Impacts of Rumen Degradable or Undegradable Protein Supplementation with or without Salt on Nutrient Digestion, and VFA Concentrations.

ABSTRACT: The objectives of this study were to evaluate the effect of differences in protein type and delivery method on rumen dynamics and nutrient digestion. Cows were allotted to rumen degradable protein (RDP) or rumen undegradable protein (RUP) and self-fed (SF) salt-limited pressed blocks or hand-fed (HF) loose supplement, resulting in four dietary treatments. There was a delivery effect (p = 0.04) on neutral detergent fiber (NDF) intake, as the SF animals consumed more NDF than HF animals. The RDP-SF animals had greater NDF digestibility (p = 0.04) and water intake (p = 0.03) than the three other treatments. Supplement intake displayed a protein type effect (p = 0.03), as RDP-supplemented animals consumed more supplement on a g·kg body weight (BW)-1 d-1 basis than RUP animals. There was an effect of protein type (p = 0.02) and delivery method (p = 0.03) on fluid flow rate, with RUP and HF cows having greater liquid flow rates. Ruminal pH was lower (p < 0.01) in RDP-HF cows than RDP-SF cows at all hours, except 4-h post-feeding. RDP-SF animals had the greatest (p < 0.01) concentrations of ruminal ammonia. Valerate ruminal concentrations were greater (p = 0.04) in RDP supplemented animals compared to RUP supplemented animals. In conclusion, self-fed supplements containing RDP may enhance the use of low-quality forages and increase ruminal ammonia concentrations.

Project description:This study was conducted to evaluate the effect of polyacrylamide (PAM) supplementation on the intake, digestion, weight gain, metabolism and growth of lambs. A total of ten 30 days old male small-tailed Han lambs with a body weight of 7.7±0.5 kg were divided into two equal groups (n = 5 each) and fed a basal diet or diet supplemented with 2.0 g of PAM per kg diet. The duration of the experiment was 210 days and experimental diets were fed ad libitum throughout the experimental period. Voluntary feed intake (VFI) was measured on daily basis, while body weight was measured on every ten days of the experiment.Two digestive and metabolic trials were conducted at the lamb's age of 95 to 103 days (Trial 1) and at the age of 210 to 218 days (Trial 2). At the end of experiment, all lambs were slaughtered to determine carcass characteristics. Results of the current study showed that supplementation of PAM in the diet of lambs increased the VFI and daily body gain by 14.4% (P < 0.05) and 15.2% (P < 0.01), respectively. In Trial 1, PAM supplementation in the diet increased the digestibility of dry matter (DM), organic matter (OM), crude protein (CP), cellulose, energy, and nitrogen retention by 7.9%, 5.4%, 6.4%, 9.6%, 4.3% and 30.3% (P < 0.01), respectively, and in Trial 2, PAM supplementation in the diet increased the digestibility of DM, OM, CP, cellulose, energy, and nitrogen retention by 9.3%, 7.9%, 7.7%, 11.6%, 6.9% and 38.5% (P < 0.01), respectively. Results of carcass parameter explored that supplementation of PAM in the diet increased the carcass, net meat and lean meat weights by 24.5%, 25.5%, and 30.6% (P < 0.01), respectively, however, PAM supplementation in the diet did not influence the contents of DM, OM, or CP in fresh liver, leg muscle, and rumen tissue; in addition, the CP contents in the Longissimus dorsi muscle was decreased by the supplementation of PAM in the diet. In summary, supplementation of 2.0 g of PAM per kg diet increased the VFI, nutrient digestibility, nitrogen retention, and carcass yield of lambs.

Project description:This study was conducted to investigate the effect of dietary multi-enzyme (MCPC) supplementation on synergistically enhancing the functions of both the foregut and hindgut, ultimately improving the nutrient digestion and utilization throughout the gastrointestinal tract. In vitro results demonstrated that MCPC increased the phosphorus and reducing sugar levels in the supernatant during enzymatic hydrolysis. Furthermore, during the fermentation of the enzymatic hydrolysis products, MCPC significantly increased the FRD0 value of the enzymatic hydrolysis products from both the positive control (PC) and negative control 1 (NC1) diets (p < 0.05). MCPC reduced the T1/2 value of in vitro fermentation products from the PC diet (p < 0.01), and decreased the VF (p = 0.082) and K (p < 0.05) values for the NC1 diet. Additionally, 72 crossbred barrows [Duroc × (Landrace × Yorkshire)], weighing 25 kg, were fed one of six diets until their live weight approached 50 kg. The basal diets consisted of PC, NC1 and negative control 2 (NC2), while the remaining three diets were prepared by adding 100 mg/kg MCPC to the respective basal diets. The results showed that MCPC supplementation significantly upregulated the expression of solute carrier family 17 member 4 (SLC17A4) and vitamin D receptor (VDR) genes in the duodenum (p < 0.05), while downregulating the expression of Calbindin-D28k (CaBP-D28K) and solute carrier family 1 member 4 (SLC1A4) genes (p < 0.05) in growing pigs. Moreover, MCPC supplementation significantly upregulated the expression of VDR, glucose transporter 2 (GLUT2) and intestinal fatty acid binding protein (FABP2) genes in the jejunum of growing pigs. Furthermore, MCPC supplementation significantly increased the relative abundances of Bacteroidota, Prevotella and Phascolarctobacterium (p < 0.05), while reducing the relative abundances of Verrucomicrobiota and Clostridium_sensu_stricto_1 (p < 0.05) in the colon of growing pigs. In conclusion, MCPC enhances nutrient digestion and absorption in the foregut, provides fermentable substrates for hindgut microbial fermentation, and improves gut microbiota composition. This improves hindgut fermentation and supports the synergistic interaction between the foregut and hindgut, ultimately improving nutrient utilization and benefiting animal health.
