ABSTRACT: Adenia viridiflora Craib. is an indigenous plant found in Thailand, Cambodia and Vietnam that has become threatened owing to lack of knowledge about its agricultural management. This plant is now rare in the wild and was registered in the Plant Genetic Conservation Project under the initiation of Her Royal Highness Princess Maha Chakri Sirindhorn (RSPG) to promote sustainable conservation and optimally beneficial utilization. A. viridiflora has a long history of utilization as a nutrient-rich source with medicinal properties but scientific evidence of the veracity of these claims is limited. Here, the nutritional compositions, phenolic contents and antioxidant activities of different plant parts (young shoots and old leaves) of A. viridiflora were investigated using plants collected from four areas of Thailand as Kamphaeng Phet (KP), Muang Nakhon Ratchasima (MN), Pakchong Nakhon Ratchasima (PN) and Uthai Thani (UT) at different harvesting periods (March-April, May-June and July-August). Results indicated that young shoots provided higher energy, protein, fat, dietary fiber, phosphorus, sodium, and zinc than old leaves. By contrast, nutrients such as total sugar, vitamin C, carotenoids, potassium, calcium, magnesium, and iron contents were higher in old leaves that also exhibited higher phenolic contents and most antioxidant activities than young shoots. Generally, most nutrients, phenolic contents, and antioxidant activities exhibited no clear trend among different plant origins. The harvesting period of July-August provided a suitable climate for biosynthesis of most nutrients, while high phenolics were mainly found in samples harvested in March-April. No clear trend was observed in the prevalence of antioxidant activities that varied according to assay techniques.

Project description:Forage plants are valuable because they maintain wild and domesticated herbivores, and sustain the delivery of meat, milk and other commodities. Forage plants contain different quantities of fibre, lignin, minerals and protein, and vary in the proportion of their tissue that can be digested by herbivores. These nutritive components are important determinants of consumer growth rates, reproductive success and behaviour. A dataset was compiled to quantify variation in forage plant nutritive values within- and between-plant species, and to assess variation between plant functional groups and bioclimatic zones. 1255 geo-located records containing 3774 measurements of nutritive values for 136 forage plant species grown in 30 countries were obtained from published articles. Spatial variability in forage nutritive values indicated that climate modified plant nutritive values. Forage plants grown in arid and equatorial regions generally contained less digestible material than those grown in temperate and tundra regions; containing more fibre and lignin, and less protein. These patterns may reveal why herbivore body sizes, digestion and migration strategies are different in warmer and drier regions. This dataset also revealed the capacity for variation in the nutrition provided by forage plants, which may drive consumer species coexistence. The proportion of the plant tissue that was digestible ranged between species from 2 to 91%. The amount of fibre contained within plant material ranged by 23-90%, protein by 2-36%, lignin by 1-21% and minerals by 2-22%. On average, grasses and tree foliage contained the most fibre, whilst herbaceous legumes contained the most protein and tree foliage contained the most lignin. However, there were individual species within each functional group that were highly nutritious. This dataset may be used to identify forage plant species or mixtures of species from different functional groups with useful nutritional traits which can be cultivated to enhance livestock productivity and inform wild herbivore conservation strategies.

