Project description:Atmospheric particulate matter (PM) is a recognized risk factor for the global burden of disease in human populations. We are presenting here the application of toxicogenomics in the evaluation of the toxic effects of organic content of atmospheric particle matter (PM), from urban and rural environments (city of Barcelona and village of La Pobla, NE Spain), using the developing zebrafish embryo. The main goal is to identify the metabolic pathways involved in the adverse effects observed in zebrafish embryos exposed to PM organic content from urban and rural environments, also allowing the selection of genes of interest that are differentially expressed. The relevance of particle size to the PM toxicity is also addressed. Indeed, the zebrafish embryos were exposed to PM of aerodynamic diameter larger than 7.2 μm and smaller than 0.5 μm (PM10 and PM0.5, respectively). PM0.5 concentrated biological and toxic activities linked to organic substances. Transcriptomic analyses showed strong induction of the AhR signalling pathway (a.k.a. dioxin-like activity) for embryos exposed to both rural and urban extracts, correlating with the concentrations of PAHs. Urban extracts, with strong contribution of traffic emissions, specifically de-regulated oxidative stress-related genes, as well as pancreatic and eye-lens specific genes, two organs known to be affected by air pollution in humans. Exposure to rural extracts, with high contribution of wood burning emissions, affected genes implicated in basic cellular functions, in agreement with their strong embryotoxicity. Extracts from rural and urban samples elicited both common and specific transcriptome responses, suggesting different potentially adverse outcomes depending on PM source and composition. The PM filter samples used in the present study were PM10 and PM0.5 from the urban site - total of 4 samples from 2 sampling months; and PM0.5 from the rural site - total of 2 samples from 2 sampling months (PM10 samples from the rural site were not tested due to their very low concentration in organic compounds, and insignificant biological activity, previously measured). Sampling occurred during the cold periods of the year 2012/2013. The organic fraction of PM samples was extracted by sonication using a mixture of dichloromethane:methanol. Then, the extracts were filtered, evaporated and re-dissolved in Dimethyl sulfoxide (DMSO). Zebrafish fertilized eggs were exposed to PM organic extracts (0.2% DMSO) from the urban and rural sites from 24 to 120h post-fertilization. Total RNA was isolated from whole embryos (pools of 20 individuals), using Trizol reagent protocol (Invitrogen Life Technologies, Carlsberg, CA). The microarray study was performed using the Agilent Two-colour D. rerio Oligo Microarray v3 platform, following the Agilent Microarray â?? Based Gene Expression Analysis protocol. Three biological replicates were performed for each PM sample and controls. Biological replicates, a technical replicate and a self-to-self, were simultaneously amplified and labelled using Cyanine 3 (Cy3) and Cyanine 5 (Cy5) dye. After cRNA purification (RNeasy Kit, Quiagen GmbH, Hilden, Germany), the cRNA concentration and labelling were quantified in the NanoDrop spectrophotometer, obtaining incorporation rates in the range of 15-20pmol of cyanide dye/µl cRNA and yield values > 0.825ug, as recommended. cRNA was fragmented and hybridized in 4x44K arrays, for 17h at 65oC. After the hybridization the array slides were washed, and immediately scanned using Microarray Scanner Agilent G2505C system. Data was extracted using Agilent Feature Extraction Software v10.5.1.1, and the quality of microarray data was evaluated using the Quality Control report provided by Agilent Software. Based on the microarray data analysis, 22 primers were selected and designed using Primer Express 2.0 software (Applied Biosystems, Foster City, CA). Genes were quantified by real-time PCR to validate the microarray data, and a subset of 13 genes were further selected by their robust behaviour in the validation assays, allowing to differentiate the toxic potential of PM from urban and rural sources.

