Project description:The review aimed to collect information about the wearable wireless sensor system (WWSS) for cattle and to conduct a systematic literature review on the accuracy of predicting the physiological parameters of these systems. The WWSS was categorized as an ear tag, halter, neck collar, rumen bolus, leg tag, tail-mounted, and vaginal mounted types. Information was collected from a web-based search on Google, then manually curated. We found about 60 WWSSs available in the market; most sensors included an accelerometer. The literature evaluating the WWSS performance was collected through a keyword search in Scopus. Among the 1875 articles identified, 46 documents that met our criteria were selected for further meta-analysis. Meta-analysis was conducted on the performance values (e.g., correlation, sensitivity, and specificity) for physiological parameters (e.g., feeding, activity, and rumen conditions). The WWSS showed high performance in most parameters, although some parameters (e.g., drinking time) need to be improved, and considerable heterogeneity of performance levels was observed under various conditions (average I2 = 76%). Nevertheless, some of the literature provided insufficient information on evaluation criteria, including experimental conditions and gold standards, to confirm the reliability of the reported performance. Therefore, guidelines for the evaluation criteria for studies evaluating WWSS performance should be drawn up.

Project description:Wearable non-invasive biosensors for the continuous monitoring of metabolites in sweat can detect a few analytes at sufficiently high concentrations, typically during vigorous exercise so as to generate sufficient quantity of the biofluid. Here we report the design and performance of a wearable electrochemical biosensor for the continuous analysis, in sweat during physical exercise and at rest, of trace levels of multiple metabolites and nutrients, including all essential amino acids and vitamins. The biosensor consists of graphene electrodes that can be repeatedly regenerated in situ, functionalized with metabolite-specific antibody-like molecularly imprinted polymers and redox-active reporter nanoparticles, and integrated with modules for iontophoresis-based sweat induction, microfluidic sweat sampling, signal processing and calibration, and wireless communication. In volunteers, the biosensor enabled the real-time monitoring of the intake of amino acids and their levels during physical exercise, as well as the assessment of the risk of metabolic syndrome (by correlating amino acid levels in serum and sweat). The monitoring of metabolites for the early identification of abnormal health conditions could facilitate applications in precision nutrition.

Project description:Levodopa (L-Dopa) is considered to be one of the most effective therapies available for Parkinson's disease (PD) treatment. The therapeutic window of L-Dopa is narrow due to its short half-life, and long-time L-Dopa treatment will cause some side effects such as dyskinesias, psychosis, and orthostatic hypotension. Therefore, it is of great significance to monitor the dynamic concentration of L-Dopa for PD patients with wearable biosensors to reduce the risk of complications. However, the high concentration of interferents in the body brings great challenges to the in vivo monitoring of L-Dopa. To address this issue, we proposed a minimal-invasive L-Dopa biosensor based on a flexible differential microneedle array (FDMA). One working electrode responded to L-Dopa and interfering substances, while the other working electrode only responded to electroactive interferences. The differential current response of these two electrodes was related to the concentration of L-Dopa by eliminating the common mode interference. The differential structure provided the sensor with excellent anti-interference performance and improved the sensor's accuracy. This novel flexible microneedle sensor exhibited favorable analytical performance of a wide linear dynamic range (0-20 μM), high sensitivity (12.618 nA μM-1 cm-2) as well as long-term stability (two weeks). Ultimately, the L-Dopa sensor displayed a fast response to in vivo L-Dopa dynamically with considerable anti-interference ability. All these attractive performances indicated the feasibility of this FDMA for minimal invasive and continuous monitoring of L-Dopa dynamic concentration for Parkinson's disease.

Project description:Most wearable biosensors struggle to balance flexibility and conductivity in their sensing interfaces. In this study, we propose a wearable sensor featuring a highly stretchable, three-dimensional conductive network structure based on liquid metal. The sensor interface utilizes a patterned Ga@MXene hydrogel system, where gallium (Ga) grafted onto MXene provides enhanced electrical conductivity and malleability. MXene provides excellent conductivity and a three-dimensional layered structure. Additionally, the chitosan (CS) hydrogel, with its superior water absorption and stretchability, allows the electrode to retain sweat and closely stick to the skin. The sensor demonstrates a low limit of detection (0.77 μM), high sensitivity (1.122 μA⋅μM⁻1⋅cm⁻2), and a broad detection range (10-1,000 μM), meeting the requirements for a wide range of applications. Notably, the sensor can also induce perspiration in the wearer. The three-dimensional porous structure of the Ga@MXene/CS biosensor ensures excellent conductivity and flexibility, making it suitable for a variety of applications.

Project description:Chronic diseases call for routine management of frequent monitoring of specific biomarkers. Traditional in vitro diagnostics technologies suffer from complex sampling processes and long detection intervals, which cannot meet the need of continuous monitoring. Wearable devices taking advantage of compact size, rapid detection process, and small sample consumption are promising to take the place of endpoint detection, providing more comprehensive information about human health. Here, we proposed a fully integrated wearable system with an ultrasensitive 3D-structured biosensor for real-time monitoring of multiple metabolites. The 3D-structured biosensor shows wide linear ranges of 400-1,400 μM and 0.1-8 mM and high sensitivities of 460.5 and 283.09 μA/(mM·cm2) for lactate and glucose detection, respectively. We have conducted in vivo animal experiments, and the proposed wearable biosensor demonstrated high consistency with established methods. We envision that this system could provide a real-time wearable detection platform for multiple biomarker detection.

