Phytochemical Composition, Anti-Inflammatory and ER Stress-Reducing Potential of Sambucus ebulus L. Fruit Extract.

ABSTRACT: Sambucus ebulus L. (SE) fruits are used for their immunostimulation, hematopoietic and antiviral potential. Recently, we focused on analyzing the mechanism underlying SE fruit aqueous extract's (FAE) immunomodulation and anti-inflammatory activities, with attention to its endoplasmic reticulum (ER) stress-reducing potential. J774A.1 macrophages were treated with SE FAE alone or in conditions of lipopolysaccharides (LPS) stimulation. Using GC-MS and LC-MS/MS, its phytochemical composition was analyzed. To measure transcription and protein levels, we used qPCR and Western blot, respectively. The prevailing phytochemicals in SE FAE were hydroxycinnamic acids, proanthocyanidins and anthocyanins. The content of some amino acids, organic acids, alcohols, fatty acids and esters were newly reported. Extracts exerted an immunostimulation potential by stimulating IL-6, TNFα, Ccl2, COX2 and iNOS transcription, without inducing ER stress. SE FAE suppressed the LPS-induced transcription of inflammation related genes (IL-1β, IL-6, TNFα, Ccl2, Icam-1, Fabp4, COX2, iNOS, Noxo1, IL-1ra, Sirt-1) and reduced the protein levels of iNOS, peIF2α, ATF6α and CHOP. The effects were comparable to that of salicylic acid. SE suppresses LPS-stimulated inflammatory markers on the transcription and translation levels. Targeting ER stress is possibly another mechanism underlying its anti-inflammatory potential. These findings reveal the potential of SE fruits as a beneficial therapeutic of inflammation and ER stress-related pathological conditions.

Project description:The excessive release of reactive oxygen species (ROS) can result in the development of chronic inflammation. The mechanisms involved in inflammation are various, with endoplasmic reticulum (ER) stress known to be among them. We have previously shown that black ginseng (BG) reduced lipid accumulation in and enhanced the antioxidant function of the liver in vitro and in vivo mostly due to ginsenoside Rb1, Rg3 and Rk1 components. Therefore, this study investigated the antioxidant effect of BG on the intestines and its possible mechanistic pathway through ER stress. The results showed that BG extract decreased ROS and nitric oxide (NO) production and reduced inducible nitric oxide synthase (iNOS) expression levels in vitro, and these results were confirmed by zebrafish embryos in vivo. However, this phenotype was abolished in the absence of inositol-requiring enzyme 1 (IRE1α) but not in the absence of protein kinase RNA (PKR)-like ER-resistant kinase (PERK) or X-box-binding protein 1 (XBP1) in the mouse embryo fibroblast (MEF) knockout (KO) cells, suggesting that BG elicits an antioxidant effect in an IRE1α-dependent manner. Antioxidant and anti-inflammatory effects were assessed in the liver and intestines of the mouse model affected by nonalcoholic fatty liver disease (NAFLD), which was induced by a high-fat/high-fructose diet. In the liver, BG treatment rescued NAFLD-induced glutathione (GSH), catalase (CAT), tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 expression. In the intestines, BG also rescued NAFLD-induced shortened villi, inflammatory immune cell infiltration, upregulated IL-6, cytosine-cytosine-adenosine-adenosine-thymidine (CCAAT)/enhancer-binding homologous protein (CHOP) and binding immunoglobulin protein (BiP) expression. In conclusion, our results show that BG reduces ROS and NO production followed by inflammation in an IRE1α-dependent and XBP1-independent manner. The results suggest that BG provides antioxidant and anti-inflammatory effects through an ER stress mechanism.

