Putative Familial Transmissible Bacteria of Various Body Niches Link with Home Environment and Children's Immune Health.

ABSTRACT: Owing to their significant impact on children's long-term health, familial factors in the microbiomes of children have attracted increasing attention. However, the mechanism underlying microbiome transmission across generations remains unclear. A significantly lower alpha diversity was observed in the gut flora of children than in the gut flora of parents and grandparents; the alpha diversity of oral and skin microbiota was relatively higher in children than in their predecessors. Gut, oral, and skin microbiome was more similar between family members than between unrelated individuals. Meanwhile, 55.05%, 61.09%, and 76.73% of amplicon sequence variants (ASVs) in children's gut, oral, and skin microbiomes, respectively, were transmitted from all family members. Among these, the most transmissible ASVs belonged to Methylophilaceae, Solimonadaceae, Neisseriaceae, and Burkholderiaceae, which were defined as "putative familial transmissible bacteria." Furthermore, we found that the time spent with parents/grandparents and children's dietary preferences were important factors that influenced the proportion of the transmissible microbiome. Moreover, the majority of transmissible ASVs (85.06%), especially those of Ruminococcaceae and Lachnospiraceae, were significantly associated with the immune indices, such as CD3⁺, CD4⁺, CD8⁺, IgG, and IgA. IMPORTANCE Our study revealed that the children's microbiota was partially transmitted from their family members and specific putative transmissible ASVs were associated with the immune system of children. These findings suggest that home life plays a key role in the shaping of young children's microbiomes and has long-term health benefits.

Project description:BackgroundIn modern lifestyles, people make their everyday tasks easier by using household appliances, for example dishwashers. Previous studies showed massive contamination of dishwasher rubber seals with fungi, thus bacterial community, able to survive under harsh conditions, remain undetermined.MethodsBacteria that colonise the extreme environment of household dishwasher rubber seals were investigated using cultivation-dependent and metagenomic approaches. All bacterial isolates were tested for resistance to seven selected antibiotics. Same time bacterial diversity of tap water, connected to the dishwashers was investigated.ResultsAll 30 dishwashers investigated were colonised by various bacteria. Cultivation approaches resulted in 632 bacterial isolates in total, belonging to four phyla, eight classes, 40 genera and 74 species. The majority were Gram-positive, as solely Firmicutes (dominated by the Bacillus cereus group) and Actinobacteria. Gammaproteobacteria were primarily represented by Stenotrophomonas maltophilia, Pseudomonas aeruginosa and Escherichia coli. Metagenomic assessment of the bacterial biodiversity of the dishwasher rubber seals confirmed the predominance of Gram-positive bacteria, as primarily Actinobacteria, followed by Proteobacteria dominated by Gammaproteobacteria, and by pathogenic species such as Escherichia sp., Acinetobacter baumannii, Pseudomonas sp., Stenotrophomonas maltophilia, and Enterobacter sp.. Metagenomic assessment of bacterial biodiversity in the tap water connected to dishwashers revealed predominance of Gram-negative bacteria, in particular Proteobacteria, mainly represented by Tepidimonas sp.. Actinobacteria showed low numbers while no Firmicutes were detected in the tap water. The bacterial diversity of tap water was also lower, 23 genera compared to 39 genera on dishwasher rubber seals. Only 13 out of 49 genera identified by metagenomics approach was found in both environments, of those Gordonia was enriched while half of 13 genera were depleted in dishwashers compared to tap water.ConclusionsThese data indicate that colonisation of dishwasher rubber seals probably depends primarily on the bacterial input from the dirty vessels, and much less on the bacteria in the tap water. Based on the antibiotic resistance data, the dishwasher rubber seal bacterial isolates do not represent a serious threat for the spread of antibiotic resistance into the household environment. Nevertheless dishwashers cannot be ignored as potential sources of human infections, in particular for immuno-compromised individuals.

Project description:The home learning environment plays an important role for children's early competencies development. In particular, the early home literacy environment (HLE) that consists of all literacy resources and interactions in a family that support children's linguistic and literacy learning is closely associated with children's language comprehension and production. A key aspect of the HLE is shared reading that should start early in children's life and should be part of a regular routine in the family. However, parental attitudes toward (shared) reading have hardly been analyzed. In this longitudinal study, we analyzed the associations between parental attitudes toward shared reading and children's linguistic competencies and whether these associations may be mediated by the HLE. Further, we were interested in changes of parental attitudes over time and their association with child and family background characteristics. The sample consisted of N = 133 children with an average age of about 3 years at t1. Children were tested two more times with a 6-month period in-between each assessment. Parental attitudes toward shared reading, socioeconomic status (SES), and the HLE were assessed via parental survey. Children's sentence comprehension, productive language, and grammar were measured with a standardized test battery. Children whose parents had a more positive attitude toward shared reading not only lived in a greater quality HLE but also performed better in the linguistic tests. In a structural equation model, an indirect effect was found showing that the HLE mediated the effect of parental attitudes on children's linguistic competencies. Further, parental attitudes toward shared reading did not change significantly across t1 to t3, and a lower score in the SES scale was associated with a less positive attitude toward shared reading. Consequently, parental attitudes toward shared reading seem to be an important basis for individual differences in the quality of the HLE and also for children's linguistic competencies. As these attitudes vary in the context of different family SES backgrounds, they may be a good target for interventions to support the quality of the HLE and young children's linguistic learning.
