New distribution records, first host plant record and DNA barcoding of the Neotropical plume moth Oidaematophoruspseudotrachyphloeus Gielis (Lepidoptera, Pterophoridae).

Background

Oidaematophoruspseudotrachyphloeus Gielis, 2011 (Lepidoptera, Pterophoridae) is a little-known Neotropical plume moth previously recorded in Ecuador, Peru and Argentina. Its host plants and DNA barcodes are unknown.

New information

Oidaematophoruspseudotrachyphloeus is recorded for the first time from Chile, based on six specimens from the Azapa Valley (Arica Province) and two from Guayacán (Coquimbo Province). Those from the Azapa Valley were reared from folivorous larvae collected on Ambrosiacumanensis Kunth (Asteraceae), representing the first host plant record for this plume moth. The first DNA barcode sequences of O.pseudotrachyphloeus are provided and used to explore relationships with congenerics.

A new distribution and host record for the rare moth, Callioratis millari (Lepidoptera: Geometridae), and some ecological observations.

Project description:Callioratis millari Hampson (Lepidoptera: Geometridae) is a Critically Endangered moth endemic to South Africa. Despite extensive searches, it was previously known only from the Entumeni Nature Reserve in KwaZulu-Natal, where its larvae exclusively feed on the cycad Stangeria eriopus (Kunze) Baill (Cycadales: Stangeriaceae). In July 2022, a new population of C. millari was discovered in the Kabouga section of Addo Elephant National Park in the Eastern Cape. Larvae of C. millari were feeding on the cycad Encephalartos caffer (Thunb.) Lehm (Cycadales: Zamiaceae), which also constitutes a new host record. In June 2023, we determined larval incidence and herbivory at this new locality, offering insights into the ecological requirements of C. millari. Known C. millari localities, although ecologically different, share low altitudes (700-950 m a.s.l.), moderate to high rainfall, and grassy habitats with sparse woody cover. A total of 59 larvae were counted in Kabouga, mostly in the fifth and sixth (final) instars. Herbivory incidence was lower on smaller plants and those covered by other vegetation. The flight period of adult C. millari likely occurs between mid-March and April in Kabouga, but further investigation is needed to clarify this. The peak period of larval occurrence in Kabouga occurs during the driest and coldest months of the year (May-July). Considering limited habitat availability, host plant poaching, and the risk of untimely fires, the species should be considered highly threatened. This study adds to our understanding of the biology of C. millari and provides information on its ecological requirements and may contribute to making informed management decisions.

New records of six moth (Lepidoptera: Erebidae, Lasiocampidae) species in south African countries, with comments on their distribution.

Project description:BackgroundSouthern Africa hosts a high diversity of ecosystems and habitats with a tremendous diversity of Lepidoptera. Although it is one of the most studied parts of the Afrotropics, the knowledge on diversity and distribution of south African moth fauna remains insufficient. To partly fill this gap, we surveyed macromoths by automatic light traps in five localities in two relatively less sampled south African countries.New informationWe reported six species and one genus (Remigioides) of moths which had not yet been recorded in Namibia or Zimbabwe. Although none of these records broadened the known distribution of individual species to a new biogeographical region, they still fill important gaps in their distributions. The known distributional ranges of two species have been substantially extended, although they are still within the same biogeographical regions: ca. 800 km southwards for Remigioides remigina (Mabille, 1884) and ca. 600 km westwards for Haplopacha cinerea Aurivillius, 1905.

Exotic-looking Neotropical Tischeriidae (Lepidoptera) and their host plants.

Project description:Seven new species of Tischeriidae are described from the Neotropics: Astrotischeria jociui Diškus & Stonis, sp. nov. (feeding on Wissadula excelsior (Cav.) C. Presl., Malvaceae), A. atlantica Diškus & Stonis, sp. nov. (feeding on Baccharis spicata (Lam.) Baill., Asteraceae), A. cornuata Diškus & Stonis, sp. nov. (host plant unknown), Paratischeria guarani Diškus & Stonis, sp. nov. (feeding on Elephantopus mollis Kunth, Asteraceae), P. mesoamericana Diškus & Stonis, sp. nov. (feeding on Montanoa hibiscifolia Benth., Asteraceae), P. suprafasciata Diškus & Stonis, sp. nov. (feeding on Allophyllus edulis (A. St.-Hil., A. Juss. & Cambess.) Hieron. ex Niederl., Sapindaceae), and P. braziliensis Diškus & Stonis, sp. nov. (host plant unknown). Additionally, an updated distribution map of Paratischeria neotropicana (Diškus & Stonis, 2015), which currently has the broadest distribution range among the Neotropical Tischeriidae is provided along with new host-plant data, a list of all recorded host plants in the Neotropics, and a brief discussion on trophic relationships of Tischeriidae. It is hypothesized that host-plant distribution ranges can provide clues to potential distribution ranges of these specialized, monophagous or oligophagous, leaf miners. All new taxa are illustrated with photographs of the adults, their genitalia, and, if available, leaf mines.

