Sensorimotor Self-organization via Circular-Reactions.

ABSTRACT: Newborns demonstrate innate abilities in coordinating their sensory and motor systems through reflexes. One notable characteristic is circular reactions consisting of self-generated motor actions that lead to correlated sensory and motor activities. This paper describes a model for goal-directed reaching based on circular reactions and exocentric reference-frames. The model is built using physiologically plausible visual processing modules and arm-control neural networks. The model incorporates map representations with ego- and exo-centric reference frames for sensory inputs, vector representations for motor systems, as well as local associative learning that result from arm explorations. The integration of these modules is simulated and tested in a three-dimensional spatial environment using Unity3D. The results show that, through self-generated activities, the model self-organizes to generate accurate arm movements that are tolerant with respect to various sources of noise.

Self-Organization of Binary Colloidal Mixtures via Diffusiophoresis.

Project description:Catalytic activity of the colloids and chemotactic response to gradients of the chemicals in the solution leads to effective interaction between catalytic colloids. In this paper, we simulate mixtures of active and passive colloids via a Brownian dynamics algorithm. These particles interact via phoretic interactions, which are determined by two independent parameters, surface activity and surface mobility. We find rich dynamic structures by tuning passive colloids' surface mobility, size, and area fractions, which include schools of active colloids with exclusion zone, yolk/shell cluster, and stable active-passive alloys to motile clusters. Dynamical cluster can also be formed due to the nonreciprocity of the phoretic interaction. Increasing the size ratio of passive colloids to active colloids favors the phase separation of active and passive colloids, resulting in yolk/shell structure. Increasing the area fraction of active colloids tends to transfer from dynamical clusters into stable alloys. The simulated binary active colloid systems exhibit intriguing nonequilibrium phenomena that mimic the dynamic organizations of active/passive systems.

Sensorimotor organization of a sustained involuntary movement.

Project description:Involuntary movements share much of the motor control circuitry used for voluntary movement, yet the two can be easily distinguished. The Kohnstamm phenomenon (where a sustained, hard push produces subsequent involuntary arm raising) is a useful experimental model for exploring differences between voluntary and involuntary movement. Both central and peripheral accounts have been proposed, but little is known regarding how the putative Kohnstamm generator responds to afferent input. We addressed this by obstructing the involuntary upward movement of the arm. Obstruction prevented the rising EMG pattern that characterizes the Kohnstamm. Importantly, once the obstruction was removed, the EMG signal resumed its former increase, suggesting a generator that persists despite peripheral input. When only one arm was obstructed during bilateral involuntary movements, only the EMG signal from the obstructed arm showed the effect. Upon release of the obstacle, the obstructed arm reached the same position and EMG level as the unobstructed arm. Comparison to matched voluntary movements revealed a preserved stretch response when a Kohnstamm movement first contacts an obstacle, and also an overestimation of the perceived contact force. Our findings support a hybrid central and peripheral account of the Kohnstamm phenomenon. The strange subjective experience of this involuntary movement is consistent with the view that movement awareness depends strongly on efference copies, but that the Kohnstamm generator does not produces efference copies.

Sensorimotor transformation via sparse coding.

Project description:Sensorimotor transformation is indispensable to the accurate motion of the human body in daily life. For instance, when we grasp an object, the distance from our hands to an object needs to be calculated by integrating multisensory inputs, and our motor system needs to appropriately activate the arm and hand muscles to minimize the distance. The sensorimotor transformation is implemented in our neural systems, and recent advances in measurement techniques have revealed an important property of neural systems: a small percentage of neurons exhibits extensive activity while a large percentage shows little activity, i.e., sparse coding. However, we do not yet know the functional role of sparse coding in sensorimotor transformation. In this paper, I show that sparse coding enables complete and robust learning in sensorimotor transformation. In general, if a neural network is trained to maximize the performance on training data, the network shows poor performance on test data. Nevertheless, sparse coding renders compatible the performance of the network on both training and test data. Furthermore, sparse coding can reproduce reported neural activities. Thus, I conclude that sparse coding is necessary and a biologically plausible factor in sensorimotor transformation.

