Assessing the Impact of Deep Neural Network-Based Image Denoising on Binary Signal Detection Tasks.

ABSTRACT: A variety of deep neural network (DNN)-based image denoising methods have been proposed for use with medical images. Traditional measures of image quality (IQ) have been employed to optimize and evaluate these methods. However, the objective evaluation of IQ for the DNN-based denoising methods remains largely lacking. In this work, we evaluate the performance of DNN-based denoising methods by use of task-based IQ measures. Specifically, binary signal detection tasks under signal-known-exactly (SKE) with background-known-statistically (BKS) conditions are considered. The performance of the ideal observer (IO) and common linear numerical observers are quantified and detection efficiencies are computed to assess the impact of the denoising operation on task performance. The numerical results indicate that, in the cases considered, the application of a denoising network can result in a loss of task-relevant information in the image. The impact of the depth of the denoising networks on task performance is also assessed. The presented results highlight the need for the objective evaluation of IQ for DNN-based denoising technologies and may suggest future avenues for improving their effectiveness in medical imaging applications.

Project description:PurposeImage quality of positron emission tomography (PET) is limited by various physical degradation factors. Our study aims to perform PET image denoising by utilizing prior information from the same patient. The proposed method is based on unsupervised deep learning, where no training pairs are needed.MethodsIn this method, the prior high-quality image from the patient was employed as the network input and the noisy PET image itself was treated as the training label. Constrained by the network structure and the prior image input, the network was trained to learn the intrinsic structure information from the noisy image and output a restored PET image. To validate the performance of the proposed method, a computer simulation study based on the BrainWeb phantom was first performed. A 68Ga-PRGD2 PET/CT dataset containing 10 patients and a 18F-FDG PET/MR dataset containing 30 patients were later on used for clinical data evaluation. The Gaussian, non-local mean (NLM) using CT/MR image as priors, BM4D, and Deep Decoder methods were included as reference methods. The contrast-to-noise ratio (CNR) improvements were used to rank different methods based on Wilcoxon signed-rank test.ResultsFor the simulation study, contrast recovery coefficient (CRC) vs. standard deviation (STD) curves showed that the proposed method achieved the best performance regarding the bias-variance tradeoff. For the clinical PET/CT dataset, the proposed method achieved the highest CNR improvement ratio (53.35% ± 21.78%), compared with the Gaussian (12.64% ± 6.15%, P = 0.002), NLM guided by CT (24.35% ± 16.30%, P = 0.002), BM4D (38.31% ± 20.26%, P = 0.002), and Deep Decoder (41.67% ± 22.28%, P = 0.002) methods. For the clinical PET/MR dataset, the CNR improvement ratio of the proposed method achieved 46.80% ± 25.23%, higher than the Gaussian (18.16% ± 10.02%, P < 0.0001), NLM guided by MR (25.36% ± 19.48%, P < 0.0001), BM4D (37.02% ± 21.38%, P < 0.0001), and Deep Decoder (30.03% ± 20.64%, P < 0.0001) methods. Restored images for all the datasets demonstrate that the proposed method can effectively smooth out the noise while recovering image details.ConclusionThe proposed unsupervised deep learning framework provides excellent image restoration effects, outperforming the Gaussian, NLM methods, BM4D, and Deep Decoder methods.

Project description:BackgroundSingle photon emission computed tomography (SPECT) is an important functional tool for clinical diagnosis and scientific research of brain disorders, but suffers from limited spatial resolution and high noise due to hardware design and imaging physics. The present study is to develop a deep learning technique for SPECT image reconstruction that directly converts raw projection data to image with high resolution and low noise, while an efficient training method specifically applicable to medical image reconstruction is presented.MethodsA custom software was developed to generate 20,000 2-D brain phantoms, of which 16,000 were used to train the neural network, 2,000 for validation, and the final 2,000 for testing. To reduce development difficulty, a two-step training strategy for network design was adopted. We first compressed full-size activity image (128×128 pixels) to a one-D vector consisting of 256×1 pixels, accomplished by an autoencoder (AE) consisting of an encoder and a decoder. The vector is a good representation of the full-size image in a lower-dimensional space and was used as a compact label to develop the second network that maps between the projection-data domain and the vector domain. Since the label had 256 pixels only, the second network was compact and easy to converge. The second network, when successfully developed, was connected to the decoder (a portion of AE) to decompress the vector to a regular 128×128 image. Therefore, a complex network was essentially divided into two compact neural networks trained separately in sequence but eventually connectable.ResultsA total of 2,000 test examples, a synthetic brain phantom, and de-identified patient data were used to validate SPECTnet. Results obtained from SPECTnet were compared with those obtained from our clinic OS-EM method. Images with lower noise and more accurate information in the uptake areas were obtained by SPECTnet.ConclusionsThe challenge of developing a complex deep neural network is reduced by training two separate compact connectable networks. The combination of the two networks forms the full version of SPECTnet. Results show that the developed neural network can produce more accurate SPECT images.