Project description:The development of wearable biosensors for continuous noninvasive monitoring of target biomarkers is limited to assays of a single sampled biofluid. An example of simultaneous noninvasive sampling and analysis of two different biofluids using a single wearable epidermal platform is demonstrated here. The concept is successfully realized through sweat stimulation (via transdermal pilocarpine delivery) at an anode, alongside extraction of interstitial fluid (ISF) at a cathode. The system thus allows on-demand, controlled sampling of the two epidermal biofluids at the same time, at two physically separate locations (on the same flexible platform) containing different electrochemical biosensors for monitoring the corresponding biomarkers. Such a dual biofluid sampling and analysis concept is implemented using a cost-effective screen-printing technique with body-compliant temporary tattoo materials and conformal wireless readout circuits to enable real-time measurement of biomarkers in the sampled epidermal biofluids. The performance of the developed wearable device is demonstrated by measuring sweat-alcohol and ISF-glucose in human subjects consuming food and alcoholic drinks. The different compositions of sweat and ISF with good correlations of their chemical constituents to their blood levels make the developed platform extremely attractive for enhancing the power and scope of next-generation noninvasive epidermal biosensing systems.

Project description:Personalized monitoring of female hormones (for example, oestradiol) is of great interest in fertility and women's health. However, existing approaches usually require invasive blood draws and/or bulky analytical laboratory equipment, making them hard to implement at home. Here we report a skin-interfaced wearable aptamer nanobiosensor based on target-induced strand displacement for automatic and non-invasive monitoring of oestradiol via in situ sweat analysis. The reagentless, amplification-free and 'signal-on' detection approach coupled with a gold nanoparticle-MXene-based detection electrode offers extraordinary sensitivity with an ultra-low limit of detection of 0.14 pM. This fully integrated system is capable of autonomous sweat induction at rest via iontophoresis, precise microfluidic sweat sampling controlled via capillary bursting valves, real-time oestradiol analysis and calibration with simultaneously collected multivariate information (that is, temperature, pH and ionic strength), as well as signal processing and wireless communication with a user interface (for example, smartphone). We validated the technology in human participants. Our data indicate a cyclical fluctuation in sweat oestradiol during menstrual cycles, and a high correlation between sweat and blood oestradiol was identified. Our study opens up the potential for wearable sensors for non-invasive, personalized reproductive hormone monitoring.

Project description:Transdermal alcohol biosensors have the ability to detect the alcohol that emanates from the bloodstream and diffuses through the skin. However, previous biosensors have suffered from long-term fouling of the sensor element and drift in the resulting sensor readings over time. Here, we report a wearable alcohol sensor platform that solves the problem of sensor fouling by enabling drift-free signals in vivo for up to 24 h and an interchangeable cartridge connection that enables consecutive days of measurement. We demonstrate how alcohol oxidase enzyme and Prussian Blue can be combined to prevent baseline drift above 25 nA, enabling sensitive detection of transdermal alcohol. Laboratory characterization of the enzymatic alcohol sensor demonstrates that the sensor is mass-transfer-limited by a diffusion-limiting membrane of lower permeability than human skin and a linear sensor range between 0 mM and 50 mM. Further, we show continuous transdermal alcohol data recorded with a human subject for two consecutive days. The non-invasive sensor presented here is an objective alternative to the self-reports used commonly to quantify alcohol consumption in research studies.

Project description:Accurate continuous direct measurement of the blood pressure is currently available thru direct invasive methods via intravascular needles, and is mostly limited to use during surgical procedures or in the intensive care unit (ICU). Non-invasive methods that are mostly based on auscultation or cuff oscillometric principles do provide relatively accurate measurement of blood pressure. However, they mostly involve physical inconveniences such as pressure or stress on the human body. Here, we introduce a new non-invasive mechanism of tissue-informative measurement, where an experimental phenomenon called subcutaneous tissue pressure equilibrium is revealed and related for application in detection of absolute blood pressure. A prototype was experimentally verified to provide an absolute blood pressure measurement by wearing a watch-type measurement module that does not cause any discomfort. This work is supposed to contribute remarkably to the advancement of continuous non-invasive mobile devices for 24-7 daily-life ambulatory blood-pressure monitoring.

Project description:BackgroundMillions of catheters for invasive arterial pressure monitoring are placed annually in intensive care units, emergency rooms, and operating rooms to guide medical treatment decision-making. Accurate assessment of arterial blood pressure requires an IV pole-attached pressure transducer placed at the same height as a reference point on the patient's body, typically, the heart. Every time a patient moves, or the bed is adjusted, a nurse or physician must adjust the height of the pressure transducer. There are no alarms to indicate a discrepancy between the patient and transducer height, leading to inaccurate blood pressure measurements.MethodsWe present a low-power wireless wearable tracking device that uses inaudible acoustic signals emitted from a speaker array to automatically compute height changes and correct the mean arterial blood pressure. Performance of this device was tested in 26 patients with arterial lines in place.ResultsOur system calculates the mean arterial pressure with a bias of 0.19, inter-class correlation coefficients of 0.959 and a median difference of 1.6 mmHg when compared to clinical invasive arterial measurements.ConclusionsGiven the increased workload demands on nurses and physicians, our proof-of concept technology may improve accuracy of pressure measurements and reduce the task burden for medical staff by automating a task that previously required manual manipulation and close patient surveillance.