Project description:The diverse climate types and the complex anthropogenic source emissions in China lead to the great regional differences of air pollution mechanisms. The COVID-19 lockdown has given us a precious opportunity to understand the effect of weather conditions and anthropogenic sources on the distribution of air pollutants in different climate zones. In this study, to understand the impact of meteorological and socio-economic factors on air pollution during COVID-19 lockdown, we divided 358 Chinese cities into eight climate regions. Temporal, spatial and diurnal variations of six major air pollutants from January 1 to April 18, 2020 were analyzed. The differences in the characteristics of air pollutants in different climate zones were obvious. PM2.5 reduced by 59.0%-64.2% in cold regions (North-East China (NEC) and North-Western (NW)), while O3 surged by 99.0%-99.9% in warm regions (Central South (CS) and Southern Coast (SC)). Diurnal variations of atmospheric pollutants were also more prominent in cold regions. Moreover, PM2.5, PM10, CO and SO2 showed more prominent reductions (20.5%-64.2%) in heating regions (NEC, NW, NCP and MG) than no-heating regions (0.8%-48%). Climate has less influence on NO2, which dropped by 41.2%-57.1% countrywide during the lockdown. The influences of weather conditions on the atmospheric pollutants in different climate zones were different. The wind speed was not the primary reason for the differences in air pollutants in different climate zones. Temperature, precipitation, and air pollution emissions led to prominent regional differences in air pollutants throughout the eight climates. The effect of temperature on PM, SO2, CO, and NO2 varied obviously with the latitude, at which condition temperature was negatively correlated to PM, SO2, CO, and NO2 in the north but positively in the south. The temperature was positively correlated to ozone in different climate zones, and the correlation was the highest in NEC and the lowest in SC. The rainfall has a strong removal effect on atmospheric pollutants in the climate regions with more precipitation, but it increases the pollutant concentrations in the climate regions with less precipitation. In regions with more emission sources, air pollutants experienced more significant variations and returned to pre-lockdown levels earlier.

Project description:Air quality in urban areas and megacities is dependent on emissions, physicochemical process and atmospheric conditions in a complex manner. The impact on air quality metrics of the COVID-19 lockdown measures was evaluated during two periods in Athens, Greece. The first period involved stoppage of educational and recreational activities and the second severe restrictions to all but necessary transport and workplace activities. Fresh traffic emissions and their aerosol products in terms of ultrafine nuclei particles and nitrates showed the most significant reduction especially during the 2nd period (40-50%). Carbonaceous aerosol both from fossil fuel emissions and biomass burning, as well as aging ultrafine and accumulation mode particles showed an increase of 10-20% of average before showing a decline (5 to 30%). It is found that removal of small nuclei and Aitken modes increased growth rates and migration of condensable species to larger particles maintaining aerosol volume.

Project description:Most of the state governments in United States (U.S.) issued lockdown or business restrictions amid the COVID-19 pandemic in March 2020, which created a unique opportunity to evaluate the air quality response to reduced economic activities. Data acquired from 28 long-term air quality stations across the U.S. revealed widespread but nonuniform reductions of nitrogen dioxide (NO2) and carbon monoxide (CO) during the first phase of lockdown (March 15-April 25, 2020) relative to a pre-lockdown reference period and historical baselines established in 2017-2019. The reductions, up to 49% for NO2 and 37% for CO, are statistically significant at two thirds of the sites and tend to increase with local population density. Significant reductions of particulate matter (PM2.5 and PM10) only occurred in the Northeast and California/Nevada metropolises where NO2 declined the most, while the changes in ozone (O3) were mixed and relatively minor. These findings are consistent with lower transportation and utility demands that dominate NO2 and CO emissions, especially in major urban areas, due to the lockdown. This study provides an insight into potential public health benefits with more aggressive air quality management, which should be factored into strategies to reopen the U.S. and global economy.

