Regulation of Swine Growth by Backfat Tissue during Growing and Finishing Stages.

ABSTRACT: Recently, interest in the function of pig backfat (BF) has increased in the field of livestock animals, and many transcriptome-based studies using commercial pig breeds have been conducted. However, there is a lack of comprehensive studies regarding the biological mechanisms of Korean native pigs (KNPs) and Yorkshire pig crossbreeds. In this study, therefore, BF samples of F1 crossbreeds of KNPs and Yorkshire pigs were investigated to identify differentially expressed genes (DEGs) and their related terms using RNA-sequencing analysis. DEG analysis identified 611 DEGs, of which 182 were up-regulated and 429 were down-regulated. Lipid metabolism was identified in the up-regulated genes, whereas growth and maturation-related terminologies were identified in the down-regulated genes. LEP and ACTC1 were identified as highly connected core genes during functional gene network analysis. Fat tissue was observed to affect lipid metabolism and organ development due to hormonal changes driven by transcriptional alteration. This study provides a comprehensive understanding of BF contribution to crossbreeds of KNPs and Yorkshire pigs during growth periods.

Project description:BackgroundThe interaction of the gut microbiota with key metabolic and physiological processes may be associated with poor growth outcomes in animals born with intrauterine growth restriction (IUGR).ResultsGrowth performance, plasma hormone concentrations, and intestinal microbiota composition were analyzed in IUGR pigs and in normal birth weight (NBW) pigs when the NBW pigs reached 25, 50, and 100 kg of body weight (BW). Compared to NBW pigs, IUGR pigs had lower initial, weaned, and final BW, and lower average daily gain and average daily feed intake in all the considered time points. In the 25 kg BW group, IUGR pigs had higher concentrations of plasma ghrelin and pancreatic polypeptide (PP), but lower insulin concentration than NBW pigs, while the situation was reversed in the 50 kg BW group. As compared to NBW pigs, IUGR pigs had higher microbial alpha diversity in the jejunum and ileum; in the 50 and 100 kg BW groups, IUGR pigs had higher Firmicutes abundance but lower Proteobacteria abundance in the jejunum, and lower Lactobacillus abundance in the jejunum and ileum; in the 25 kg BW group, IUGR pigs showed higher unclassified Ruminococcaceae abundance in the ileum; and in 25 and 50 kg BW groups, IUGR pigs showed lower Ochrobactrum abundance in the jejunum. Spearman's correlation revealed that Lactobacillus was negatively correlated with growth performance, while unclassified Ruminococcaceae was positively correlated. Predictive metagenomic analysis detected significantly different expression of genes in the intestinal microbiota between IUGR and NBW pigs, suggesting different metabolic capabilities between the two groups.ConclusionsGrowing-finishing IUGR pigs showed lower growth performance, higher microbial alpha diversity, and differences in plasma hormone concentrations compared to NBW pigs. Alterations in the abundance of Firmicutes, Proteobacteria, Ruminococcaceae, Lactobacillus, and Ochrobactrum in the small intestine may be associated with IUGR, and may therefore serve as a future target for gut microbiota intervention in growing-finishing IUGR pigs.

Project description:BackgroundGrowing and finishing performances of pigs strongly influence farm efficiency and profitability. The performances of the pigs rely on the herd health status and also on several non-infectious factors. Many recommendations for the improvement of the technical performances of a herd are based on the results of studies assessing the effect of one or a limited number of infections or environmental factors. Few studies investigated jointly the influence of both type of factors on swine herd performances. This work aimed at identifying infectious and non-infectious factors associated with the growing and finishing performances of 41 French swine herds.ResultsTwo groups of herds were identified using a clustering analysis: a cluster of 24 herds with the highest technical performance values (mean average daily gain = 781.1 g/day +/- 26.3; mean feed conversion ratio = 2.5 kg/kg +/- 0.1; mean mortality rate = 4.1% +/- 0.9; and mean carcass slaughter weight = 121.2 kg +/- 5.2) and a cluster of 17 herds with the lowest performance values (mean average daily gain =715.8 g/day +/- 26.5; mean feed conversion ratio = 2.6 kg/kg +/- 0.1; mean mortality rate = 6.8% +/- 2.0; and mean carcass slaughter weight = 117.7 kg +/- 3.6). Multiple correspondence analysis was used to identify factors associated with the level of technical performance. Infection with the porcine reproductive and respiratory syndrome virus and the porcine circovirus type 2 were infectious factors associated with the cluster having the lowest performance values. This cluster also featured farrow-to-finish type herds, a short interval between successive batches of pigs (≤3 weeks) and mixing of pigs from different batches in the growing or/and finishing steps. Inconsistency between nursery and fattening building management was another factor associated with the low-performance cluster. The odds of a herd showing low growing-finishing performance was significantly increased when infected by PRRS virus in the growing-finishing steps (OR = 8.8, 95% confidence interval [95% CI]: 1.8-41.7) and belonging to a farrow-to-finish type herd (OR = 5.1, 95% CI = 1.1-23.8).ConclusionsHerd management and viral infections significantly influenced the performance levels of the swine herds included in this study.

Project description:Traditionally, swine diets have been formulated to meet nutrient requirements at the lowest cost with little regard toward minimizing environmental impacts. The overall objective of this study was to evaluate the relative differences among four grower-finisher feeding programs, using precision diet formulation practices, on growth performance, carcass composition, nitrogen utilization efficiency, and environmental impacts. In experiment 1, four 4-phase growing-finishing feeding programs consisting of diets containing corn and soybean meal (CSBM), low protein CSBM supplemented with crystalline amino acids (LP), CSBM with 30% distillers dried grains with solubles (DDGS), and DDGS supplemented with crystalline Ile, Val, and Trp (DDGS + IVT) were fed to 288 mixed sex pigs (initial body weight [BW] = 36.9 ± 4.2 kg) for 12 wk to determine effects on growth performance and carcass characteristics. Pigs fed with CSBM had greater (P < 0.05) final BW than those fed with LP and DDGS, and greater gain efficiency than pigs fed with LP. Pigs fed with DDGS + IVT tended to have greater (P = 0.06) backfat depth than pigs fed with DDGS, and less (P < 0.05) loin muscle area than pigs fed with CSBM. In experiment 2, nitrogen (N) and phosphorus (P) balance of barrows (n = 32; initial BW = 59.9 ± 5.1 kg) fed with each of the phase-2 diets from experiment 1 was determined in a 12-d metabolism study (7 d adaptation and 5 d collection). Pigs fed with CSBM had a greater (P < 0.05) amount of N retained than pigs fed with other diets, but also had a greater (P < 0.05) amount of urinary N excretion and blood urea N than pigs fed with LP and DDGS + IVT diets. Pigs fed with LP tended (P = 0.07) to have the greatest N utilization efficiency but the least (P < 0.05) P retained as a percentage of P intake among dietary treatments. Diet composition and data collected from experiments 1 and 2 were used to calculate life cycle assessment environmental impacts using Opteinics software (BASF, Lampertheim, Germany). The CSBM feeding program had the least impact on climate change, marine and freshwater eutrophication, and fossil resource use. The LP feeding program had the least impact on acidification, terrestrial eutrophication, and water use, while the DDGS feeding programs had the least impact on land use. These results indicate that feeding CSBM diets optimized growth performance and carcass composition while simultaneously reducing impacts on climate change, marine and freshwater eutrophication, and fossil resource use compared with the other feeding programs evaluated.
