The complete mitochondrial genome of a polyphagous insect: Colasposoma dauricum (Coleoptera: Chrysomelidae: Eumolpinae).

ABSTRACT: Colasposoma dauricum Mannerheim, 1849, is an important insect pest distributed in most areas of China. The complete mitochondrial genome of C. dauricum was sequenced and analyzed. The phylogenetic relationships between C. dauricum and other 10 species in the superfamily Chrysomeloidea were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that C. dauricum is closely related to Basilepta fulvipes in the same subfamily.

Project description:The sweet potato leaf beetle Colasposoma dauricum auripenne (Motschlsky, 1860) (Coleoptera: Chrysomelidae: Eumolpinae) is one of the main pests in sweet potato. In this study, the complete mitochondrial genome of Colasposoma dauricum auripenne (Motschlsky, 1860) (Coleoptera: Chrysomelidae: Eumolpinae) has been sequenced. The mitogenome length is 15,614 bp (GenBank: MW528208), which includes 13 typical insect protein-coding genes (PCGs), 2 ribosomal RNA genes (rRNAs), and 22 transfer RNA genes (tRNAs), and it contains 78.6% A + T and 21.4% G + C. All PCGs start with ATD (ATT, ATA, ATG) codon and stop with termination codon TAA, TAG and single T. The phylogenetic tree showed that Colasposoma had been cluster together, and form a sister group relationship with Pseudocolaspis. The complete mitogenome of C. dauricum auripenne would help understand Chrysomelidae evolution.

Mitochondrial genome of Chrysochares punctatus (Coleoptera: Chrysomelidae: Eumolpinae) and phylogenetic analysis.

Project description:Here, we determined the nearly complete mitochondrial genome (mitogenome) of Chrysochares punctatus (Coleoptera: Chrysomelidae: Eumolpinae), an important insect pest on Apocynum venetum in Northwestern China. This mitogenome was 14,451 bp long, encoding 13 protein-coding genes (PCGs), 21 transfer RNA genes (tRNAs), and 2 ribosomal RNA genes. The C. punctatus mitogenome presented an A + T content of 75.11%, with a positive AT-skew (0.064) and a negative GC-skew (-0.192). Ten PCGs started with a typical ATN codon, whereas the remaining three PCGs started with AAC (cox1) and TTG (nad1 and nad2). All tRNAs had a typical secondary cloverleaf structure, except for trnS1 which lacked the dihydrouridine arm. Bayesian phylogenetic analysis based on the nucleotide sequences of 13 PCGs recovered a phylogeny within Chrysomelidae: (((Chrysomelinae + Galerucinae), (((Eumolpinae, Lamprosomatinae), Cassidinae), Criocerinae)), Bruchinae).

Complete mitochondrial genome of Basilepta melanopus Lefevre, 1893 (coleoptera: chrysomelidae: eumolpinae), a tea pest from Southern China.

Project description:The tea pest, Basilepta melanopus Lefèvre 1893 (Chrysomelidae), belongs to the subfamily Eumolpinae. In this study, the complete mitochondrial genome sequence of B. melanopus from southern China was sequenced using the next-generation sequencing technique, assembled, and annotated using bioinformatics tools. The complete mitochondrial genome was 15,905 bp in length. The overall GC content was 22.51%, in which the percentages for the bases A, T, C, and G were 41.23%, 36.26%, 8.92%, and 13.59%, respectively. Thirty-seven genes were predicted, including 13 protein-coding, 22 transfer RNA, and two ribosomal RNA genes. Phylogenetic analysis based on the complete mitochondrial genome sequences of 18 Chrysomelidae taxa revealed that B. melanopus was closely related to Basilepta fulvipes.

The complete mitochondrial genome of invasive insect Leptinotarsa decemlineata Say 1824 (Coleoptera: Chrysomelidae).