Project description:BackgroundSmall-grain winter cereals can be utilized as early spring pastures in temperate climates to relieve grazing pressure and potentially mitigate feed shortages. This study was conducted to determine the effects of triticale and oat cereal pastures grazed by sheep during early spring on forage yields, nutritive values, and nutritional requirements of sheep.MethodsThe research was carried out over three consecutive years, from 2015 to 2017, at the Sheep Research Institute in Bandırma-Balıkesir, located in the Marmara region of Türkiye. The treatments were arranged in a completely randomized block design, with the two forage species, triticale and oat, randomized within each of three blocks. The animal material for the study consisted of 24 Karacabey Merino sheep, each 2 years old, with an average live weight of 57.6 ± 0.5 kg, all in the late lactation stage. In each replication, four sheep were included, resulting in a total of 12 sheep grazing in each of the triticale and oat pastures. The sheep grazed exclusively on the cereal pastures without any additional feed, and had unrestricted access to water throughout the entire period of the experiment. The dry matter yields (DMY), dry matter intakes (DMI), nutritive values, and mineral contents of the cereal species were determined.ResultsThe DMY showed significant differences over the years (P < 0.05). No differences in DMY were observed between pastures, with oats yielding 11.99 t ha-1 and triticale yielding 11.08 t ha-1. During the grazing period, the change in DMY was significant in all years (P < 0.05). The average DMI of the sheep was 2,003.5 g d-1 for triticale and 2,156.6 g d-1 for oat, respectively, and DMI exhibited no significant differences across pastures. Although there was no difference in DMI between 2015 and 2016, the lowest consumption occurred in 2017 (P < 0.05). Additionally, while DMI showed different trends each year based on the periods, it generally decreased by the end of the grazing period. While both pastures provided similar nutritive values, significant differences were observed in the crude protein (CP), acid detergent fiber (ADF), in vitro true DM digestibility (DDM), and metabolisable energy (ME) values across the years. Over the years, as the grazing period progressed, CP levels decreased while neutral detergent fiber (NDF), ADF, and acid detergent lignin (ADL) increased, resulting in reduced DDM and ME values. The phosphorus (P) content in triticale was higher than in oats, but there were no differences in the content of other minerals between them. Between the years, significant differences were observed in the levels of phosphorus (P) and iron (Fe), while changes in other elements were insignificant. The variation in mineral content during the grazing process differed over the three years. Study results indicated that the nutritional values of triticale and oat pastures are similar, and both can effectively be used to provide sufficient feed to meet the early spring forage requirements for sheep.

Project description:BackgroundClinical cases of leishmaniasis caused by Leishmania (Mundinia) parasites have been increasingly reported in Southeast Asia, particularly Thailand. Recent evidence has shown that Leishmania (Mundinia) parasites successfully developed into infective metacyclic promastigotes in Culicoides biting midges, strongly supporting their putative role in disease transmission. However, Culicoides diversity, host preference, and Leishmania prevalence in endemic areas remain largely unknown.MethodsWe investigated the seasonal dynamics, infection prevalence, and blood meal identification of Culicoides collected from the emerging focus of visceral leishmaniasis in Lampang Province, Northern Thailand, during 2021-2023. Midge samples were molecularly screened for Leishmania using SSU rRNA-qPCR and ITS1-PCR, followed by Sanger plasmid sequencing, and parasite haplotype diversity was analyzed. Host blood meal origins were comparatively identified using host-specific Cytb-PCRs and a nanopore-based metabarcoding approach.ResultsA total of 501 parous and gravid females and 46 blood-engorged ones belonging to at least 17 species of five subgenera (Remmia, Trithecoides, Avaritia, Hoffmania, and Meijerehelea) and two species groups (Shortti and Calvipalpis) were collected with temporal differences in abundance. Leishmania was detected by SSU rRNA-qPCR in 31 samples of at least 11 midge species, consisting of Culicoides oxystoma, C. guttifer, C. orientalis, C. mahasarakhamense, C (Trithecoides) spp., C. innoxius, C. shortti, C. arakawae, C. sumatrae, C. actoni, and C. fulvus, with the overall infection prevalence of 5.7%. The latter six species represent the new records as putative leishmaniasis vectors in Northern Thailand. The ITS1-PCR and plasmid sequencing revealed that Leishmania martiniquensis was predominantly identified in all qPCR-positive species, whereas L. orientalis was identified only in three C. oxystoma samples. The most dominant haplotype of L. martiniquensis in Thailand was genetically intermixed with those from other geographical regions, confirming its globalization. Neutrality test statistics were also significantly negative on regional and country-wide scales, suggesting rapid population expansion or selective sweeps. Nanopore-based blood meal analysis revealed that most Culicoides species are mammalophilic, with peridomestic and wild mammals (cow, pig, deer, and goat-like species) and humans as hosts, while C. guttifer and C. mahasarakhamense fed preferentially on chickens.ConclusionsThis study revealed seasonal dynamics and sympatric circulation of L. martiniquensis and L. orientalis in different species of Culicoides. Evidence of human blood feeding was also demonstrated, implicating Culicoides as putative vectors of human leishmaniasis in endemic areas. Further research is therefore urgently needed to develop vector control strategies and assess the infection status of their reservoir hosts to effectively minimize disease transmission.
