ABSTRACT: Sea surface temperature (SST) is crucial for understanding the physical characteristics and ecosystems of coastal seas. SST varies near the tidal flat, where exposure and flood recur according to the tidal cycle. However, the variability of SST near the tidal flat is poorly understood owing to difficulties in making in-situ observations. The high resolution of Landsat 8 enabled us to determine the variability of SST near the macro tidal flat. The spatial distribution of the SST extracted from Landsat 8 changed drastically. The seasonal SST range was higher near the tidal flat than in the open sea. The maximum seasonal range of coastal SST exceeded 23 °C, whereas the range in the open ocean was approximately 18 °C. The minimum and maximum horizontal SST gradients near the tidal flat were approximately - 0.76 °C/10 km in December and 1.31 °C/10 km in June, respectively. The heating of sea water by tidal flats in spring and summer, and cooling in the fall and winter might result in a large horizontal SST gradient. The estimated heat flux from the tidal flat to the seawater based on the SST distribution shows seasonal change ranging from - 4.85 to 6.72 W/m².

Project description:Coastal zones with dense populations, low elevations and/or inadequate adaptive capacity are on the frontline of unprecedented impacts from climate change. The Gulf of Guinea (GoG), stretching from Liberia to Gabon, is in particular vulnerable to coastal flooding caused by local and/or climate-induced sea level rise. In this region, interannual to decadal coastal sea level changes remain poorly understood, mainly due to a lack of tide gauge stations. Here we use nearly three decades (1993-2021) of satellite altimetry data to study the link between the Equatorial Atlantic and coastal GoG sea level variability. The rate of mean sea level rise increased from 3.47 to 3.89 ± 0.10 mm/yr from the Equatorial oceanic domain to the GoG coastal area, with an acceleration of 0.094 ± 0.050 mm/yr2. This corresponds to a mean sea level rise of about 8.9 cm over the entire altimetry period, 1993-2021. We focus on the (extreme) warm/cold events that occur in both the GoG during Atlantic Niños, and along the Angola-Namibia coast during Benguela Niños. Both events are driven by remote forcing via equatorial Kelvin waves and local forcing by local winds, freshwater fluxes and currents intensifications. Analysis of altimetry-based sea level, sea surface temperature anomalies, 20 °C isotherm based PIRATA moorings, and the Argo-based steric and thermometric sea level allows us to follow the coastal trapped waves (CTWs) along the GoG, and its link with major events observed along the strong Equatorial Atlantic warmings in 2010, 2012, 2019 and 2021. Both 2019 and 2021 warming have been identified as the warmest event ever reported in this region during the last 40 years. A lag of 1 month is observed between equatorial and West African coastal trapped wave propagation. This observation may help to better anticipate and manage the effects of extreme events on local ecosystems, fisheries, and socio-economic activities along the affected coastlines. In order to enable informed decision-making and guarantee the resilience of coastal communities in the face of climate change, it emphasises the significance of ongoing study in this field.

Project description:BackgroundAlkaline phosphatase (AP) catalyzes the hydrolytic cleavage of phosphate monoesters under alkaline conditions and plays important roles in microbial ecology and molecular biology applications. Here, we report on the first isolation and biochemical characterization of a thermolabile AP from a metagenome.ResultsThe gene encoding a novel AP was isolated from a metagenomic library constructed with ocean-tidal flat sediments from the west coast of Korea. The metagenome-derived AP (mAP) gene composed of 1,824 nucleotides encodes a polypeptide with a calculated molecular mass of 64 kDa. The deduced amino acid sequence of mAP showed a high degree of similarity to other members of the AP family. Phylogenetic analysis revealed that the mAP is shown to be a member of a recently identified family of PhoX that is distinct from the well-studied classical PhoA family. When the open reading frame encoding mAP was cloned and expressed in recombinant Escherichia coli, the mature mAP was secreted to the periplasm and lacks an 81-amino-acid N-terminal Tat signal peptide. Mature mAP was purified to homogeneity as a monomeric enzyme with a molecular mass of 56 kDa. The purified mAP displayed typical features of a psychrophilic enzyme: high catalytic activity at low temperature and a remarkable thermal instability. The optimal temperature for the enzymatic activity of mAP was 37°C and complete thermal inactivation of the enzyme was observed at 65°C within 15 min. mAP was activated by Ca(2+) and exhibited maximal activity at pH 9.0. Except for phytic acid and glucose 1-phosphate, mAP showed phosphatase activity against various phosphorylated substrates indicating that it had low substrate specificity. In addition, the mAP was able to remove terminal phosphates from cohesive and blunt ends of linearized plasmid DNA, exhibiting comparable efficiency to commercially available APs that have been used in molecular biology.ConclusionsThe presented mAP enzyme is the first thermolabile AP found in cold-adapted marine metagenomes and may be useful for efficient dephosphorylation of linearized DNA.

Project description:A method to assess global land surface water (fw) inundation dynamics was developed by exploiting the enhanced fw sensitivity of L-band (1.4 GHz) passive microwave observations from the Soil Moisture Active Passive (SMAP) mission. The L-band fw (fwLBand ) retrievals were derived using SMAP H-polarization brightness temperature (Tb ) observations and predefined L-band reference microwave emissivities for water and land endmembers. Potential soil moisture and vegetation contributions to the microwave signal were represented from overlapping higher frequency Tb observations from AMSR2. The resulting fwLBand global record has high temporal sampling (1-3 days) and 36-km spatial resolution. The fwLBand annual averages corresponded favourably (R=0.85, p-value<0.001) with a 250-m resolution static global water map (MOD44W) aggregated at the same spatial scale, while capturing significant inundation variations worldwide. The monthly fwLBand averages also showed seasonal inundation changes consistent with river discharge records within six major US river basins. An uncertainty analysis indicated generally reliable fwLBand performance for major land cover areas and under low to moderate vegetation cover, but with lower accuracy for detecting water bodies covered by dense vegetation. Finer resolution (30-m) fwLBand results were obtained for three sub-regions in North America using an empirical downscaling approach and ancillary global Water Occurrence Dataset (WOD) derived from the historical Landsat record. The resulting 30-m fwLBand retrievals showed favourable spatial accuracy for water (commission error 31.46%, omission error 30.20%) and land (commission error 0.87%, omission error 0.96%) classifications and seasonal wet and dry periods when compared to independent water maps derived from Landsat-8 imagery. The new fwLBand algorithms and continuing SMAP and AMSR2 operations provide for near real-time, multi-scale monitoring of global surface water inundation dynamics and potential flood risk.
