The complete mitochondrial genome of a medical important wasp, Vespa magnifica (Hymenoptera, Vespidae).

ABSTRACT: Vespa magnifica (Smith) is an aggressive social wasp species of Vespidae family. This species is of medical importance for its dangerous sting, traditional medicinal use and valuable venom components. Here, a complete mitogenome of V. magnifica was presented. It was 16,730 bp in length with nucleotide composition of AT: 79.4% and CG: 20.6%. In total, 13 protein-coding genes (PCGs), 22 transfer RNA, and two ribosomal RNA genes were annotated in this mitogenome. Phylogenetic analysis was performed using V. magnifica with 20 other species of Vespidae. The result indicated those species of genus Vespa fell into a paraphyletic group. Moreover, the Vespa species with large body size were clustered into a clade. This mitogenome resource can contribute to further phylogenetic and taxonomic study on genus Vespa.

Project description:We analyzed the complete mitochondrial genome of a social wasp, Vespa simillima simillima from South Korea prior to a systematic study on Korean Vespidae. The mitogenome is 16,740 bp in length, includes 13 protein-coding genes (PCGs), 22 tRNAs, 2 rRNAs, and a 228 bp short A + T-rich region. The overall base composition is 82.0% AT and 18.0% GC. The maximum-likelihood analysis suggested that V. s. simillima is closely related to V. bicolor, another species of Vespidae.

Complete mitochondrial genome of the Oriental Hornet, Vespa orientalis F. (Hymenoptera: Vespidae).

Project description:The Oriental Hornet (Vespa orientalis) is a social insect belonging to the Vespiade family (Wasps, Hornets, Yellowjackets), genus Vespa (true Hornets). The oriental hornet is a scavenger and an agricultural pest, especially to bee farmers, but is also recently described as a harvester of solar energy. Here, we report the mitochondrial genome sequence of the Oriental Hornet, Vespa orientalis F., which may play a vital role in understanding this wasp biology, light trapping and generation of electricity. The mitochondrial genome of this hornet is 16,099 bp in length, containing 13 protein-coding genes, 21 transfer RNA genes, and 2 ribosomal RNA genes. The overall base composition of the heavy-strand is 40.3% A, 5.9% C, 13.2% G, and 40.6% T, the percentages of A and T being higher than that of G and C. The mitochondrial genome of the Oriental Hornet, Vespa orientalis F. represents the first mitogenome of a solar energy harvesting insect.

Complete mitochondrial genome of the yellow-legged Asian hornet, Vespa velutina nigrithorax (Hymenoptera: Vespidae).

Project description:The yellow-legged Asian hornet, Vespa velutina nigrithorax, which originated from Asia, has invaded several countries, including South Korea. In Korea, V. velutina nigrithorax predation on honeybees is one of the most serious factors threatening apiculture. We sequenced the complete mitochondrial genome (mitogenome) of V. velutina to better understand the mitogenomic characteristics of this species. The 16,475 bp mitogenome of V. velutina consists of a typical set of genes, with an arrangement identical to that of congeneric species. Vespa velutina possesses the shortest A + T-rich region (132 bp) among congeneric Vespa, and this is also the shortest in the superfamily Vespoidea. Phylogenetic analysis using the 13 protein-coding genes of Vespoidea species indicated that each family forms strongly supported monophyletic groups (Bayesian posterior probability =1; ML, 100%). Moreover, V. velutina and V. bicolor form strongly supported sister groups (Bayesian posterior probability =1; ML, 100%).

Complete mitochondrial DNA sequence of the paper wasp Polistes riparius (Hymenoptera: Vespidae).

Project description:The paper wasp Polistes riparius is distributed in cold regions of northern East Asia to Russia. P. riparius are characterized by having longer cells than those of the closely related P. chinensis, which has a similar life history, as an adaptation to cold regions. The phylogenetic relationships of paper wasps have recently been studied; however, the genetic diversity and population structure of P. riparius has not been determined. The present study is the first to analyze the complete mitochondrial genome using next generation sequencing of P. riparius collected from Sapporo, Hokkaido Prefecture, Japan. The genome consisted of a closed loop that was 16,383 bp-long and included 13 protein coding genes (PCGs), 22 tRNA genes, two rRNA genes, and one AT-rich control region. The average AT content was 84.54%. The heavy (H)-strand was predicted to have 12 PCGs and 14 tRNA genes, while the light (L)-strand was predicted to contain one PCGs, eight tRNA genes, and two rRNA genes. All PCGs started with ATG. Stop codons were of two types: TAA for 11 genes (ND1, ND2, ND3, ND4L, ND5, ND6, COXI, COXII, COXIII, COB, ATP6 and ATP8) and TAG for two genes (ND3 and ND4). The molecular phylogenetic relationship based on the maximum likelihood method using 13 PCGs was consistent with some previous studies in which a closely relationship between P. riparius and P. jokahamae.

The mitochondrial genome of a social wasp, Vespula rufa (Linnaeus, 1758) (Hymenoptera: Vespidae).

Project description:For an ongoing systematic study of Korean Vespidae, we analyzed the complete mitochondrial genome of a social wasp, Vespula rufa (Linnaeus 1758), from the South Korea. The mitogenome is 17,521 bp in length, comprising 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, and two ribosomal RNA (rRNA) genes. The nucleotide composition is 40.5% adenines, 43.0% thymines, 6.0% guanines, and 10.5% cytosines. The GC content is 16.5%. A maximum-likelihood analysis was conducted to understand phylogenetic relationships, based on 13 complete mitogenome sequences of Vespinae species. We recognized that V. rufa is being placed basal within the genus Vespula. The complete mitochondrial genome of V. rufa provides useful genetic information for further studies.