DNA barcoding insect-host plant associations.

Project description:Short-sequence fragments ('DNA barcodes') used widely for plant identification and inventorying remain to be applied to complex biological problems. Host-herbivore interactions are fundamental to coevolutionary relationships of a large proportion of species on the Earth, but their study is frequently hampered by limited or unreliable host records. Here we demonstrate that DNA barcodes can greatly improve this situation as they (i) provide a secure identification of host plant species and (ii) establish the authenticity of the trophic association. Host plants of leaf beetles (subfamily Chrysomelinae) from Australia were identified using the chloroplast trnL(UAA) intron as barcodes amplified from beetle DNA extracts. Sequence similarity and phylogenetic analyses provided precise identifications of each host species at tribal, generic and specific levels, depending on the available database coverage in various plant lineages. The 76 species of Chrysomelinae included-more than 10 per cent of the known Australian fauna-feed on 13 plant families, with preference for Australian radiations of Myrtaceae (eucalypts) and Fabaceae (acacias). Phylogenetic analysis of beetles shows general conservation of host association but with rare host shifts between distant plant lineages, including a few cases where barcodes supported two phylogenetically distant host plants. The study demonstrates that plant barcoding is already feasible with the current publicly available data. By sequencing plant barcodes directly from DNA extractions made from herbivorous beetles, strong physical evidence for the host association is provided. Thus, molecular identification using short DNA fragments brings together the detection of species and the analysis of their interactions.

Phylogeny and host-plant relationships of the Australian Myrtaceae leafmining moth genus Pectinivalva (Lepidoptera, Nepticulidae), with new subgenera and species.

Project description:The phylogeny of the mainly Australian nepticulid genus Pectinivalva Scoble, 1983 is investigated on the basis of morphology, and a division into three monophyletic subgenera is proposed on the basis of these results. These subgenera (Pectinivalva, Casanovula Hoare, subgen. n. and Menurella Hoare, subgen. n. ) are described and diagnosed, the described species of Pectinivalva are assigned to them, and representative new species are described in each: Pectinivalva (Pectinivalva) mystaconota Hoare, sp. n., Pectinivalva (Casanovula) brevipalpa Hoare, sp. n., Pectinivalva (Casanovula) minotaurus Hoare, sp. n., Pectinivalva (Menurella) scotodes Hoare, sp. n., Pectinivalva (Menurella) acmenae Hoare, sp. n., Pectinivalva (Menurella) xenadelpha Van Nieukerken & Hoare, sp. n., Pectinivalva (Menurella) quintiniae Hoare & Van Nieukerken, sp. n., and Pectinivalva (Menurella) tribulatrix Van Nieukerken & Hoare, sp. n. Pectinivalva (Menurella) quintiniae (from Quintinia verdonii, Paracryphiaceae) is the first known member of the genus with a host-plant not belonging to Myrtaceae. Pectinivalva (Menurella) xenadelpha from Mt Gunung Lumut, Kalimantan, Borneo, is the first pectinivalvine reported from outside Australia. Keys to the subgenera of Nepticulidae known from Australia, based on adults, male and female genitalia, and larvae, are presented. Host-plant relationships of Pectinivalva are discussed with relation to the phylogeny, and a list of known host-plants of Pectinivalva, including hosts of undescribed species, is presented. DNA barcodes are provided for most of the new and several unnamed species.

Patterns of genetic variation among geographic and host-plant associated populations of the peach fruit moth Carposina sasakii (Lepidoptera: Carposinidae).

Project description:BackgroundPopulations of herbivorous insects may become genetically differentiated because of local adaptation to different hosts and climates as well as historical processes, and further genetic divergence may occur following the development of reproductive isolation among populations. Here we investigate the population genetic structure of the orchard pest peach fruit moth (PFM) Carposina sasakii (Lepidoptera: Carposinidae) in China, which shows distinct biological differences when characterized from different host plants. Genetic diversity and genetic structure were assessed among populations from seven plant hosts and nine regions using 19 microsatellite loci and a mitochondrial sequence.ResultsStrong genetic differentiation was found among geographical populations representing distinct geographical regions, but not in host-associated populations collected from the same area. Mantel tests based on microsatellite loci indicated an association between genetic differentiation and geographical distance, and to a lesser extent environmental differentiation. Approximate Bayesian Computation analyses supported the scenario that PFM likely originated from a southern area and dispersed northwards before the last glacial maximum during the Quaternary.ConclusionsOur analyses suggested a strong impact of geographical barriers and historical events rather than host plants on the genetic structure of the PFM; however, uncharacterized environmental factors and host plants may also play a role. Studies on adaptive shifts in this moth should take into account geographical and historical factors.