Project description:BackgroundThe antioxidant and anticancer potential of natural compounds, particularly from medicinal plants, is increasingly being explored as alternatives to synthetic antioxidants and chemotherapeutics. Boascia coriacea (Pax) has been traditionally used for treating various ailments, including oxidative stress-related diseases and prostate cancer. However, there is a paucity of empirical evidence to validate the ethnomedicinal claims, hence this study.MethodsThe antioxidant capacity of the extract was assessed using 1,1-Diphenyl-2-picryl Hydrazyl radical (DPPH) radical scavenging and hydrogen peroxide scavenging assays, alongside total antioxidant capacity. In vitro cytotoxicity was determined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on Vero CCL-81 normal cells and DU-145 prostate cancer cells. Gene expression levels of ar, bcl-2, caspase 3, cdk1, and p53 were quantified using qPCR to elucidate the mechanisms of action. Phytochemical analysis was conducted using gas chromatography-mass spectrometry.ResultsThe studied plant extract exhibited significant DPPH radical scavenging activity, with an EC50 of 0.008 μg/ml, 10-fold lower than that of L-ascorbic acid (0.08 μg/ml), indicating potent antioxidant capacity. Similarly, the extract demonstrated substantial hydrogen peroxide scavenging activity, albeit with lower efficacy (EC50 of 1039.10 μg/ml) compared to L-ascorbic acid, and a total antioxidant capacity of 454.39±25.26 μg AAE/mg dw. In vitro cytotoxicity assay revealed a CC50 of 68.61 μg/ml against Vero CCL-81 cells and an IC50 of 32.16 μg/ml against DU-145 cells, with a superior selectivity index of 2.13, compared to doxorubicin's 1.46. The extract significantly downregulated the expression of ar, bcl-2, normalised caspase 3, cdk1 genes while upregulating p53 in DU-145 cells, suggesting its role in inducing apoptosis and inhibiting cancer cell proliferation. Phytochemical analysis identified 19 compounds, including lup-20(29)-en-3-one (7.99%) and lupeol (59.49%), which are associated with anticancer activity.ConclusionThe ethyl acetate stem bark extract of B. coriacea demonstrates significant antioxidant and anticancer activities, potentially through modulation of apoptosis and cell cycle pathways. The presence of bioactive compounds supports its potential as a therapeutic agent, warranting further investigation for developing novel treatments for prostate cancer and oxidative stress-related conditions.

Project description:Rodents have remained a menace to humans, hence there is need to evaluate for anti-rodent activity of cheap and environment friendly control methods. This study aimed at evaluating the anti-rodenticidal activity of Thevetia (T.) peruviana fruit methanol extract. T. peruviana fruit was sampled, dried and extracted using methanol/water in the ratio of 3:1 by volume. Phytochemicals; alkaloids, phenols, flavonoids, glycosides, saponins, and tannins were determined qualitatively and quantitatively in the fruit extract. The extract was then characterized using Fourier Transform Infrared (FTIR) and Gas Chromatography Mass Spectrophotometer (GC-MS). Anti-rodent activity of the extracts was determined on a total of 25 mice with body weights of 20-25 g. The animals 8-12 weeks' old were grouped into 5 cages (5 animals per cage), marked and allowed to acclimatize with laboratory conditions of 25 °C, warm or less humid for 5 days with enough water and food. Extract dose (between 0.3 and 0.5 g of extract/kg body weight) was then administered in a single dose by gavage using intubation canula for 7 days and the animals observed for any toxicity and mortality. The data was subjected to probit analysis and ANOVA. Phytochemical screening showed that the extracts contained glycosides, phenols, saponins, alkaloids, triterpenoids, and flavonoids in different abundance. T. peruviana fruit contained 125.13 ± 1.04 mg/g in GAE phenolic content, 85.70 mg/g in RE of dry weight of flavonoids, 10.50 ± 0.01 mg/g in TAE of Tannins, 16.50± 0.21 mg/g alkaloid content, and 8.28± 0.11 mg/g saponin content. The FTIR spectrophotometer depicted O - H, CH2, C[bond, double bond]O, C-O-C functional groups in wave numbers of 3335, 2932, 1599, and 1001 cm-1 respectively. The T. peruviana fruit methanol extracts depicted high acute toxicity with an average of 300 mg/kg upon oral administration in Balb C mice species. The fruit extract from T. peruviana revealed presence of alkaloids, phenols, glycosides, saponins, tannins. These participated synergistically in killing the rats and the postmortem examination report indicated that the tested extract induced a number of physical changes in the mice and therefore the T. peruviana's fruit extract can be utilized as a natural alternative anti-rodent in agriculture production before and after harvesting.