Self-organization of maze-like structures via guided wrinkling.

Project description:Sophisticated three-dimensional (3D) structures found in nature are self-organized by bottom-up natural processes. To artificially construct these complex systems, various bottom-up fabrication methods, designed to transform 2D structures into 3D structures, have been developed as alternatives to conventional top-down lithography processes. We present a different self-organization approach, where we construct microstructures with periodic and ordered, but with random architecture, like mazes. For this purpose, we transformed planar surfaces using wrinkling to directly use randomly generated ridges as maze walls. Highly regular maze structures, consisting of several tessellations with customized designs, were fabricated by precisely controlling wrinkling with the ridge-guiding structure, analogous to the creases in origami. The method presented here could have widespread applications in various material systems with multiple length scales.

Thermoresponsive Polyphosphoester via Polycondensation Reactions: Synthesis, Characterization, and Self-Assembly.

Project description:Using a novel strategy, amphiphilic polyphosphoesters based on poly(oxyethylene H-phosphonate)s (POEHP) with different poly(ethylene glycol) segment lengths and aliphatic alcohols with various alkyl chain lengths were synthesized using polycondensation reactions. They were characterized by 1H NMR, 13C {H} NMR 31P NMR, IR, and size exclusion chromatography (SEC). The effects of the polymer structure on micelle formation and stability, micelle size, and critical micelle temperature were studied via dynamic light scattering (DLS). The hydrophilic/hydrophobic balance of these polymers can be controlled by changing the chain lengths of hydrophilic PEG and hydrophobic alcohols. A solubilizing test, using Sudan III, revealed that hydrophobic substances can be incorporated inside the hydrophobic core of polymer associates. Loading capacity depends on the length of alkyl side chains. The results obtained indicate that these structurally flexible polymers have the potential as drug carriers.

Functional organization of human sensorimotor cortex for speech articulation.

Project description:Speaking is one of the most complex actions that we perform, but nearly all of us learn to do it effortlessly. Production of fluent speech requires the precise, coordinated movement of multiple articulators (for example, the lips, jaw, tongue and larynx) over rapid time scales. Here we used high-resolution, multi-electrode cortical recordings during the production of consonant-vowel syllables to determine the organization of speech sensorimotor cortex in humans. We found speech-articulator representations that are arranged somatotopically on ventral pre- and post-central gyri, and that partially overlap at individual electrodes. These representations were coordinated temporally as sequences during syllable production. Spatial patterns of cortical activity showed an emergent, population-level representation, which was organized by phonetic features. Over tens of milliseconds, the spatial patterns transitioned between distinct representations for different consonants and vowels. These results reveal the dynamic organization of speech sensorimotor cortex during the generation of multi-articulator movements that underlies our ability to speak.

Femtosecond photoelectron circular dichroism of chemical reactions.

Project description:Understanding the chirality of molecular reaction pathways is essential for a broad range of fundamental and applied sciences. However, the current ability to probe chirality on the time scale of primary processes underlying chemical reactions remains very limited. Here, we demonstrate time-resolved photoelectron circular dichroism (TRPECD) with ultrashort circularly polarized vacuum-ultraviolet (VUV) pulses from a tabletop source. We demonstrate the capabilities of VUV-TRPECD by resolving the chirality changes in time during the photodissociation of atomic iodine from two chiral molecules. We identify several general key features of TRPECD, which include the ability to probe dynamical chirality along the complete photochemical reaction path, the sensitivity to the local chirality of the evolving scattering potential, and the influence of electron scattering off dissociating photofragments. Our results are interpreted by comparison with high-level ab-initio calculations of transient PECDs from molecular photoionization calculations. Our experimental and theoretical techniques define a general approach to femtochirality.

Regaining motor control in musician's dystonia by restoring sensorimotor organization.