The complete mitochondrial genome of Orancistrocerusaterrimusaterrimus and comparative analysis in the family Vespidae (Hymenoptera, Vespidae, Eumeninae).

Project description:To date, only one mitochondrial genome (mitogenome) in the Eumeninae has been reported in the world and this is the first report in China. The mitogenome of O.a.aterrimus is 17 972 bp long, and contains 38 genes, including 13 protein coding genes (PCGs), 23 tRNA genes, two rRNA genes, a long non-coding region (NCR), and a control region (CR). The mitogenome has 79.43% A + T content, its 13 PCGs use ATN as the initiation codon except for cox1 using TTG, and nine genes used complete translation termination TAA and four genes have incomplete stop codon T (cox2, cox3, nad4, and cytb). Twenty-two of 23 tRNAs can form the typical cloverleaf secondary structure except for trnS1. The CR is 1 078 bp long with 84.69% A+T content, comprising 28 bp tandem repeat sequences and 13 bp T-strech. There are two gene rearrangements which are an extra trnM2 located between trnQ and nad2 and the trnL2 in the upstream of nad1. Within all rearrangements of these mitogenomes reported in the family Vespidae, the translocation between trnS1 and trnE genes only appears in Vespinae, and the translocation of trnY in Polistinae and Vespinae. The absent codons of 13 PCGs in Polistinae are more than those both in Vespinae and Eumeninae in the family Vespidae. The study reports the complete mitogenome of O.a.aterrimus, compares the characteristics and construct phylogenetic relationships of the mitogenomes in the family Vespidae.

The complete mitochondrial genome of a forensic potential wasp, Vespa auraria (Smith).

Project description:Vespa auraria (Smith) is a common wasp species which is broadly distributed in south Asia. This species was spotted in filed forensic studies and disrupted the insect succession on the decaying corpse. Complete mitochondrial genome of V. auraria was presented here for its forensically potential influence. The mitogenome was assemblied to 18613 bp in length. The nucleotide composition of present mitogenome was calculated (A: 40.1%, G: 5.5%, T: 41.7%, C: 12.7%). Gene annotation analysis found 22 tRNA, 2 rRNA, and 13 protein-coding genes (PCGs). All the tRNA sequences could be folded into the typical structure of clover-leaf with the excepstion of tRNA-Ser (AGN). Phylogenetic development was analyzed based on V. auraria with other species of Hymenoptera.

Complete mitochondrial genome of a parasitic wasp Microplitis pallidipes (Hymenoptera: Braconidae: Microgastrinae).

Project description:The complete mitochondrial genome of Microplitis pallidipes contains 15931 bp, with an A + T content of 86.5% and consists of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a control region (GenBank accession no. MN892396). All of the 22 tRNA genes displayed an usual clover-leaf structure. Gene rearrangement events occurred in this species, there are eight tRNA genes changed their positions or/and directions. 13 PCGs started with ATN. Ten PCGs used the typical stop codon 'TAA' and 'TAG', three PCGs terminated with incomplete stop codons (TA). Phylogenetic analyses within the microgastroid complex were performed based on mitochondrial protein-coding genes.

Characterization of Complete Mitochondrial Genome and Phylogenetic Analysis of a Nocturnal Wasps-Provespa barthelemyi (Hymenoptera: Vespidae).

Project description:Genus Provespa contains nocturnal wasps mainly found in the southeastern region of Asia. There are no complete genome resources available of this genus, which hinders the study of its phylogenetic evolution and the origin of nocturnal behavior in the Vespidae family. Through high-throughput sequencing, we obtained the mitochondrial genome of Provespa barthelemyi (Hymenoptera: Vespidae), which is 17,721 base pairs in length and contains 13 protein-coding genes (PCGs), 22 tRNAs, and two rRNAs. We identified four gene rearrangement events of P. barthelemyi that frequently occur in the Vespidae family. We used Maximum Likelihood (ML) methodologies to construct a phylogenetic tree based on the sequenced mitochondrial genome and the available data of reported species belonging to Vespinae. Our findings confirmed the monophyly of Vespinae. Our study reports the first complete mitochondrial genome of Provespa and compares its characteristics with other mitochondrial genomes in the family Vespidae. This research should shed light on the phylogenetic relationships and ecological characteristics of the Vespidae family.

The invasion, provenance and diversity of Vespa velutina Lepeletier (Hymenoptera: Vespidae) in Great Britain.

Project description:The yellow-legged or Asian hornet (Vespa velutina colour form nigrithorax) was introduced into France from China over a decade ago. Vespa velutina has since spread rapidly across Europe, facilitated by suitable climatic conditions and the ability of a single nest to disperse many mated queens over a large area. Yellow-legged hornets are a major concern because of the potential impact they have on populations of many beneficial pollinators, most notably the western honey bee (Apis mellifera), which shows no effective defensive behaviours against this exotic predator. Here, we present the first report of this species in Great Britain. Actively foraging hornets were detected at two locations, the first around a single nest in Gloucestershire, and the second a single hornet trapped 54 km away in Somerset. The foraging activity observed in Gloucestershire was largely restricted to within 700 m of a single nest, suggesting highly localised movements. Genetic analyses of individuals from the Gloucestershire nest and the single hornet from Somerset suggest that these incursions represent an expansion of the European population, rather than a second incursion from Asia. The founding queen of the Gloucestershire nest mated with a single male, suggesting that sexual reproduction may have occurred in an area of low nest density. Whilst the nest contained diploid adult males, haploid 'true' males were only present at the egg stage, indicating that the nest was detected and removed before the production of queens. Members of the public reported additional dead hornets associated with camping equipment recently returned from France and imported timber products, highlighting possible pathways of incursion. The utility of microsatellites to inform surveillance during an incursion and the challenge of achieving eradication of this damaging pest are discussed.