Project description:In 2020, Covid-19-related mobility restrictions resulted in the most extensive human-made air-quality changes ever recorded. The changes in mobility are quantified in terms of outdoor air pollution (concentrations of PM2.5 and NO2) and the associated health impacts in four UK cities (Greater London, Cardiff, Edinburgh and Belfast). After applying a weather-corrected machine learning (ML) technique, all four cities show NO2 and PM2.5 concentration anomalies in 2020 when compared with the ML-predicted values for that year. The NO2 anomalies are -21% for Greater London, -19% for Cardiff, -27% for Belfast and -41% for Edinburgh. The PM2.5 anomalies are 7% for Greater London, -1% for Cardiff, -15% for Edinburgh, -14% for Belfast. All the negative anomalies, which indicate air pollution at a lower level than expected from the weather conditions, are attributable to the mobility restrictions imposed by the Covid-19 lockdowns. Spearman rank-order correlations show a significant correlation between the lowering of NO2 levels and reduction in public transport (p < 0.05) and driving (p < 0.05), which is associated with a decline in NO2-attributable mortality. These positive effects of the mobility restrictions on public health can be used to evaluate policies for improved outdoor air quality.Policy relevanceFinding the means to curb air pollution is very important for public health. Empirical evidence at a city scale reveals significant correlations between the reduction in vehicular transport and in ambient NO2 concentrations. The results provide justification for city-level initiatives to reduce vehicular traffic. Well-designed and effective policy interventions (e.g. the promotion of walking and cycling, remote working, local availability of services) can substantially reduce long-term air pollution and have positive health impacts.

Project description:Indoor VOC air concentrations of many compounds are higher than outdoor concentrations due to indoor sources. However, most studies have measured residential indoor air in urban centers so the typical indoor air levels in suburban and rural regions have not been well characterized. Indoor VOC air concentrations were measured in 100 homes in suburban and rural areas in NJ to provide background levels for investigations of the impact from subsurface contamination sources. Of the 57 target compounds, 23 were not detected in any of the homes, and 14 compounds were detected in at least 50% of the homes with detection limits of approximately 1 microg/m3. The common compounds identified included aromatic and aliphatic hydrocarbons from mobile sources, halogenated hydrocarbons commonly used in consumer products or from chlorinated drinking water, acetone and 2-butanone emitted from cosmetic products, and Freons. Typical concentrations were in the low microg/m3 range, though values of tens, hundreds or even thousands of microg/m3 were measured in individual homes in which activities related to specific sources of VOCs were reported. Compounds with known similar sources were highly correlated. The levels observed are consistent with concentrations found in the air of urban homes.

Project description:The outbreak of coronavirus named COVID-19, initially identified in Wuhan, China in December 2019, has spread rapidly at the global scale. Most countries have rapidly stopped almost all activities including industry, services and transportation of goods and people, thus decreasing air pollution in an unprecedented way, and providing a unique opportunity to study air pollutants. While satellite data have provided visual evidence for the global reduction in air pollution such as nitrogen dioxide (NO2) worldwide, precise and quantitative information is missing at the local scale. Here we studied changes in particulate matter (PM2.5, PM10), carbon monoxide (CO), NO2, sulfur dioxide (SO2) and ozone (O3) at 10 urban sites in Hangzhou, a city of 7.03 million inhabitants, and at 1 rural site, before city lockdown, January 1-23, during city lockdown, January 24-February 15, and during resumption, February 16-28, in 2020. Results show that city lockdown induced a sharp decrease in PM2.5, PM10, CO, and NO2 concentrations at both urban and rural sites. The NO2 decrease is explained by reduction in traffic emissions in the urban areas, and by lower regional transport in rural areas during lockdown, as expected. SO2 concentrations decreased from 6.3 to 5.3 μg m-3 in the city, but increased surprisingly from 4.7 to 5.8 μg m-3 at the rural site: this increase is attributed both to higher coal consumption for heating and emissions from traditional fireworks of the Spring Eve and Lantern Festivals during lockdown. Unexpectedly, O3 concentrations increased by 145% from 24.6 to 60.6 μg m-3 in the urban area, and from 42.0 to 62.9 μg m-3 in the rural area during the lockdown. This finding is explained by the weakening of chemical titration of O3 by NO due to reductions of NOx fresh emissions during the non-photochemical reaction period from 20:00 PM to 9:00 AM (local time). During the lockdown, compared to the same period in 2019, the daily average concentrations in the city decreased by 42.7% for PM2.5, 47.9% for PM10, 28.6% for SO2, 22.3% for CO and 58.4% for NO2, which is obviously explained by the absence of city activities. Overall, we observed not only the expected reduction in some atmospheric pollutants (PM, SO2, CO, NO2), but also unexpected increases in SO2 in the rural areas and of ozone (O3) in both urban and rural areas, the latter being paradoxically due to the reduction in nitrogen oxide levels. In other words, the city lockdown has improved air quality by reducing PM2.5, PM10, CO, and NO2, but has also decreased air quality by augmenting O3 and SO2.

