cox2, atp8, atp6, cox3, nad2, nad5, nad6, nad3, nad4L, nad1, nad4, cytB, and cox1), 2 tRNAs (tRNA-Met and tRNA-Trp), and 2 rRNAs (12S and 16S RNA). The genome structure of the B. virginica was completely identical to mitochondrial genomes of other hydrozoans that belonged to Leptothecata and Anthoathecata. Molecular phylogenetic analysis within hydrozoan species showed that B. virginica was the closest to the hydrozoan Laomedea flexuosa.

Two new species of Hydromedusae from Queensland, Australia (Hydrozoa, Leptothecata).

Project description:Two new species of small hydromedusae were found during routine monitoring in coastal waters of eastern Australia and are here described. The first, Melicertissaantrichardsoni Uribe-Palomino & Gershwin, sp. n., from Moreton Bay, Queensland, is placed in its genus because of its possession of both cordyli and eight-fold symmetry. It differs from its congeners in two conspicuous features: firstly, having small, oval split gonads located adjacent to the base of the stomach, and secondly, in its extremely small size at maturity (2 mm bell diameter, compared to the next smallest species at 7 mm). Moreover, it possesses a unique combination of other characters. This species appears to be endemic to Moreton Bay. The second new species, Paraloveniayongalensis Gershwin & Uribe-Palomino, sp. n., from the Great Barrier Reef, Queensland, is placed in its genus because of its two opposite normal tentacles and two opposite marginal clusters of cirri. It differs from its congeners primarily in a more rounded body than the others; the shape, length, and position of its short spindle-shaped, distal gonads; possession of subumbrellar nematocyst clusters; and possession of statocysts. These discoveries bring the total number of Melicertissa species to eight and the total number of Paralovenia species to three. The discovery of these two micromedusae underscores the need for further examination of the often-ignored minute and/or gelatinous fauna.

First record of two Leptothecata medusae (Cnidaria, Hydrozoa) in Colombia with annotations on their distribution and ecology.

Project description:BackgroundHydromedusae are a group of planktonic cnidarians that represent the medusoid phase in the life cycle of most members of the class Hydrozoa, whose primary function is to produce and release gametes. These organisms are generally small and translucent, with slight pigmentation, except for those that inhabit great depths and exhibit the typical body shape of a jellyfish. In Colombia, studies on this group are limited due to the scarcity of updated taxonomic information and the small number of expert scientists. The most recent inventory records 71 species in the waters of the Pacific and Atlantic Oceans.New informationIn this study, we report for the first time the presence of Eucheilotaduodecimalis and Eutoninascintillans in Colombia, based on 28 zooplankton samples collected from seven stations between February and May 2024, using horizontal tows with a simple conical zooplankton net. Our research emphasises the significance of scientific exploration in new geographic regions and provides valuable data on the distribution and ecology of these species, contributing to a deeper understanding of their population dynamics.

Phylogenetic analysis of higher-level relationships within Hydroidolina (Cnidaria: Hydrozoa) using mitochondrial genome data and insight into their mitochondrial transcription.

Project description:Hydrozoans display the most morphological diversity within the phylum Cnidaria. While recent molecular studies have provided some insights into their evolutionary history, sister group relationships remain mostly unresolved, particularly at mid-taxonomic levels. Specifically, within Hydroidolina, the most speciose hydrozoan subclass, the relationships and sometimes integrity of orders are highly unsettled. Here we obtained the near complete mitochondrial sequence of twenty-six hydroidolinan hydrozoan species from a range of sources (DNA and RNA-seq data, long-range PCR). Our analyses confirm previous inference of the evolution of mtDNA in Hydrozoa while introducing a novel genome organization. Using RNA-seq data, we propose a mechanism for the expression of mitochondrial mRNA in Hydroidolina that can be extrapolated to the other medusozoan taxa. Phylogenetic analyses using the full set of mitochondrial gene sequences provide some insights into the order-level relationships within Hydroidolina, including siphonophores as the first diverging clade, a well-supported clade comprised of Leptothecata-Filifera III-IV, and a second clade comprised of Aplanulata-Capitata s.s.-Filifera I-II. Finally, we describe our relatively inexpensive and accessible multiplexing strategy to sequence long-range PCR amplicons that can be adapted to most high-throughput sequencing platforms.

