Snails associated with the coral-killing sponge Terpios hoshinota in Okinawa Island, Japan.

ABSTRACT: Terpios hoshinota is a thin encrusting sponge that overgrows live scleractinian corals and it is linked to coral loss in many reefs. However, our knowledge of the species associated with this sponge species is poor. During a periodical survey of T. hoshinota in 2020, we found tiny snails crawling on the sponge in the subtropical waters around Okinawa Island, Japan. We observed egg capsules inside the sponge tissue and veliger larvae released from the egg capsules. Molecular analyses of both the snails and veliger larvae (cytochrome oxidase I, COI) showed that they were identical and belonged to Joculator sp. (family Cerithiopsidae). There was no direct observation of predation on the sponge by this snail; however, to the best of our knowledge, this is the first report on a close association between a snail and the sponge T. hoshinota.

Contact Reactions between Individuals of the Coral-killing sponge, <i>Terpios hoshinota</i>.

Project description:Outbreaks of the coral-killing sponge Terpios hoshinota Rützler and Muzik, 1993 have become a threat to corals and result in coral reef deterioration. This species has an increasing distribution in the Indo-Pacific Ocean and thrives in patches on some reefs in Okinawa, Japan. However, the dispersal process and mechanisms involved remain unknown. We observed the self and non-self recognition capabilities of T. hoshinota by performing contact assays in aquarium and in the field. In the contact assays (indirect and direct contact), allogeneic sets did not fuse and showed a rejection reaction as they formed boundaries (approx. 0.2 mm width) between their tissues. Although the initial reaction between individuals involved adhesion in allogeneic sets, the two individuals remained distant from each other. Histological observations showed that soft tissues (such as collagen) were not present in the boundary zones. These boundaries were maintained for more than 2 weeks. Boundary formations were also confirmed at three field sites in Okinawa, Japan. Our results suggest that T. hoshinota can distinguish self and non-self individuals. Contact assays are a useful method for evaluating the spatial distribution and local population structures of T. hoshinota in coral reefs.

Short-term in situ shading effectively mitigates linear progression of coral-killing sponge Terpios hoshinota.

Project description:The coral-killing sponge, Terpios hoshinota is a global invasive species that has conquered coral patches within a short span of time, which has led to a significant decline in living coral cover at various geographical locations. In this study, we surveyed the linear progression and impact of the Terpios invasion on live coral patches along Palk Bay, Indian Ocean, from August 2013 to August 2015. The field inventory revealed an extensive fatality rate of 76% as a result of Terpios outbreak. Experimental findings showed that symbiotic cyanobacteria act as a nutritional factory for the aggressive growth of Terpios. Shading hypothetically impairs the nutritional symbiont of the invasive species: the effect of sunlight on cyanobacterial biomass and its influence on Terpios progression over live coral patches was tested through in situ shading experiments. This study showed that artificial shading with cotton fabric could effectively mitigate sponge growth on live coral without affecting coral homeostasis.

Disappearance and Return of an Outbreak of the Coral-killing Cyanobacteriosponge Terpios hoshinota in Southern Japan.

Project description:Masashi Yomogida, Masaru Mizuyama, Toshiki Kubomura, and James Davis Reimer (2017) Terpios hoshinota is cyanobacteriosponge that can cause serious damage to coral reef ecosystems by undergoing rapid breakouts in which it smothers and encrusts hard substrates, killing living sessile benthic organisms and reducing biodiversity of the a ected area. The reasons for these outbreaks are still unclear, as are long-term prognoses of affected reefs. Some reports have suggested outbreaks may not be permanent, but very little long-term monitoring information exists. In this study, we report on a T. hoshinota outbreak (~24% coverage) at Yakomo, Okinoerabu-jima Island, Kagoshima, Japan between 2010 to 2014. In this period, the existing outbreak was seen to almost completely disappear (~0%) after unusually severe Typhoon Songda passed by in early June 2011. After this, macroalgae and a cyanobacterial bloom became the dominant benthos, but by September 2014, T. hoshinota coverage had recovered to approximately half of its pre-typhoon coverage, suggesting the conditions that had caused the outbreak still persisted at Yakomo. While the conditions promoting T. hoshinota growth at this site remain uncertain, it appears that subtropical typhoons could play an important role in the dynamics of T. hoshinota outbreaks and disappearances.

Prevalence, complete genome, and metabolic potentials of a phylogenetically novel cyanobacterial symbiont in the coral-killing sponge, Terpios hoshinota.

