TTP protects against acute liver failure by regulating CCL2 and CCL5 through m6A RNA methylation.

ABSTRACT: Tristetraprolin (TTP), an important immunosuppressive protein regulating mRNA decay through recognition of the AU-rich elements (AREs) within the 3'-UTRs of mRNAs, participates in the pathogenesis of liver diseases. However, whether TTP regulates mRNA stability through other mechanisms remains poorly understood. Here, we report that TTP was upregulated in acute liver failure (ALF), resulting in decreased mRNA stabilities of CCL2 and CCL5 through promotion of N6-methyladenosine (m6A) mRNA methylation. Overexpression of TTP could markedly ameliorate hepatic injury in vivo. TTP regulated the mRNA stabilization of CCL2 and CCL5. Interestingly, increased m6A methylation in CCL2 and CCL5 mRNAs promoted TTP-mediated RNA destabilization. Moreover, induction of TTP upregulated expression levels of WT1 associated protein, methyltransferase like 14, and YT521-B homology N6-methyladenosine RNA binding protein 2, which encode enzymes regulating m6A methylation, resulting in a global increase of m6A methylation and amelioration of liver injury due to enhanced degradation of CCL2 and CCL5. These findings suggest a potentially novel mechanism by which TTP modulates mRNA stabilities of CCL2 and CCL5 through m6A RNA methylation, which is involved in the pathogenesis of ALF.

Project description:Ferroptosis is a recently discovered form of programmed cell death, but its regulatory mechanisms remain poorly understood. Here, we show that the RNA-binding protein ZFP36/TTP (ZFP36 ring finger protein) plays a crucial role in regulating ferroptosis in hepatic stellate cells (HSCs). Upon exposure to ferroptosis-inducing compounds, the ubiquitin ligase FBXW7/CDC4 (F-box and WD repeat domain containing 7) decreased ZFP36 protein expression by recognizing SFSGLPS motif. FBXW7 plasmid contributed to classical ferroptotic events, whereas ZFP36 plasmid impaired FBXW7 plasmid-induced HSC ferroptosis. Interestingly, ZFP36 plasmid inhibited macroautophagy/autophagy activation by destabilizing ATG16L1 (autophagy related 16 like 1) mRNA. ATG16L1 plasmid eliminated the inhibitory action of ZFP36 plasmid on ferroptosis, and FBXW7 plasmid enhanced the effect of ATG16L1 plasmid on autophagy. Importantly, ZFP36 plasmid promoted ATG16L1 mRNA decay via binding to the AU-rich elements (AREs) within the 3'-untranslated region. The internal mutation of the ARE region abrogated the ZFP36-mediated ATG16L1 mRNA instability, and prevented ZFP36 plasmid-mediated ferroptosis resistance. In mice, treatment with erastin and sorafenib alleviated murine liver fibrosis by inducing HSC ferroptosis. HSC-specific overexpression of Zfp36 impaired erastin- or sorafenib-induced HSC ferroptosis. Noteworthy, we analyzed the effect of sorafenib on HSC ferroptosis in fibrotic patients with hepatocellular carcinoma receiving sorafenib monotherapy. Attractively, sorafenib monotherapy led to ZFP36 downregulation, ferritinophagy activation, and ferroptosis induction in human HSCs. Project description:To probe the binding sites of MeCbl on GSDME, 0.4 μM recombinant human GSDME (CSB-EP006766HU) purchased from CUSABIO (Wuhan, China) was incubated with 10 μM MeCbl in 100 μL buffer (20 mM HEPES, 150 mM NaCl and 50 μM DPA) at room temperature for 2 h and protected from light. 0.4 μM recombinant human GSDME was incubated with ddH2O as negative control, or with HOCbl for binding sites analysis between GSDME and cobalamin. All groups were equally prepared ten incubation samples and mixed for further analysis. Trypsin and Glu C were then added in the mixture overnight, and formic acid was added to adjust the pH down to three to terminate the enzyme digestion. Activated the C18 desalting column with 100 μL 100% acetonitrile, then centrifuged at 400 g for 3 min. Added 100 μL of 0.1% formic acid and centrifuged at 400 g for 3 min for column equilibration. Replaced with new EP tube, added samples to column, and centrifuged at 400 g for 3 min. Washed the column twice with 100 μL of 0.1% formic acid and centrifuged at 400 g for 3 min, washed once again with 100 μL water at pH10. The eluents were collected with 70% acetonitrile, each sample was lyophilized and stored at -80°C until further mass spectrometry analysis. Prepared the mobile phase solution, A (0.1% (v/v) formic acid in ddH2O), B (0.1% (v/v) formic acid and 80% acetonitrile in ddH2O), the lyophilized samples were dissolved with 10 µL solution A, then centrifuged at 14000 g for 20 min at 4°C, 400 ng supernatant of the samples were injected onto a Thermo Scientific™ UltiMate™ 3000 UHPLC coupled to a timsTOF HT mass spectrometer (BRUKER). Peptides were then separated over a 100 μm ⅹ15 cm ReproSil-Pur C18-AQ 1.5-µm silica beads (Beijing Qinglian Biotech Co.,Ltd, Beijing, China), at a flow rate of 300 nL/min using a gradient: 0-4 min (5-10% B), 4-46 min (10-24% B), 46-53 min (36% B), 53-54 min (95% B), 54-60 (95% B). The timsTOF HT mass spectrometer was used for liquid quality detection with Captive Spray source, and the mass spectrum was collected in DDA mode. The scanning range of the mass spectrum was m/z 100-1700, the primary mass spectrum resolution was set to 60,000 (1222 m/z), and the cumulative time was set to 100 ms in the TIMS tunnel. The capillary voltage is set at 1.6 kV and the mobility is 0.6-1.6 cm2/(V). The total cycle time was 1.1s with 10 PASEF cycles. The mass spectrometry raw data was processed by FragPipe to search against the target protein database. The precursor ion mass tolerance was ±15 ppm; the fragment mass tolerance was ±0.02 Da. The static modification was carbamidomethylation of cysteine; the dynamic modifications were oxidation of methionine, acetylation of N-termini of peptides and C63H91CoN13O14P or C62H88CoN13O14P of cysteine; a maximum of two missed cleavages were allowed.

