L-Arginine Supplementation for Nulliparous Sows during the Last Third of Gestation.

ABSTRACT: We evaluated the effects of L-arginine supplementation during the last third of gestation on molecular mechanisms related to skeletal muscle development of piglets and litter traits at birth. Twenty-three nulliparous sows averaging 205.37 ± 11.50 kg of body weight were randomly assigned to the following experimental treatments: control (CON), where pregnant sows were fed diets to meet their nutritional requirements; arginine (ARG), where sows where fed CON + 1.0% L-arginine. Skeletal muscle from piglets born from sows from ARG group had greater mRNA expression of MYOD (p = 0.043) and MYOG (p ≤ 0.01), and tended to present greater mRNA expression (p = 0.06) of IGF-2 gene compared to those born from CON sows. However, there were no differences (p > 0.05) in the histomorphometric variables of fetuses' skeletal muscle. The total weight of born piglets, total weight of born alive piglets, piglet weight at birth, coefficient of variation of birth weight, and the incidence of intrauterine growth restriction (IUGR) piglets did not differ between groups. No stillborn piglets (p < 0.01) were verified in the ARG sows compared to CON group. The blood levels of estradiol (p = 0.035) and urea (p = 0.03) were higher in ARG sows compared to those from the CON group. In summary, our data show that arginine supplementation of nulliparous sows at late gestation enhance mRNA expression of key myogenic regulatory factors, which likely contribute to improve animal growth rates in later stages of development.

Project description:BackgroundThe prolificacy of sows (litter size at birth) has markedly increased, leading to higher post-natal mortality. Heat stress can exacerbate this issue. Arginine plays an important role in several physiological pathways; its effect on gestating sows can depend on the period of supplementation. This study evaluated the effects of arginine supplementation on the productive performance and physiological status of sows during different gestation periods and seasons, using a multi-omics approach.MethodsA total of 320 sows were divided into 4 groups over 2 seasons (warm/cold); a control group (CO) received a standard diet (including 16.5 g/d of arginine) and 3 other groups received the standard diet supplemented with 21.8 g/d of arginine (38.3 g/d of arginine) either during the first 35 d (Early35), the last 45 d (Late45) or throughout the entire gestation period (COM). The colostrum was analyzed for nutritional composition, immunoglobulins and metabolomic profile. Urine and feces were analyzed on d 35 and 106 for the metabolomic and microbial profiles. Piglet body weight and mortality were recorded at birth, d 6, d 26, and on d 14 post-weaning.ResultsInteractions between arginine and season were never significant. The Early35 group had a lower percentage of stillborn (P < 0.001), mummified (P = 0.002) and low birthweight (LBW) piglets (P = 0.02) than the CO group. The Late45 group had a lower percentage of stillborn piglets (P = 0.029) and a higher percentage of high birthweight piglets (HBW; P < 0.001) than the CO group. The COM group had a higher percentage of LBW (P = 0.004) and crushed piglets (P < 0.001) than the CO group. Arginine supplementation modifies the metabolome characterization of colostrum, urine, and feces. Creatine and nitric oxide pathways, as well as metabolites related to microbial activity, were influenced in all matrices. A slight trend in the beta diversity index was observed in the microbiome profile on d 35 (P = 0.064).ConclusionsArginine supplementation during early gestation reduced the percentage of stillborn and LBW piglets, while in the last third of pregnancy, it favored the percentage of HBW pigs and reduced the percentage of stillbirths, showing that arginine plays a significant role in the physiology of pregnant sows.

