Carboxylato-Pillar[6]arene-Based Fluorescent Indicator Displacement Assays for Caffeine Sensing.

ABSTRACT: In the present work, we have developed a new indicator displacement system based on pillararene for anionic water-soluble carboxylato pillar [6] arene (WP6) and aromatic fluorescent dye safranine T (ST). A large fluorescence enhancement and colour change of ST were observed after complexation with electron-rich cavity in WP6 because of host-guest twisted intramolecular charge-transfer interactions. The constructed pillararene-indicator displacement system can be applied for caffeine selective detection in water.

Project description:The complexation of three cationic fluorescent dye guests with the anionic host carboxylato-pillar[6]arene (WP6) was investigated by optical and NMR spectroscopy. Among the selected indicators - a stilbazolium dye (i1) and two naphthalimide derivatives with positively charged 'anchor' groups (i2 and i3) - i1 gave a large turn-on, i2 and i3 a large turn-off fluorescence response to the complexation. The size selectivity of the complex formation of pillararenes was demonstrated by comparing the binding constants of the complexes of the three indicators with WP6 and its smaller homologue, WP5. The systems WP6·i1 and WP6·i2 were tested as indicator displacement assays for the sensing of monoamine neurotransmitters. The WP6·i1 system functioned as a turn-off, the WP6·i2 system as a turn-on sensor for neurotransmitters, and both assays showed a good selectivity to histamine over the other neurotransmitter analytes.

Selective Fluorescent Sensing for Iron in Aqueous Solution by A Novel Functionalized Pillar[5]arene.

Project description:Iron ion is one of the most physiologically important elements in metabolic processes, indispensable for all living systems. Since its excess can lead to severe diseases, new approaches for its monitoring in water samples are urgently needed to meet requirements. Here, we firstly report a novel and universal route for the synthesis of a series of pillar[n]arene derivates containing one benzoquinone unit by photocatalysis. With this in hand, an anthracene - appended water - soluble pillar[5]arene (H) with excellent fluorescence sensing potency was prepared. H enabled the ultrasensitive detection of iron ions in aqueous solution with limits of detection of 10-8 M. Over a wide range of metal ions, H exhibited specific selectivity toward Fe3+ . More importantly, H could still properly operate in a simulated sewage sample, coexisting with multiple interference ions.

Novel Competitive Fluorescence Sensing Platform for L-carnitine Based on Cationic Pillar[5]Arene Modified Gold Nanoparticles.

Project description:Supramolecular host-guest interaction and sensing between cationic pillar[5]arenes (CP5) and L-carnitine were developed by the competitive host-guest recognition for the first time. The fluorescence sensing platform was constructed by CP5 functionalized Au nanoparticles (CP5@Au-NPs) as receptor and probe (rhodamine 123, R123), which shown high sensitivity and selectivity for L-carnitine detection. Due to the negative charge and molecular size properties of L-carnitine, it can be highly captured by the CP5 via electrostatic interactions and hydrophobic interactions. The host-guest mechanism between PP5 and L-carnitine was studied by ¹H NMR and molecular docking, indicating that more affinity binding force of CP5 with L-carnitine. Therefore, a selective and sensitive fluorescent method was developed. It has a linear response of 0.1⁻2.0 and 2.0⁻25.0 μM and a detection limit of 0.067 μM (S/N = 3). The fluorescent sensing platform was also used to detect L-carnitine in human serum and milk samples, which provided potential applications for the detection of drugs abuse and had path for guarding a serious food safety issues.

Multistimuli Sensitive Behavior of Novel Bodipy-Involved Pillar[5]arene-Based Fluorescent [2]Rotaxane and Its Supramolecular Gel.

Project description:Fluorescent [2]rotaxane BC12P5 is successfully constructed with 1,4-diethoxypillar[5]arene as wheel over a long alkyl axle with Bodipy chromophore as one stopper for the first time. NMR spectra clearly reveal its molecular shuttle nature triggered by multiple external stimuli including solvent polarity and temperature. In particular, the fluorescence nature introduced into [2]rotaxane BC12P5 renders it a good sensor for the external stimuli. Nevertheless, the supramolecular gel successfully fabricated from this novel rotaxane system via self-assembly in dimethyl sulfoxide (DMSO) also shows reversible gel-sol phase transition upon multiple external stimuli such as heating/cooling, shaking/resting, or the addition of different anions. Interestingly, exposure of the supramolecular gel film to HCl or ammonia vapor induces the change in the film fluorescence intensity, endowing this system with a potential application in gas detecting.

A stimuli-responsive pillar[5]arene-based hybrid material with enhanced tunable multicolor luminescence and ion-sensing ability.

Project description:Tunable luminescent materials are becoming more and more important owing to their broad application potential in various fields. Here we construct a pillar[5]arene-based hybrid material with stimuli-responsive luminescent properties and ion-sensing abilities from a pyridine-modified conjugated pillar[5]arene and a planar chromophore oligo(phenylenevinylene) upon coordination of Cd (II) metal cores. This new material not only shows an optimized luminescence due to the minimized π-π stacking and efficient charge transfer properties benefitting from the existence of pillar[5]arene rings, but also exhibits tunable multicolor emission induced by different external stimuli including solvent, ions and acid, indicating great application potential as a fluorescent sensory material, especially for Fe3+. With this pillar[5]arene-based dual-ligand hybrid material, valid optimization and regulation on the fluorescence of the original chromophore have been achieved, which demonstrates a plausible strategy for the design of tunable solid-state luminescent materials and also a prototypical model for the effective regulation of fluorescent properties of planar π systems using synthetic macrocycle-based building blocks.