Project description:Terpios hoshinota is an aggressive, space-competing sponge that kills various stony corals. Outbreaks of this species have led to intense damage to coral reefs in many locations. Here, the first large-scale 16S rRNA gene survey across three oceans revealed that bacteria related to the taxa Prochloron, Endozoicomonas, SAR116, Ruegeria, and unclassified Proteobacteria were prevalent in T. hoshinota. A Prochloron-related bacterium was the most dominant and prevalent cyanobacterium in T. hoshinota. The complete genome of this uncultivated cyanobacterium and pigment analysis demonstrated that it has phycobiliproteins and lacks chlorophyll b, which is inconsistent with the definition of Prochloron. Furthermore, the cyanobacterium was phylogenetically distinct from Prochloron, strongly suggesting that it should be a sister taxon to Prochloron. Therefore, we proposed this symbiotic cyanobacterium as a novel species under the new genus Candidatus Paraprochloron terpiosi. Comparative genomic analyses revealed that 'Paraprochloron' and Prochloron exhibit distinct genomic features and DNA replication machinery. We also characterized the metabolic potentials of 'Paraprochloron terpiosi' in carbon and nitrogen cycling and propose a model for interactions between it and T. hoshinota. This study builds a foundation for the study of the T. hoshinota microbiome and paves the way for better understanding of ecosystems involving this coral-killing sponge.

Population genetics and demography of the coral-killing cyanobacteriosponge, <i>Terpios hoshinota,</i> in the Indo-West Pacific.

Project description:The first occurrence of the cyanobacteriosponge Terpios hoshinota was reported from coral reefs in Guam in 1973, but was only formally described in 1993. Since then, the invasive behavior of this encrusting, coral-killing sponge has been observed in many coral reefs in the West Pacific. From 2015, its occurrence has expanded westward to the Indian Ocean. Although many studies have investigated the morphology, ecology, and symbiotic cyanobacteria of this sponge, little is known of its population genetics and demography. In this study, a mitochondrial cytochrome oxidase I (COI) fragment and nuclear ribosomal internal transcribed spacer 2 (ITS2) were sequenced to reveal the genetic variation of T. hoshinota collected from 11 marine ecoregions throughout the Indo-West Pacific. Both of the statistical parsimony networks based on the COI and nuclear ITS2 were dominated by a common haplotype. Pairwise F ST and Isolation-by-distance by Mantel test of ITS2 showed moderate gene flow existed among most populations in the marine ecoregions of West Pacific, Coral Triangle, and Eastern Indian Ocean, but with a restricted gene flow between these regions and Maldives in the Central Indian Ocean. Demographic analyses of most T. hoshinota populations were consistent with the mutation-drift equilibrium, except for the Sulawesi Sea and Maldives, which showed bottlenecks following recent expansion. Our results suggest that while long-range dispersal might explain the capability of T. hoshinota to spread in the IWP, stable population demography might account for the long-term persistence of T. hoshinota outbreaks on local reefs.

Draft genome of Myxosarcina sp. strain GI1, a baeocytous cyanobacterium associated with the marine sponge Terpios hoshinota.

Project description:To date, genome sequences (complete or in draft form) from only six baeocytous cyanobacteria in four genera have been reported: Xenococcus, Chroococcidiopsis, Pleurocapsa, and Stanieria. To expand our knowledge on the diversity of baeocytous cyanobacteria, this study sequenced the genome of GI1, which is a Myxosarcina-like baeocytous cyanobacterium. GI1 is of interest not only because of its phylogenetic niche, but also because it is a cyanobiont isolated from the marine cyanobacteriosponge Terpios hoshinota, which has been shown to cause the death of corals. The ~7 Mb draft GI1 genome contains 6,891 protein-coding genes and 62 RNA genes. A comparison of genomes among the sequenced baeocytous cyanobacterial strains revealed the existence or absence of numerous discrete genes involved in nitrogen metabolism. It will be interesting to determine whether these genes are important for cyanobacterial adaptations and interactions between cyanobionts and their marine sponge hosts.

Two new species of Asellota (Crustacea, Isopoda) from coral reefs on Iriomote Island, Okinawa, Japan.

Project description:Pleurocope iriomotensis sp. n. and Prethura tuberculata sp. n. are described from Iriomote Island, Ryukyu Archipelago, southern Japan. These are the first records of Pleurocope from the Pacific and of Prethura from the Asian Pacific coast. Pleurocope iriomotensis differs from its congeners in having lateral spine-like processes on pereonite 4 and coxal plates of pereonite 7. Prethura tuberculata can be distinguished from its single congener in having a lateral short projection of protopod of pleopod 2.
