Initial Study on COMT and DRD2 Gene Polymorphisms as Well as the Influence of Temperament and Character Trait on the Severity of Alcohol Craving in Alcohol-Dependent Patients.

ABSTRACT: The main aim of this work was to determine the impact of COMT and DRD2 gene polymorphisms together with temperament and character traits on alcohol craving severity alcohol-dependent persons. The sample comprised of 89 men and 16 women (aged 38±7). For the sake of psychological assessment various analytic methods have been applied like the Short Alcohol Dependence Data Questionnaire (SADD), Penn Alcohol Craving Scale (PACS) or Temperament and Character Inventory (TCI) test. The SNP polymorphism of the analyzed genes was determined by Real Time PCR test. The results showed, that the COMT polymorphismmay have an indirected relationship with the intensity and changes in alcohol craving during abstinence. The DRD2 receptor gene polymorphisms are related with the intensity of alcohol craving. It seems that the character traits like "self-targeting", including "self-acceptance", are more closely related to the severity of alcohol craving and polymorphic changes in the DRD2 receptor than temperamental traits. Although this is a pilot study the obtained results appeared to be promising and clearly indicate the link betweengene polymorphisms alcohol craving and its severity.

Project description:Several lines of evidence suggest that genes involved in dopamine (DA) transmission may contribute to creativity. Among these genes, the catechol-O-methyltransferase gene (COMT) and the dopamine D2 receptor gene (DRD2) are the most promising candidates. Our previous study has revealed evidence for the involvement of DRD2 in creative potential. The present study extended our previous study by systematically exploring the association of COMT with creative potential as well as the interaction between COMT and DRD2. Twelve single nucleotide polymorphisms (SNPs) covering COMT were genotyped in 543 healthy Chinese college students whose creative potentials were assessed by divergent thinking tests. Single SNP analysis showed that rs174697 was nominally associated with verbal originality, two SNPs (rs737865 and rs5993883) were nominally associated with figural fluency, and two SNPs (rs737865 and rs4680) were nominally associated with figural originality. Haplotype analysis showed that, the TCT and CCT haplotype (rs737865-rs174675-rs5993882) were nominally associated with figural originality, and the TATGCAG and CGCGGGA haplotype (rs4646312-rs6269-rs4633-rs6267-rs4818-rs4680-rs769224) were nominally associated with figural originality and verbal flexibility, respectively. However, none of these nominal findings survived correction for multiple testing. Gene-gene interaction analysis identified one significant four-way interaction of rs174675 (COMT), rs174697 (COMT), rs1076560 (DRD2), and rs4436578 (DRD2) on verbal fluency, one significant four-way interaction of rs174675 (COMT), rs4818 (COMT), rs1076560 (DRD2), and rs4648317 (DRD2) on verbal flexibility, and one significant three-way interaction of rs5993883 (COMT), rs4648319 (DRD2), and rs4648317 (DRD2) on figural flexibility. In conclusion, the present study provides nominal evidence for the involvement of COMT in creative potential and suggests that DA related genes may act in coordination to contribute to creativity.

Project description:BackgroundMultiple factors influence the decision to enter a career in medicine and choose a specialty. Previous studies have looked at personality differences in medicine but often were unable to describe the heterogeneity that exists within each specialty. Our study used a person-centered approach to characterize the complex relations between the personality profiles of resident physicians and their choice of specialty.Methods169 resident physicians at a large Midwestern US training hospital completed the Temperament and Character Inventory (TCI) and the Satisfaction with Life Scale (SWLS). Clusters of personality profiles were identified without regard to medical specialty, and then the personality clusters were tested for association with their choice of specialty by co-clustering analysis. Life satisfaction was tested for association with personality traits and medical specialty by linear regression and analysis of variance.ResultsWe identified five clusters of people with distinct personality profiles, and found that these were associated with particular medical specialties Physicians with an "investigative" personality profile often chose pathology or internal medicine, those with a "commanding" personality often chose general surgery, "rescuers" often chose emergency medicine, the "dependable" often chose pediatrics, and the "compassionate" often chose psychiatry. Life satisfaction scores were not enhanced by personality-specialty congruence, but were related strongly to self-directedness regardless of specialty.ConclusionsThe personality profiles of physicians were strongly associated with their medical specialty choices. Nevertheless, the relationships were complex: physicians with each personality profile went into a variety of medical specialties, and physicians in each medical specialty had variable personality profiles. The plasticity and resilience of physicians were more important for their life satisfaction than was matching personality to the prototype of a particular specialty.