Phylogeny, host use, and diversification in the moth family Momphidae (Lepidoptera: Gelechioidea).

Project description:Insect herbivores and their hostplants constitute much of Earth's described biological diversity, but how these often-specialized associations diversify is not fully understood. We combined detailed hostplant data and comparative phylogenetic analyses of the lepidopteran family Momphidae to explore how shifts in the use of hostplant resources, not just hostplant taxon, contribute to the diversification of a phytophagous insect lineage. We inferred two phylogenetic hypotheses emphasizing relationships among species in the nominate genus, Mompha Hübner. A six-gene phylogeny was constructed with reared exemplars and collections from hostplants in the family Onagraceae from western and southwestern USA, and a cytochrome c oxidase subunit 1 (COI) phylogeny was inferred from collections and publicly available accessions in the Barcode of Life Data System. Species delimitation analyses combined with morphological data revealed ca. 56 undescribed species-level taxa, many of which are hostplant specialists on Onagraceae in the southwestern USA. Our phylogenetic reconstructions divided Momphidae into six major clades: 1) an Onagraceae flower- and fruit-boring clade, 2) a Melastomataceae-galling clade, 3) a leafmining clade A, 4) a leafmining clade B, 5) a Zapyrastra Meyrick clade, and 6) a monobasic lineage represented by Mompha eloisella (Clemens). Ancestral trait reconstructions using the COI phylogeny identified leafmining on Onagraceae as the ancestral state for Momphidae. Our study finds that shifts along three hostplant resource axes (plant taxon, plant tissue type, and larval feeding mode) have contributed to the evolutionary success and diversification of momphids.

DNA barcoding of fogged caterpillars in Peru: A novel approach for unveiling host-plant relationships of tropical moths (Insecta, Lepidoptera).

Project description:The present study aimed to perform molecular identification of lepidopteran larvae from canopy fogging including gut-content analyses. A total of 130 lepidopteran larvae were selected from 37 fogging samples at the Panguana station, district Yuyapichis, province Puerto Inca, department Huánuco, Peru. Target trees were pre-identified and subsequently submitted to molecular confirmation of identity with three markers (rbcL, psbA and trnL-F). The COI gene of 119 lepidopteran larvae was successfully sequenced and found to belong to 92 species: Comparison of DNA barcodes with the reference database of adult moths resulted in 65 (55%) matches at species level, 32 (27%) at genus level, 19 (16%) at subfamily or family level, three just to order level. Three larvae could not be assigned to a family. For these larvae the fogged target tree now suggests a potential host-plant relationship. Molecular gut content analysis, based on High-Throughput-Sequencing was successfully tested for ten larvae corroborating feeding on the target plant in some cases but elucidating several other cases of potential 'alternative feeding'. We propose a larger-scale approach using this rapid and efficient method including molecular gut-content analyses for comprehensively testing the ratio of 'alternative feeders' and pitfalls caused by collateral fogging of larvae from neighboring trees.

Codling Moth (Lepidoptera: Tortricidae) Establishment in China: Stages of Invasion and Potential Future Distribution.

Project description:Codling moth (Cydia pomonella L.) is an internal feeding pest of apples and can cause substantial economic losses to fruit growers due to larval feeding which in turn degrades fruit quality and can result in complete crop loss if left uncontrolled. Although this pest originally developed in central Asia, it was not known to occur in China until 1953. For the first three decades the spread of codling moth within China was slow. Within the last three decades, addition of new commercial apple orchards and improved transportation, this pest has spread to over 131 counties in seven provinces in China. We developed regional (China) and global ecological niche models using MaxEnt to identify areas at highest potential risk of codling moth establishment and spread. Our objectives were to 1) predict the potential distribution of codling moth in China, 2) identify the important environmental factors associated with codling moth distribution in China, and 3) identify the different stages of invasion of codling moth in China. Human footprint, annual temperature range, precipitation of wettest quarter, and degree days ≥10 °C were the most important predictors associated with codling moth distribution. Our analysis identified areas where codling moth has the potential to establish, and mapped the different stages of invasion (i.e., potential for population stabilization, colonization, adaptation, and sink) of codling moth in China. Our results can be used in effective monitoring and management to stem the spread of codling moth in China.