The mitochondrial genome was a linear form (15,752 bp long, 70.4% AT), consisting of 13 protein coding genes (cox1, cox2, atp8, atp6, cox3, nad2, nad5, nad6, nad3, nad4L, nad1, nad4, and cytB), two tRNAs (tRNA-Met and tRNA-Trp), and two rRNAs (12S and 16S). Mitochondrial gene arrangement of the S. saltatrix was completely identical to already-known mitochondrial genomes of hydrozoans. Molecular phylogenetic analysis using 13 protein-coding genes showed that S. saltatrix was closely related to the hydrozoan Clava multicornis.

The linear mitochondrial genome of commensal hydroid Eutima japonica (Cnidaria, Hydrozoa, Eirenidae).

Project description:Here, we present the whole mitochondrial genome of commensal hydroid Eutima japonica McCrady 1859 (family Eirinidae); this is the first specimen of the family to have its mitogenome sequenced. The linear mitogenome is 15,315 bp in length and consists of 13 protein-coding genes (PCGs), large and small ribosomal subunits (rRNA), methionine and tryptophan transfer RNA (tRNA) genes (trnM and trnW), and a partial copy of cytochrome oxidase subunit I (cox1) pseudogene, as is typical for the class Hydrozoa. Nucleotide sequences of two cox1 genes at two ends of the linear mitogenome form a part of inverted terminal repeat. The overall genomic structure and gene arrangement of 13 PCGs were identical to the reported mitochondrial genomes of hydrozoans, except for the positions of two tRNA genes. Phylogenetic analysis of E. japonica 13 PCGs and other cnidarians recovers a closest relationship with the derived cluster of two hydrozoans, Laomedea flexuosa and Obelia longissimi within Leptothecata.

Project description:In this study, we sequenced and analyzed the complete mitochondrial genome of Cladonema multiramosum Zhou et al., 2022 from Fujian, China. The length of the linear mitochondrial genome is 15164 bp, containing 13 protein-coding genes (cox2, atp8, atp6, cox3, nad2, nad5, nad6, nad3, nad4L, nad1, nad4, cob, cox1), two tRNAs (trnM and trnW) and 2 rRNAs (12S and 16S). The arrangement of mitogenomes show some little differences in different hydrozoan groups. The phylogenetic analysis of 13 protein coding genes (PCGs) in Cnidarians showed that C. multiramosum was closely related to Cladonema pacificum.

The mtgenome was a complete linear form (15,047 bp in length, 30.9% A, 42.1% T, 12.5% C, and 14.5% G), including 13 protein coding genes (PCGs) (cox1, cox2, cox3, atp6, atp8, nad1, nad2, nad3, nad4, nad4L, nad5, nad6, and cytb), 2 tRNAs (tRNAMet and tRNATrp), and 2 rRNAs (12S and 16S rRNA). The genome structure of the T. lata was completely identical to those of other species within the subclass Hydroidolina. In addition, our molecular phylogenetic analysis using 13 PCGs within hydrozoans showed that T. lata was the closest to Turritopsis dohrnii.

Project description:Eirene ceylonensis, a hydrozoan jellyfish species with a complex polymorphic life cycle, is widely distributed in the Chinese coastal sea. In this study, we conducted sequencing and analysis of the first complete mitochondrial genome of E. ceylonensis, obtained from the coastal sea of Qinhuangdao, China. The linear mitochondrial genome is 14,997 bp in length with the overall AT content being 72.8%, encoding 13 protein-coding genes (PCGs), two transfer RNA (tRNA) genes (tRNA-Met and tRNA-Trp) and two ribosomal RNA (rRNA) genes (rrnS and rrnL). Phylogenetic analysis of 13 PCGs suggests that the E. ceylonensis is closely related to Laomedea flexuosa. The availability of the complete mitochondrial genome of E. ceylonensis will be useful for studying the evolutionary relationships of hydrozoan jellyfish species.
