Single-Shot Multicontrast X-ray Imaging for In Situ Visualization of Chemical Reaction Products.

ABSTRACT: We present the application of single-shot multicontrast X-ray imaging with an inverted Hartmann mask to the time-resolved in situ visualization of chemical reaction products. The real-time monitoring of an illustrative chemical reaction indicated the formation of the precipitate by the absorption, differential phase, and scattering contrast images obtained from a single projection. Through these contrast channels, the formation of the precipitate along the mixing line of the reagents, the border between the solid and the solution, and the presence of the scattering structures of 100-200 nm sizes were observed. The measurements were performed in a flexible and robust setup, which can be tailored to various imaging applications at different time scales.

In situ single-shot diffractive fluence mapping for X-ray free-electron laser pulses.

Project description:Free-electron lasers (FELs) in the extreme ultraviolet (XUV) and X-ray regime opened up the possibility for experiments at high power densities, in particular allowing for fluence-dependent absorption and scattering experiments to reveal non-linear light-matter interactions at ever shorter wavelengths. Findings of such non-linear effects are met with tremendous interest, but prove difficult to understand and model due to the inherent shot-to-shot fluctuations in photon intensity and the often structured, non-Gaussian spatial intensity profile of a focused FEL beam. Presently, the focused beam is characterized and optimized separately from the actual experiment. Here, we present the simultaneous measurement of XUV diffraction signals from solid samples in tandem with the corresponding single-shot spatial fluence distribution on the actual sample. Our in situ characterization scheme enables direct monitoring of the sample illumination, providing a basis to optimize and quantitatively understand FEL experiments.

Single-shot X-ray imaging of two-dimensional strain fields in colloidal crystals.

Project description:We used a soft X-ray free-electron laser and the Bragg coherent diffraction imaging method to characterize the defect structure of colloidal crystals. The single-shot X-ray pulse allowed us to reach four powder rings and measured all six reflections of the hexagonal lattice. We reproduced the static shape of the 2D crystal and mapped out the 2D strain tensors inside the crystal. The observed defect structures agreed with electron microscope images of similar colloidal samples.

Magnetic Resonance Imaging in Situ Visualization of an Electrochemical Reaction under Forced Hydrodynamic Conditions.

Project description:Magnetic resonance imaging (MRI) has proven to be a powerful tool for the characterization and investigation of in situ chemical reactions. This is more relevant when dealing with complex systems, where the spatial distribution of the species, partition equilibrium, flow patterns, among other factors have a determining effect over mass transport and therefore over the reaction rate. The advantage of MRI is that it provides spatial information in a noninvasive way and does not require any molecular sensor or sample extraction. In this work, MRI is used to fully characterize an electrochemical reaction under forced hydrodynamic conditions. Reaction rates, flow patterns, and quantitative concentration of the chemical species involved are spatially monitored in situ in a complex system that involves metallic pieces and a heterogeneous cementation reaction. Experimental data are compared with numerical simulations.

In-situ visualization of sound-induced otolith motion using hard X-ray phase contrast imaging.

Project description:Regarding the basics of ear structure-function relationships in fish, the actual motion of the solid otolith relative to the underlying sensory epithelium has rarely been investigated. Otolith motion has been characterized based on a few experimental studies and on approaches using mathematical modeling, which have yielded partially conflicting results. Those studies either predicted a simple back-and-forth motion of the otolith or a shape-dependent, more complex motion. Our study was designed to develop and test a new set-up to generate experimental data on fish otolith motion in-situ. Investigating the basic parameters of otolith motion requires an approach with high spatial and temporal resolution. We therefore used hard X-ray phase contrast imaging (XPCI). We compared two anatomically well-studied cichlid species, Steatocranus tinanti and Etroplus maculatus, which, among other features, differ in the 3D shape of their otoliths. In a water-filled tank, we presented a pure tone of 200 Hz to 1) isolated otoliths embedded in agarose serving as a simple model or 2) to a fish (otoliths in-situ). Our new set-up successfully visualized the motion of otoliths in-situ and therefore paves the way for future studies evaluating the principles of otolith motion.

Single-shot X-ray absorption spectroscopy at X-ray free electron lasers.

Project description:X-ray Absorption Spectroscopy (XAS) is a widely used X-ray diagnostic method for studying electronic and structural properties of matter. At first glance, the relatively narrow bandwidth and the highly fluctuating spectral structure of X-ray Free Electron Lasers (XFEL) sources seem to require accumulation over many shots to achieve high data quality. To date the best approach to implementing XAS at XFEL facilities has been using monochromators to scan the photon energy across the desired spectral range. While this is possible for easily reproducible samples such as liquids, it is incompatible with many important systems. Here, we demonstrate collection of single-shot XAS spectra over 10s of eV using an XFEL source, with error bars of only a few percent. We additionally show how to extend this technique over wider spectral ranges towards Extended X-ray Absorption Fine Structure measurements, by concatenating a few tens of single-shot measurements. Our results pave the way for future XAS studies at XFELs, in particular those in the femtosecond regime. This advance is envisioned to be especially important for many transient processes that can only be initiated at lower repetition rates, for difficult to reproduce excitation conditions, or for rare samples, such as those encountered in high-energy density physics.

