A Mix of Dietary Fibres Changes Interorgan Nutrients Exchanges and Muscle-Adipose Energy Handling in Overfed Mini-Pigs.

ABSTRACT: This study evaluates the capacity of a bread enriched with fermentable dietary fibres to modulate the metabolism and nutrients handling between tissues, gut and peripheral, in a context of overfeeding. Net fluxes of glucose, lactate, urea, short chain fatty acids (SCFA), and amino acids were recorded in control and overfed female mini-pigs supplemented or not with fibre-enriched bread. SCFA in fecal water and gene expressions, but not protein levels or metabolic fluxes, were measured in muscle, adipose tissue, and intestine. Fibre supplementation increased the potential for fatty acid oxidation and mitochondrial activity in muscle (acox, ucp2, sdha and cpt1-m, p < 0.05) as well as main regulatory transcription factors of metabolic activity such as pparα, pgc-1α and nrf2. All these features were associated with a reduced muscle fibre cross sectional area, resembling to controls (i.e., lean phenotype). SCFA may be direct inducers of these cross-talk alterations, as their feces content (+52%, p = 0.05) was increased in fibre-supplemented mini-pigs. The SCFA effects could be mediated at the gut level by an increased production of incretins (increased gcg mRNA, p < 0.05) and an up-regulation of SCFA receptors (increased gpr41 mRNA, p < 0.01). Hence, consumption of supplemented bread with fermentable fibres can be an appropriate strategy to activate muscle energy catabolism and limit the establishment of an obese phenotype.

Project description:Perivascular adipose tissue (PVAT), a special type of adipose tissue, closely surrounds vascular adventitia and produces numerous bioactive substances to maintain vascular homeostasis. PVAT dysfunction has a crucial role in regulating vascular remodeling, but the exact mechanisms remain unclear. In this study, we investigated whether and how obesity-induced PVAT dysfunction affected adventitia remodeling in early vascular injury stages. Mini pigs were fed a high sugar and fat diet for 6 months to induce metabolic syndrome and obesity. In the mini pigs, left carotid vascular injury was then generated using balloon dilation. Compared with normal mini pigs, obese mini pigs displayed significantly enhanced vascular injury-induced adventitial responses, evidenced by adventitia fibroblast (AF) proliferation and differentiation, and adventitia fibrosis, as well as exacerbated PVAT dysfunction characterized by increased accumulation of resident macrophages, particularly the M1 pro-inflammatory phenotype, increased expression of leptin and decreased expression of adiponectin, and production of pro-inflammatory cytokines interleukin (IL)-1β and IL-18. Primary AFs cultured in PVAT-conditioned medium from obese mini pigs also showed significantly increased proliferation and differentiation. We further revealed that activated nod-like receptor protein 3 (NLRP3) inflammasome and its downstream products, i.e., IL-1 family members such as IL-1β and IL-18 were upregulated in the PVAT of obese mini pigs; PVAT dysfunction was also demonstrated in preadipocytes treated with palmitic acid. Finally, we showed that pretreatment with IL-1 receptor (IL-1R) antagonist or IL-1R knockdown blocked AF proliferation and differentiation in AFs cultured in PVAT-conditioned medium. These results demonstrate that obesity-induced PVAT dysfunction aggravates adventitial remodeling after early vascular injury with elevated AF proliferation and differentiation via activating the NLRP3/IL-1 signaling pathway.

Project description:Aerial ammonia exposure leads to tissue damage and metabolic dysfunction. However, it is unclear how different organs are coordinated to defend against aerial ammonia exposure. Twenty-four pigs were randomly divided into 4 groups, exposed to 0, 10, 25 or 35 mg/m3 ammonia respectively for 25 d. After above 25 mg/m3 ammonia exposure, decreased aspartate (P = 0.016), glutamate (P = 0.030) and increased ornithine (P = 0.002) were found in the ammonia-removing liver, and after high ammonia (35 mg/m3) exposure, glutamine synthetase (GS) expression was increased (P = 0.012). An increased glutamate (P = 0.004) and decreased glutaminase (GLS) expression (P = 0.083) were observed in the lungs after high ammonia exposure. There was also an increasing trend of glutamine in the kidneys after high ammonia exposure (P = 0.066). For branched-chain amino acid (BCAA) catabolism, high ammonia exposure increased BCAA content in both the lungs and muscle (P < 0.05), whereas below 25 mg/m3 ammonia exposure increased BCAA only in the lungs (P < 0.05). The expression of BCAA transaminase (BCAT1/2) and dehydrogenase complex (BCKDHA/B and DBT) were inhibited to a varying degree in the liver, lungs and muscle after above 25 mg/m3 ammonia exposure, especially high ammonia exposure. The expression of BCKDH complex and glutamate-glutamine metabolism-related genes were highly expressed in the liver, followed by the lungs and muscle (P < 0.01), whereas the BCAT2 expression was highest in the lungs (P = 0.002). Altogether, low ammonia exposure sufficed to evoke the urea cycle to detoxify ammonia in the liver. The process of ammonia removal in the liver and potential ability of the lungs to detoxify ammonia were enhanced with increasing ammonia. Furthermore, high ammonia exposure impaired the BCAA catabolism and decreased the transcripts of the BCAA catabolism-related enzymes, resulting in high BCAA content in extrahepatic tissues. Therefore, with aerial ammonia increasing, an increased urea cycle and glutamine synthesis were ammonia defensive strategies, and high ammonia exposure impaired the BCAA catabolism.