Project description:Leptinotarsa decemlineata Say 1824, an invasive and globally devastating beetle, inflicts great damage to potato crops worldwide. The complete mitogenome of L. decemlineata is described in this study. It is a 16,741 bp long circular DNA molecule with a high A + T content of 76.9%, containing a typical 37 gene pattern. All PCGs (protein-coding genes) initiate with typical ATN codons. Most PCGs use TAN as a stop codon, whereas ND4 and COX3 use the incomplete codon TA as the stop codon. The lengths of rrnL and rrnS genes are 1,337 bp and 811 bp, respectively. All 22 tRNAs ranged from 62 to 77 bp. Phylogenetic analysis of Chrysomelidae indicated that L. decemlineata clusteres with three other Chrysomelinae species, which is consistent with previous analyses.

Project description:Aorianigripes (Baly, 1860) is one of the main pests of grapes, mainly damaging leaves, petioles and shoots and seriously affecting plant growth and development. Recently, this pest was found to damage the leaves of Ampelopsisgrossedentata, Ampelopsismegalophylla, Ampelopsischaffanjonii and Ampelopsiscantoniensis. However, the phylogenetic relationships of A.nigripes and other related family members are unclear. In this study, we sequenced and analysed the complete mitogenome of A.nigripes for the first time. The mitogenome of A.nigripes is circular and 17,306 bp in size, consisting of 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs) and two ribosomal RNA genes (rRNAs). The base composition of the A.nigripes mitogenome is 41.70% A, 33.76% T, 9.01% G and 15.53% C. The phylogenetic analysis showed that A.nigripes was clustered together with Basileptafulvipes and Colasposomadauricum.

The complete mitochondrial genome of Downesia tarsata (Coleoptera: Chrysomelidae: Cassidinae).

Project description:Downesia species are leaf-mining beetles mainly feed on Poaceae plants in the tropical and subtropical areas in Asia. In this study, we firstly sequenced and reported the complete mitochondrial genome for the genus. The complete mitogenome of Downesia tarsata is 18,557 bp in length, including 13 protein-coding genes (PCG), 22 transfer RNA (tRNA), two ribosomal RNA (rRNA), and one AT-rich region. Phylogenomic analysis indicated that D. tarsata is closely related to Agonita chinensis, and the two species belong to the same tribe of Gonophorini. The complete mitochondrial genome of D. tarsata could help clarify the phylogenetic relationship among Cassidinae species.

Project description:The complete mitogenome of Psylliodes balyi Jacoby (GenBank accession number MT644112) is 14,561 bp in length, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes, and a putative control region. The gene content and orientation of P. balyi were identical to other beetle mitogenomes. ATT, ATA, and ATG were initiation codons and TAA, TAG, and T were termination codons. All the 22 tRNAs have the typical cloverleaf secondary structure, except for trnS1 which lacked the dihydrouracil (DHU) arm. The phylogenetic relationship based on the neighbor-joining method showed that P. balyi is closely related to Agasicles hygrophila, which agrees with the conventional classification.

The complete mitochondrial genome of Monolepta hieroglyphica (Motschulsky) (Coleoptera: Chrysomelidae).

Project description:Monolepta hieroglyphica (Motschulsky) (Coleoptera: Chrysomelidae) is an important agricultural insect pest. In this study, the complete mitochondrial genome of M. hieroglyphica (GenBank accession number MW732714) was sequenced using Illumina HiSeq X Ten. The mitogenome was 16,213 bp long and comprised 13 protein-coding genes (PCGs), two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs) and a putative control region (CR). The nucleotide composition of the M. hieroglyphica mitochondrial genome was significantly biased (A, G, C and T accounted for 41.04%, 8.01%, 11.76% and 39.18%, respectively) with 80.23% A + T content. Two rRNAs were located between tRNA-Leu and the CR, separated by tRNA-Val. The CR, located between 12 s rRNA and tRNA-Ile, was 1,661 bp long. The length of the 22 tRNAs ranged from 61 to 71 bp. Phylogenetic analyses of 29 Chrysomelidae-Galerucinae species based on 13 mitochondrial protein-coding genes reconstructed using Bayesian 3.2.0 showed that the M. hieroglyphica mitogenome was clustered with the existing three different species of the Monolepta genus mitogenomes in a monophyletic manner. The M. hieroglyphica mitogenome provides an important data resource for further studies and contributes to our understanding of the phylogeny of this group.