Project description:This study was conducted to evaluate the effects of L-arginine supplementation levels during late gestation on reproductive performance and piglet uniformity in high prolific sows. A total of 60 F1 multiparous sows (Yorkshire × Landrace), with an average body weight of 238.2 kg, were allotted to one of three treatment groups in a completely randomized design. The dietary treatments were divided by the supplementation level of arginine during the late-gestation period, from day 70 to farrowing, as follows-(1) CON: corn-soybean meal-based basal diet (Arg 0.72%), (2) Arg10: basal diet + L-Arg 0.28% (Arg 1.0%), and (3) Arg15: basal diet + L-Arg 0.79% (Arg 1.5%). The same lactation diet was provided ad libitum to sows during the lactation period. There were no significant differences in body weight and backfat thickness in sows during late-gestation and lactation. Dietary arginine levels had no significant influences on the number of total born, stillbirth, and born alive. However, increasing inclusion level of L-arginine supplementation tended to increase (p < 0.10) alive litter weight linearly, and also linearly increased (p < 0.05) the piglet weight gain and litter weight gain during the lactation period. In piglet uniformity, the standard deviation of piglet birth weight (p < 0.05) and the coefficient of variation for piglet birth weight (p < 0.10) increased linearly, as dietary arginine levels increased in the late gestation period. Increasing L-arginine supplementation to late gestating sows linearly increased (p < 0.05) the blood concentrations of arginine and ornithine at day 90 and day 110 of gestation. On the other hand, dietary arginine levels in late gestation did not affect the blood parameters related to the nitrogen utilization. Increasing dietary arginine levels for the late gestating sows did not affect the milk composition for colostrum and milk at day 21 of lactation. In conclusion, the inclusion level of arginine in the diet for late gestating sows, by up to 1.5%, could improve the alive litter weight at birth and litter weight gain during lactation, whereas the piglet uniformity at birth was decreased due to the increase of survival for fetuses with light birth weight.

Project description:This study investigated the effects of supplementing sow diets with lysozyme during the late gestation to lactation stage on the performance of sows and their offspring. Sixty sows (Yorkshire × Landrace, 3 to 6 of parity) at day 85 of gestation were allocated to the following 3 dietary treatments: 1) sows fed a basal diet from late gestation to lactation (control, n = 20), 2) sows fed a basal diet with lysozyme 150 g/t (LZM 150, n = 20), and 3) sows fed a basal diet with lysozyme 300 g/t (LZM 300, n = 20). During the lactation period, sows fed diets containing lysozyme had increased average daily feed intake (ADFI) (P < 0.01) and decreased weaning-to-estrus interval (WEI, P < 0.05), but there were no significant effects on backfat during the trial among treatments. Sows fed lysozyme diets had increased (P < 0.05) serum concentration of total protein (TP) compared with those fed the control diets. Serum immunoglobulin M (IgM) of the sows increased (P < 0.05) on day 1 of lactation, immunoglobulin A (IgA) and interleukin-10 (IL-10) increased (P < 0.05) on day 7 of lactation, and immunoglobulin G (IgG) had a tendency to increase (P = 0.05) during the lactation. Milk concentration of IgA increased (P < 0.05) on day 1 and 7 of lactation and tended to be greater (P = 0.06) on day 21 of lactation. No significant differences among the dietary treatments were observed in placental tissue mRNA expression of interleukin-6 (IL-6), IL-10, tumor necrosis factor-α (TNF-α), polymeric immunoglobulin receptor (pIgR), or the concentrations of IL-6, IL-10, TNF-α, or secretory immunoglobulin A (sIgA). Moreover, there was a decrease (P < 0.05) in stillborn in sows fed lysozyme diets. The diarrhea rate decreased (P < 0.05) and serum concentrations of IgA, IgG, IgM, and IL-10 increased (P < 0.05) in piglets from sows fed the diets containing lysozyme compared with piglets from sows fed the control diet. The serum concentrations of TP increased (P < 0.05), and albumin (ALB) and globulin (GLB) had a tendency to increase (P = 0.08, P = 0.06) in piglets from sows fed the diets containing lysozyme compared with piglets from sows fed the control diet. In conclusion, this study indicates that feeding sows diets supplemented with lysozyme from the late gestation through lactation stage increased sow ADFI during the lactation, shortened the WEI, and improved the maternal and offspring health status as indicated by immunological characteristics and a reduced incidence of diarrhea in piglets.