Project description:We have investigated the impact of reduced emissions due to COVID-19 lockdown measures in spring 2020 on air quality in Canada's four largest cities: Toronto, Montreal, Vancouver, and Calgary. Observed daily concentrations of NO2, PM2.5, and O3 during a "pre-lockdown" period (15 February-14 March 2020) and a "lockdown" period (22 March-2 May 2020), when lockdown measures were in full force everywhere in Canada, were compared to the same periods in the previous decade (2010-2019). Higher-than-usual seasonal declines in mean daily NO2 were observed for the pre-lockdown to lockdown periods in 2020. For PM2.5, Montreal was the only city with a higher-than-usual seasonal decline, whereas for O3 all four cities remained within the previous decadal range. In order to isolate the impact of lockdown-related emission changes from other factors such as seasonal changes in meteorology and emissions and meteorological variability, two emission scenarios were performed with the GEM-MACH air quality model. The first was a Business-As-Usual (BAU) scenario with baseline emissions and the second was a more realistic simulation with estimated COVID-19 lockdown emissions. NO2 surface concentrations for the COVID-19 emission scenario decreased by 31 to 34% on average relative to the BAU scenario in the four metropolitan areas. Lower decreases ranging from 6 to 17% were predicted for PM2.5. O3 surface concentrations, on the other hand, showed increases up to a maximum of 21% close to city centers versus slight decreases over the suburbs, but Ox (odd oxygen), like NO2 and PM2.5, decreased as expected over these cities.Supplementary informationThe online version contains supplementary material available at 10.1007/s11869-021-01039-1.

Project description:This work aims to quantify potential pollution level changes in an urban environment (Madrid city, Spain) located in South Europe due to the lockdown measures for preventing the SARS-CoV-2 transmission. Polluting 11 species commonly monitored in urban zones were attended. Except for O3, a prompt target pollutant levels abatement was reached, intensely when implanted stricter measures and moderately along those measures' relaxing period. In the case of TH and CH4, it is evidenced a progressive diminution over the lockdown period. While the highest decreasing average changes relapsed on NOx (NO2: - 40.0% and NO: - 33.3%) and VOCs (C7H8: - 36.3% and C6H6: - 32.8%), followed by SO2 (- 27.0%), PM10 (- 19.7%), CO (- 16.6%), CH4 (- 14.7%), TH (- 11.6%) and PM2.5 (- 10.1%), the O3 level slightly raised 0.4%. These changes were consistently dependent on the measurement station location, emphasizing urban background zones for SO2, CO, C6H6, C7H8, TH and CH4, suburban zones for PM2.5 and O3, urban traffic sites for NO and PM10, and keeping variations reasonably similar at all the stations in the case of NO2. Those pollution changes were not translated in variations on geospatial pattern, except for NO, O3 and SO2. Although the researched urban atmosphere improvement was not attributable to meteorological conditions' variations, it was in line with the decline in traffic intensity. The evidenced outcomes might offer valuable clues to air quality managers in urban environments regarding decision-making in favor of applying punctual severe measures for quickly and considerably relieving polluting high load occurred in urban environments.Supplementary informationThe online version contains supplementary material available at 10.1007/s13762-022-04464-6.