Project description:Acute liver failure is a severe liver disorder that poses considerable global challenges. Previous studies on Bifidobacterium longum R0175 have mainly focused on its psychotropic functions. The current research focused on the protective efficacy of B. longum R0175 against acute liver failure caused by d-galactosamine (d-GalN) in rats and further tested the hypothesis that B. longum R0175 exerted liver-protective effects by affecting the intestinal microbiota and fecal metabolites and by inhibiting inflammation. We found that oral gavage of B. longum R0175 markedly reduced the severity of liver injury in d-GalN-treated rats, as evidenced by decreased serum levels of aspartate aminotransferase (AST) and total bile acids (TBAs) (P < 0.05). Moreover, the plasma concentrations of proinflammatory cytokines (interleukin 1β [IL-1β] and tumor necrosis factor-α [TNF-α]) and chemokines (granulocyte-macrophage colony-stimulating factor [GM-CSF], macrophage chemoattractant protein 1 [MCP-1], chemokine [C-X-C motif] ligand 1 [CXCL1], chemokine [C-C motif] ligand 5 [CCL5], and macrophage inflammatory protein-1α [MIP-1α]) were also markedly reduced (P < 0.05). Pretreatment with B. longum R0175 partially reversed the gut microbiota dysbiosis in rats with liver injury by increasing the relative abundances of potentially beneficial bacteria, such as Alloprevotella spp., and decreasing the relative abundances of potentially harmful bacteria, such as Acetatifactor muris, Butyricimonas spp., and Oscillibacter spp. Furthermore, B. longum R0175 administration partially improved the metabolic function of the intestinal microbes, as indicated by the decreased level of lithocholic acid found in the feces.IMPORTANCE Our research investigated the protective and preventive roles of B. longum R0175 in a rat model of acute liver failure. The results illustrated that this probiotic strain exhibited protective effects in rats with acute liver failure. Thus, B. longum R0175 showed clinical application prospects that required further exploration.