Project description:BackgroundThe degree of adipose tissue development at birth may influence neonatal survival and subsequent health outcomes. Despite their lower birth weights, piglets from Meishan sows (a fat breed with excellent maternal ability) have a higher survival rate than piglets from Large White sows (a lean breed). To identify the main pathways involved in subcutaneous adipose tissue maturation during the last month of gestation, we compared the proteome and the expression levels of some genes at d 90 and d 110 of gestation in purebred and crossbred Large White or Meishan fetuses gestated by sows of either breed.ResultsA total of 52 proteins in fetal subcutaneous adipose tissue were identified as differentially expressed over the course of gestation. Many proteins involved in energy metabolism were more abundant, whereas some proteins participating in cytoskeleton organization were reduced in abundance on d 110 compared with d 90. Irrespective of age, 24 proteins differed in abundance between fetal genotypes, and an interaction effect between fetal age and genotype was observed for 13 proteins. The abundance levels of proteins known to be responsive to nutrient levels such as aldolase and fatty acid binding proteins, as well as the expression levels of FASN, a key lipogenic enzyme, and MLXIPL, a pivotal transcriptional mediator of glucose-related stimulation of lipogenic genes, were elevated in the adipose tissue of pure and crossbred fetuses from Meishan sows. These data suggested that the adipose tissue of these fetuses had superior metabolic functionality, whatever their paternal genes. Conversely, proteins participating in redox homeostasis and apoptotic cell clearance had a lower abundance in Meishan than in Large White fetuses. Time-course differences in adipose tissue protein abundance were revealed between fetal genotypes for a few secreted proteins participating in responses to organic substances, such as alpha-2-HS-glycoprotein, transferrin and albumin.ConclusionsThese results underline the importance of not only fetal age but also maternal intrauterine environment in the regulation of several proteins in subcutaneous adipose tissue. These proteins may be used to estimate the maturity grade of piglet neonates.

Project description:BackgroundThis study aimed to investigate the impacts of guar gum and cellulose as the source of dietary fiber during gestation on the reproductive performance of sows.MethodsA total of 210 sows (parities 3-6) were randomly allocated into six diets (n = 35) throughout gestation to feed graded levels of dietary fiber (DF), including a corn-soybean meal-based control diet with no wheat bran inclusion (CON, 12.5% DF), a wheat bran-rich diet (DF1, 17.4% DF), and another 4 diets (DF2, 17.7% DF; DF3, 18.1% DF; DF4, 18.4% DF; DF5, 18.8% DF) in which wheat bran were equally substituted by 1%, 2%, 3% and 4% purified FIBER MIX (guar gum and cellulose, 1:4). All sows received similar DE and other nutrients throughout gestation.ResultsDF treatment during gestation resulted in normal fecal score (1 to 5 with 1 = dry and 5 = watery) in sows compared with those received the CON diet (P < 0.05). The number of total born piglets had a tendency to be affected by dietary treatment (P = 0.07), and correlation analysis revealed a linear response of total born to dietary fiber levels during gestation (P < 0.01). Sows received the DF2, DF3, and DF5 diets during gestation had a greater ADFI during lactation compared with those in the CON group (P < 0.05) without affecting the daily body weight gain of suckling piglets. Gut microbiota compositions were dramatically changed by the gestation stage and some of those were changed by DF inclusion. Fecal acetate, propionate, and butyrate of sows were markedly increased in late gestation, and butyrate contents in feces of gestating sows were significantly affected by DF levels (P < 0.01). Serum concentrations of pro-inflammatory TNF-α were decreased and anti-inflammatory IL-10 was increased on day 30 of gestation by DF levels (P < 0.05).ConclusionsIn summary, increasing dietary fiber levels by guar gum and cellulose during gestation improved the reproductive performance of sows, which might be related to changes in immunity and gut microbiota of sows.

