Clistobothrium sp. (Cestoda: Tetraphyllidea) in oarfish (Regalecus russelii) stranded on the coast of Akita Prefecture, Japan.

ABSTRACT: Oarfish (Regalecus russelii Cuvier) are mesopelagic fish with little known about their life history. Oarfish live in deep water, making it difficult for researchers to collect specimens; thus, records of their parasitic helminths are limited. Two plerocercoids were found for the first time in an oarfish stranded on the coast of Akita Prefecture, Japan. These plerocercoids were identified as Clistobothrium sp. RR-1 using morphological and molecular analyses. It was revealed that oarfish represent one of the intermediate hosts of the genus Clistobothrium, and large sharks are the definitive hosts for these parasites.

Brucella infection in rough-toothed dolphin (Steno bredanensis) with severe orchitis stranded on the Pacific coast of Japan.

Project description:In the western North Pacific, prominent granulomatous testes have been detected in many Brucella-infected common minke whales (Balaenoptera acutorostrata), but there have been no reports in toothed cetaceans. We found severe orchitis with granulomatous lesions in a rough-toothed dolphin (Steno bredanensis) stranded on the Pacific coast of Japan in 2011. Histopathological examination revealed leukocyte infiltration of the lesions. DNA from the lesion was analyzed by PCR and it showed molecular biological similarities with those of Brucella-infected common minke whales and Brucella ceti of sequence-type 27 (ST27). These results suggest that the type of Brucella ceti that infected the dolphin was ST27, which may have caused severe orchitis. This study adds to our understanding of Brucella infections in marine mammals.

Seroepidemiology of human bocavirus in Hokkaido prefecture, Japan.

Project description:A new human virus, provisionally named human bocavirus (HBoV), was discovered by Swedish researchers in 2005. A new immunofluorescence assay using Trichoplusia ni insect cells infected with a recombinant baculovirus expressing the VP1 protein of HBoV was developed, and the levels of immunoglobulin G antibody to the VP1 protein of HBoV in serum samples were measured. The overall seroprevalence rate of antibodies against the VP1 protein of HBoV in a Japanese population aged from 0 months to 41 years was 71.1% (145 of 204). The seropositive rate was lowest in the age group of 6 to 8 months and gradually increased with age. All of the children had been exposed to HBoV by the age of 6 years. A rise in titers of antibody against the VP1 protein of HBoV during the convalescent phase was observed for four patients with lower respiratory tract infections, and HBoV DNA was detected in nasopharyngeal swab and serum samples from all four patients. These results suggest that HBoV is a ubiquitous virus acquired early in life and that HBoV might play a role in the course of lower respiratory tract infections.

Porcine circoviruses in wild boars in Nagano Prefecture, Japan.

Project description:We investigated the prevalence of porcine circovirus 2 (PCV2) and 3 (PCV3) in wild boars captured or found dead in Nagano Prefecture in 2020. Based on PCR testing, 21 of 254 (8.3%) wild boars were PCV2-positive and 43 of 256 (16.8%) wild boars were PCV3-positive, 5 of 253 (2.0%) wild boars were both PCV2-positive and PCV3-positive. The frequency of detecting PCV3 in wild boars was significantly higher in adults than in juveniles (P=0.014). The PCV2-positive wild boars were found in all districts except for the North Alps and Hokushin, while PCV3-positive wild boars were found in all districts except for Suwa. This is the first report of PCV2 and PCV3 detected in wild boars in Japan.

Description and life-cycle of Taenia lynciscapreoli sp. n. (Cestoda, Cyclophyllidea).

Project description:A new species of tapeworm, Taenia lynciscapreoli sp. n. (Cestoda, Cyclophyllidea), is described from the Eurasian lynx (Lynx lynx), the main definitive host, and the roe deer (Capreolus capreolus and Capreolus pygargus), the main intermediate hosts, from Finland and Russia (Siberia and the Russian Far East). The new species was found once also in the wolf (Canis lupus) and the Eurasian elk/moose (Alces alces), representing accidental definitive and intermediate hosts, respectively. The conspecificity of adult specimens and metacestodes of Taenia lynciscapreoli sp. n. in various host species and regions, and their distinction from related species of Taenia, was confirmed by partial nucleotide sequences of the mitochondrial cytochrome c oxidase subunit 1 gene. Morphologically, Taenia lynciscapreoli sp. n. can be separated unambiguously from all other species of Taenia by the shape of its large rostellar hooks, particularly the characteristically short, wide and strongly curved blade. If the large rostellar hooks are missing, Taenia lynciscapreoli may be separated from related species by a combination of morphological features of mature proglottids. It is suggested that Taenia lynciscapreoli has been present in published materials concerning the tapeworms of Lynx lynx and Lynx pardinus in Europe, but has been misidentified as Taenia pisiformis (Bloch, 1780). Taenia lynciscapreoli sp. n. has not been found in lynx outside the range of roe deer, suggesting a transmission pathway based on a specific predator-prey relationship. The present study applies a novel, simple approach to compare qualitative interspecific differences in the shape of rostellar hooks.

Lacydoniidae (Annelida) Off the Coast of North-eastern Japan: A Description of <i>Lacydonia japonica</i> sp. nov.

Project description:Lacydonia japonica sp. nov. (Annelida, Lacydoniidae) is described based on material found in sediments collected off the Pacific coast of northern Honshu, Japan, at depths of 262 m and 407 m. The sediments were obtained by a remotely operated vehicle equipped with a suction sampler during a Tohoku Ecosystem-Associated Marine Sciences (TEAMS) project in 2019. Lacydonia japonica sp. nov. belongs to the eyeless group of lacydoniids and is discriminated from the morphologically most similar congener, Lacydonia papillata Uschakov, 1958 by its reddish pigments on both the dorsal and ventral parapodial cirri and four pigment spots on the pygidium. To assess the phylogenetic position of the new species among other lacydoniids for which sequence data are available in public databases, analyses were performed using the mitochondrial cytochrome c oxidase subunit I (COI) and 16S rRNA, as well as the nuclear 18S rRNA and 28S rRNA genes. We additionally obtained some lacydoniids by sledging off western Japan, but these were severely fragmented and broken during collection. Using the paucity of morphological data, they were left unidentified as Lacydonia sp. but included in the molecular analyses. Genetic distances between Lacydonia eliasoni Hartmann-Schröder, 1996, Lacydonia japonica, and Lacydonia sp. off western Japan were 10.4-17.1% uncorrected p-distance (11.3-18.6% K2P) in terms of 658-bp COI sequences.

A Novel Archaeal Lineage in Boiling Hot Springs around Oyasukyo Gorge (Akita, Japan).

Project description:A novel deep-branching archaeal lineage was discovered at high-temperature hot springs around Oyasukyo Gorge in Akita Prefecture, Japan. Actively boiling hot spring water contained >1×104 microbes mL-1. The microbial community composition assessed by analyzing 16S rRNA gene amplicons revealed that the dominant bacterial phyla were Proteobacteria and Aquificae (>50% of the microbial composition) in samples collected in 2016 and 2019, respectively. Approximately 10% of the reads obtained in both years were not assigned to any taxonomy. The more detailed phylogenetic positions of the unassigned sequences identified using a clone library and phylogenetic tree showed that they formed a clade that was independent, distantly related to known phyla, and had low similarity (<82%) to all other sequences in available databases. The present results suggest that this novel archaeal phylum-level lineage thrives in boiling hot springs in Japan.