Project description:BackgroundPersonality is the major predictor of people's subjective well-being (i.e., positive affect, negative affect, and life satisfaction). Recent research in countries with high-income and strong self-transcendent values shows that well-being depends on multidimensional configurations of temperament and character traits (i.e., Joint Personality Networks) that regulate the way people learn to adapt their habits to be in accord with their goals and values, rather than individual traits. To evaluate the prevalence and the associations of different Joint Personality (temperament-character) Networks with well-being in a low-income country with weak self-transcendent values, we tested their association in Bulgarian adults, a population known to have strong secular-rationalist values but weak self-transcendent values.MethodThe sample consisted of 443 individuals from Bulgaria (68.70% females) with a mean age of 34 years (SD = 15.05). Participants self-reported personality (Temperament and Character Inventory), affect (Positive Affect Negative Affect Schedule), and life satisfaction (Satisfaction with Life Scale). The personality scores were used for profiling through latent profile analysis and latent class analysis based on temperament configurations (i.e., Temperament Profiles) of high/low scores of Novelty Seeking (N/n), Harm Avoidance (H/h), Reward Dependence (R/r), and Persistence (P/s); and character configurations (i.e., Character Profiles) of high/low scores of Self-Directedness (S/s), Cooperativeness (C/c), and Self-Transcendence (T/t).ResultsWe found two Temperament Profiles and two Character Profiles that clustered into two distinctive Joint Personality Networks. All individuals in Joint Personality Network 1 had a Reliable (nhRP) Temperament Profile in combination with an Organized (SCt) Character Profile (i.e., a stable temperament and a healthy character configuration). About 71.9% in Joint Personality Network 2 had an Apathetic (sct) Character Profile in combination with Methodical (nHrp) or Reliable (nhRP) Temperament Profiles, while 28.1% had a Methodical (nHrp) Temperament Profile in combination with an Organized (SCt) Character Profile. Few people with high self-expressive values (i.e., high in all three character traits; SCT) were found. Individuals with a Joint Personality Network 1 with strong secular-rationalist values reported higher levels of positive affect and life satisfaction (p < .001), while individuals with a Joint Personality Network 2 reported higher levels of negative affect (p < .001).ConclusionsAlthough a stable temperament and a healthy character were separately important for well-being, it was clear that it was the interaction between such temperament and character configuration that yielded greater levels of subjective well-being. Nevertheless, future research needs to investigate this interaction further to evaluate other cultures with variable configurations of personality traits and values.

Project description:IntroductionA common practice in research and clinical practice is to use data considered representative of a target population to compare and understand the personality characteristics of specific groups or specific individuals. To this end, numerous studies have presented normative data for the temperament and character traits outlined in Cloninger's psychobiological model of personality. However, recent genomic evidence demonstrates that human personality is organized as a complex hierarchy that ascends beyond the individual traits to multi-trait profiles that regulate emotional reactivity (temperament profiles) or goals and values (character profiles), and then to three phenotypic networks, which integrate temperament profiles and character profiles, that regulate learning. Given this recent understanding, our aim was to provide a novel and more comprehensive description of personality features at a societal level (using a stratified sample representative of the Portuguese population) by considering personality at its higher levels of complexity.MethodsToward this goal, a stratified sample of 2,443 Portuguese adults responded to the Revised Temperament and Character Inventory (TCI-R).ResultsWe summarize the prevalence of (a) temperament profiles, (b) character profiles, and (c) integrated temperament-character networks within the whole sample, as well as for men vs. women and different age groups separately. Independent of age and education, women were more likely to be capable of resourceful productivity and helpful cooperation combined with being more intuitive, meditative and creative than men. Independent of age and gender, individuals with a degree were also more likely to present these biopsychosocial features. We also found that the organized character profile was most typical of adults in their 40s. Finally, the distribution of personality profiles across age differed as a function of gender: for men the oldest individuals had the most coherent personalities while high personality integration was most prevalent for women in their 30s.DiscussionThese results have strong implications for research and intervention. In particular, these results are relevant for understanding the epidemiology of interactions between personality, mental health and well-being, including their expressions in a national population as a function of demographic characteristics.