Synthesis, Optical Properties and Cellular Toxicity of Water-Soluble near Infrared-II Fluorescent Assemblies Based on Pillar[5]arene.

Project description:The main challenges in second near-infrared region molecular fluorophores are poor water solubility and unknown long-term toxicity at present. Herein, new NIR-II molecular fluorophores have been designed and employed to integrate biocompatible pillar[5]arene with 10 outer triethylene oxide groups for the synthesis of rotaxane IRCR. In addition, PEGylated pillar[5]arenes have been combined for the self-assembly of two supramolecular vesicular systems, i.e., PP5-IR1 and PP5-IR2, affording aqueous solubility and lowered cellular toxicity. In aqueous solution, all these fluorophores displayed room-temperature emission with λmax at 986-1013 nm and quantum yields of 0.54-1.45%. They also exhibited good chemical stability and reasonable self-assembled sizes, which may find potential applications in NIR-II imaging. In addition, PP5-IR1 can be used as a fluorescent chemosensor for selective recognition of glutathione through the cleavage of dinitrophenyl ether and release the fluorescent dye.

Project description:The current study investigated host-guest complexation in 6-p-toluidinylnaphthalene-2-sulfonate (TNS), a fluorescence probe used to investigate hydrophobic regions that contain the water-soluble cationic pillar[6]arene (CP6). After complexation with CP6, the fluorescence intensity of TNS was significantly increased. The decreases in the fluorescence intensity of the TNS•CP6 complex when phenolic food-additives are added have been used in indicator displacement assays to detect food additives in the water.

A Fluorescence Sensing Determination of 2, 4, 6-Trinitrophenol Based on Cationic Water-Soluble Pillar[6]arene Graphene Nanocomposite.

Project description:We describe a selective and sensitive fluorescence platform for the detection of trinitrophenol (TNP) based on competitive host⁻guest recognition between pyridine-functionalized pillar[6]arene (PCP6) and a probe (acridine orange, AO) that used PCP6-functionalized reduced graphene (PCP6-rGO) as the receptor. TNP is an electron-deficient and negative molecule, which is captured by PCP6 via electrostatic interactions and π⁻π interactions. Therefore, a selective and sensitive fluorescence probe for TNP detection is developed. It has a low detection limit of 0.0035 μM (S/N = 3) and a wider linear response of 0.01⁻5.0 and 5.0⁻125.0 for TNP. The sensing platform is also used to test TNP in two water and soil samples with satisfying results. This suggests that this approach has potential applications for the determination of TNP.

A pillar[5]arene-based [2]rotaxane lights up mitochondria.

Project description:Subcellular organelle-specific reagents for simultaneous targeting, imaging and treatment are highly desirable for cancer therapy. However, it remains a challenge to fabricate a single molecular platform containing a targeting group, imaging and therapeutic agents through traditional synthesis. Due to their superior sensitivity and photostability, fluorescent probes with aggregation-induced emission (AIE) characteristics have attracted more and more attention in studying the process of translocation, drug release, and excretion of nanomedicines in vitro or in vivo. We construct a pillar[5]arene-based [2]rotaxane (R1) by employing tetraphenylethene (TPE) and triphenylphosphonium (TPP) moieties as stoppers; the TPE unit retains the aggregation-induced emission (AIE) attribute and the TPP group is used as a mitochondria-targeting agent. R1 exhibits enhanced AIE, high specificity to mitochondria, and superior photostability. By introducing doxorubicin (DOX) into R1, prodrug R2 is constructed as a dual-fluorescence-quenched Förster resonance energy transfer (FRET) system, in which the TPE-based axle acts as a donor fluorophore and the DOX unit acts as the acceptor. Upon hydrolysis of R2 in endo/lysosomes, the fluorescences of the carrier and the drug recover. R1 is further utilized as a drug delivery platform to conjugate other anticancer drugs containing amine groups through imine formation to prepare prodrugs. The anticancer drugs are released from these prodrugs in the cells upon hydrolysis of the pH-responsive imine bonds.

Porous pillar[6]arene-based polymers for reversible iodine capture.

Project description:Iodine in nuclear waste can cause serious environment pollution and health risks, and has thus driven more development of materials for iodine capture. Herein, a novel porous pillar[6]arene-based polymer (P-P6APs) was easily prepared as a supramolecular adsorbent for iodine via a one-step crosslinking reaction between per-hydroxylated pillar[6]arene and decafluorobiphenyl. Nitrogen adsorption tests demonstrated that this material possessed a satisfactory surface area (S BET = 366 m2 g-1) and pore diameter (3.8 nm), which was due to the macrocyclic scaffolds. Compared with commercially available activated carbon, P-P6APs exhibited superior adsorption efficiency toward volatile iodine not only in the solution phase (water and n-hexane) but also in the gas phase. This outcome was mainly ascribed to nonspecific adsorption by the pore structure of the crosslinked material coupled with multiple intermolecular binding sites of the macrocyclic scaffold. Moreover, this supramolecular absorbent was recyclable and could be reused 5 times with no obvious loss of performance.