Multicontrast x-ray computed tomography imaging using Talbot-Lau interferometry without phase stepping.

Project description:The purpose of this work is to demonstrate that multicontrast computed tomography (CT) imaging can be performed using a Talbot-Lau interferometer without phase stepping, thus allowing for an acquisition scheme like that used for standard absorption CT.Rather than using phase stepping to extract refraction, small-angle scattering (SAS), and absorption signals, the two gratings of a Talbot-Lau interferometer were rotated slightly to generate a moiré pattern on the detector. A Fourier analysis of the moiré pattern was performed to obtain separate projection images of each of the three contrast signals, all from the same single-shot of x-ray exposure. After the signals were extracted from the detector data for all view angles, image reconstruction was performed to obtain absorption, refraction, and SAS CT images. A physical phantom was scanned to validate the proposed data acquisition method. The results were compared with a phantom scan using the standard phase stepping approach.The reconstruction of each contrast mechanism produced the expected results. Signal levels and contrasts match those obtained using the phase stepping technique.Absorption, refraction, and SAS CT imaging can be achieved using the Talbot-Lau interferometer without the additional overhead of long scan time and phase stepping.

Single-shot time-folded fluorescence lifetime imaging.

Project description:Fluorescence lifetime imaging is an important tool in bioimaging that allows one to detect subtle changes in cell dynamics and their environment. Most time-domain approaches currently involve scanning a single illumination point across the sample, which can make imaging dynamic scenes challenging, while single-shot "rapid lifetime determination" can suffer from large uncertainties when the lifetime is not appropriately sampled. Here, we propose a time-folded fluorescence lifetime imaging microscopy (TFFLIM) approach, whereby a time-folding cavity provides multiple spatially sheared replicas of the lifetime, each shifted temporally with respect to a fixed time gate. This provides a robust, single-shot FLIM approach that we experimentally validate across a broad lifetime range on fluorescent beads and Convallaria samples.

Laser-driven x-ray and proton micro-source and application to simultaneous single-shot bi-modal radiographic imaging.

Project description:Radiographic imaging with x-rays and protons is an omnipresent tool in basic research and applications in industry, material science and medical diagnostics. The information contained in both modalities can often be valuable in principle, but difficult to access simultaneously. Laser-driven solid-density plasma-sources deliver both kinds of radiation, but mostly single modalities have been explored for applications. Their potential for bi-modal radiographic imaging has never been fully realized, due to problems in generating appropriate sources and separating image modalities. Here, we report on the generation of proton and x-ray micro-sources in laser-plasma interactions of the focused Texas Petawatt laser with solid-density, micrometer-sized tungsten needles. We apply them for bi-modal radiographic imaging of biological and technological objects in a single laser shot. Thereby, advantages of laser-driven sources could be enriched beyond their small footprint by embracing their additional unique properties, including the spectral bandwidth, small source size and multi-mode emission.

Visualization of Electron Density Changes Along Chemical Reaction Pathways.

Project description:We propose a simple procedure for visualizing the electron density changes (EDC) during a chemical reaction, which is based on a mapping of rectangular grid points for a stationary structure into (distorted) positions around atoms of another stationary structure. Specifically, during a small step along the minimum energy pathway (MEP), the displacement of each grid point is obtained as a linear combination of the motion of all atoms, with the contribution from each atom scaled by the corresponding Hirshfeld weight. For several reactions (identity SN2, Claisen rearrangement, Diels-Alder reaction, [3+2] cycloaddition, and phenylethyl mercaptan attack on pericosine A), our EDC plots showed an expected reduction of electron densities around severed bonds (or those with the bond-order lowered), with the opposite observed for newly-formed or enhanced chemical bonds. The EDC plots were also shown for copper triflate catalyzed N2O fragmentation, where the N-O bond weakening initially occurred on a singlet surface, but continued on a triplet surface after reaching the minimum-energy crossing point (MECP) between the two potential energy surfaces.

Single shot multispectral multidimensional imaging using chaotic waves.

Project description:Multispectral imaging technology is a valuable scientific tool for various applications in astronomy, remote sensing, molecular fingerprinting, and fluorescence imaging. In this study, we demonstrate a single camera shot, lensless, interferenceless, motionless, non-scanning, space, spectrum, and time resolved five-dimensional incoherent imaging technique using tailored chaotic waves with quasi-random intensity and phase distributions. Chaotic waves can distinctly encode spatial and spectral information of an object in single self-interference intensity distribution. In this study, a tailored chaotic wave with a nearly pure phase function and lowest correlation noise is generated using a quasi-random array of pinholes. A unique sequence of signal processing techniques is applied to extract all possible spatial and spectral channels with the least entropy. The depth-wavelength reciprocity is exploited to see colour from depth and depth from colour and the physics of beam propagation is exploited to see at one depth by calibrating at another.