Project description:Professional musicians are an excellent model of long-term motor learning effects on structure and function of the sensorimotor system. However, intensive motor skill training has been associated with task-specific deficiency in hand motor control, which has a higher prevalence among musicians (musician's dystonia) than in the general population. Using a transcranial magnetic stimulation paradigm, we previously found an expanded spatial integration of proprioceptive input into the hand motor cortex [sensorimotor organization (SMO)] in healthy musicians. In musician's dystonia, however, this expansion was even larger. Whereas motor skills of musicians are likely to be supported by a spatially expanded SMO, we hypothesized that in musician's dystonia this might have developed too far and now disrupts rather than assists task-specific motor control. If so, motor control should be regained by reversing the excessive reorganization in musician's dystonia. Here, we test this hypothesis and show that a 15 min intervention with proprioceptive input (proprioceptive training) restored SMO in pianists with musician's dystonia to the pattern seen in healthy pianists. Crucially, task-specific motor control improved significantly and objectively as measured with a MIDI (musical instrument digital interface) piano, and the amount of behavioral improvement was significantly correlated to the degree of sensorimotor reorganization. In healthy pianists and nonmusicians, the SMO and motor performance remained essentially unchanged. These findings suggest that the differentiation of SMO in the hand motor cortex and the degree of motor control of intensively practiced tasks are significantly linked and finely balanced. Proprioceptive training restored this balance in musician's dystonia to the behaviorally beneficial level of healthy musicians.

Self-configuring feedback loops for sensorimotor control.

Project description:How dynamic interactions between nervous system regions in mammals performs online motor control remains an unsolved problem. In this paper, we show that feedback control is a simple, yet powerful way to understand the neural dynamics of sensorimotor control. We make our case using a minimal model comprising spinal cord, sensory and motor cortex, coupled by long connections that are plastic. It succeeds in learning how to perform reaching movements of a planar arm with 6 muscles in several directions from scratch. The model satisfies biological plausibility constraints, like neural implementation, transmission delays, local synaptic learning and continuous online learning. Using differential Hebbian plasticity the model can go from motor babbling to reaching arbitrary targets in less than 10 min of in silico time. Moreover, independently of the learning mechanism, properly configured feedback control has many emergent properties: neural populations in motor cortex show directional tuning and oscillatory dynamics, the spinal cord creates convergent force fields that add linearly, and movements are ataxic (as in a motor system without a cerebellum).

Circular RNA circIPO11 drives self-renewal of liver cancer initiating cells via Hedgehog signaling.

Project description:BackgroundHepatocellular carcinoma (HCC) is one of the most intractable tumors in the world due to its high rate of recurrence and heterogeneity. Liver cancer initiating cells also called cancer stem cells (CSCs) play a critical role in resistance against typical therapy and high tumor-initiating potential. However, the role of the novel circular RNA (circRNA) circIPO11 in the maintenance of liver cancer initiating cells remains elusive.MethodsCircRNAs highly conserved in humans and mice were identified from 3 primary HCC samples by circRNA array. The expression and function of circIPO11 were further evaluated by Northern blot, limiting dilution xenograft analysis, chromatin isolation by RNA purification-PCR assay (ChIRP) and HCC patient-derived tumor cells (PDC) models. CircIpo11 knockout (KO) mice were generated by a CRISPR/Cas9 technology.ResultsCircIPO11 is highly expressed in HCC tumor tissues and liver CSCs. CircIPO11 is required for the self-renewal maintenance of liver CSCs to initiate HCC development. Mechanistically, circIPO11 recruits TOP1 to GLI1 promoter to trigger its transcription, leading to the activation of Hedgehog signaling. Moreover, GLI1 is also highly expressed in HCC tumor tissues and liver CSCs, and TOP1 expression levels positively correlate with the metastasis, recurrence and survival of HCC patients. Additionally, circIPO11 knockout in mice suppresses the progression of chemically induced liver cancer development.ConclusionOur findings reveal that circIPO11 drives the self-renewal of liver CSCs and promotes the propagation of HCC via activating Hedgehog signaling pathway. Antisense oligonucleotides (ASOs) against circIPO11 combined with TOP1 inhibitor camptothecin (CPT) exert synergistic antitumor effect. Therefore, circIPO11 and the Hedgehog signaling pathway may provide new potential targets for the treatment of HCC patients.