Project description:IntroductionThe effects of maternal addition of yeast cultures on offspring gut development and intestinal microorganisms are not yet known, so the aim of this study was to investigate the effects of maternal addition of yeast cultures to the diet of sows during late gestation and lactation on growth performance, antioxidant properties and intestinal microorganisms of offspring weaned piglets.Methods40 Landrace × Yorkshire sows (3-7 of parity) with similar backfat were randomly divided into two treatment groups: control diet (CON) and control diet +2.0 g/kg yeast culture (XPC), and the trial started on day 90 of gestation and ended on day 21 of lactation.ResultsThe results showed that maternal addition of yeast culture significantly increased weaned piglet weight and mean daily gain (p < 0.05), with a tendency to increase litter weight gain (p = 0.083) and liver weight (p = 0.076) compared to the control group. The content of thymus malondialdehyde (MDA) was significantly higher (p < 0.05) and the content of colon total antioxidant capacity (T-AOC) was significantly lower (p < 0.05) in the offspring weaned piglets of the XPC group compared to the control group. The expression of thymus SOD1 and SOD2, spleen SOD1, jejunum SOD2, and colon GPX1, SOD1, and SOD2 were significantly downregulated in the XPC group of offspring weaned piglets compared with the control group (p < 0.05). The intestinal morphology and the content of short-chain fatty acids in colonic chyme did not differ between the two groups (p > 0.05). Compared with the control group, the XPC group significantly increased the relative abundance of colonic chyme Bacteroidetes (p < 0.05), tended to decrease the relative abundance of Lactobacillus (p = 0.078), and tended to increase the relative abundance of Alloprevotella (p = 0.055). The XPC group significantly upregulated Blautia and Fournierella (p < 0.05) and significantly downregulated Candidatus_Competibacter, Nitrospira, Dechloromonas, Haliangium, and Oscillospira (p < 0.05).DiscussionIn conclusion, maternal addition of yeast cultures improved the growth performance of offspring weaned piglets and changed the intestinal microbial community, but did not improve their antioxidant performance.

Project description:Pork meat is estimated to be responsible for 10-20% of human salmonellosis cases in Europe. Control strategies at the farm could reduce contamination at the slaughterhouse. One of the targeted sectors of production is maternity, where sows could be Salmonella reservoirs. The aim of this study was to assess the dynamics of shedding of Salmonella in terms of variation in both shedding prevalence and strains excreted during gestation in Quebec's maternity sector. The evolution of the fecal microbiota of these sows during gestation was also assessed to detect bacterial populations associated with these variations. A total of 73 sows both at the beginning and the end of the gestation were randomly selected and their fecal matter was analyzed. Salmonella detection was conducted using a method that includes two selective enrichment media (MSRV and TBG). Nine isolates per positive samples were collected. Among the 73 sows tested, 27 were shedding Salmonella. Sows in the first third of their gestation shed Salmonella significantly more frequently (21/27) than those in the last third (6/46) (χ2P < 0.05). The shedding status of 19 of the sows that were previously sampled in the first third of their gestation was followed, this time in the last third of their gestation, which confirmed reduction of shedding. Using 16S rRNA gene sequencing and qPCR, significant differences between the fecal flora of sows at the beginning and the end of the gestation, shedding Salmonella or not and with different parity number were detected. Using MaAsLin, multiple OTUs were found to be associated with the time of gestation, the status of Salmonella excretion and parity number. Some of the identified taxa could be linked to the reduction of the shedding of Salmonella at the end of gestation. In this study, we showed that the level of Salmonella shedding was variable during gestation with significantly higher shedding at the beginning rather than at the end of gestation. We also observed for the first time a significant change in the microbiota during sow gestation and identified interesting taxa which could be linked to a reduced Salmonella shedding.
