Antibacterial properties of poly (N,N-dimethylaminoethyl methacrylate) obtained at different initiator concentrations in solution polymerization.

ABSTRACT: The samples of poly(N,N-dimethylaminoethyl methacrylate) were synthesized by radical polymerization. The amount of monomer and solvent was constant as opposed to an amount of initiator which was changing. No clear relationship between polymerization conditions and the molecular weight of the polymer was found, probably due to the branched configuration of produced polymer. Bactericidal interactions in all samples against Gram-positive and Gram-negative bacteria have been demonstrated. However, the observed effect has various intensities, depending on the type of bacteria and the type of sample.

Thermal properties of poly(N,N-dimethylaminoethyl methacrylate).

Project description:Poly(N,N-dimethylaminoethyl methacrylate) (PDMAEMA) is a promising quite new polymer with very interesting properties. The thermal degradation process of PDMAEMA was investigated. The polymer was heated at specific time intervals, then heating was stopped, and infrared analysis was performed to obtain information on the structure of the solid residue. The thermal degradation process has a two-stage character. The limit temperature for the first decomposition step was about 390°C, after which the second stage of sample decomposition began. The order of disintegration of the macromolecules was determined. Activation energy values for the thermal decomposition process have been calculated; they are 89.8 kJ/mol for the first stage and 17.7 kJ/mol for the second stage of the degradation process.

Nanolayers of Poly(N,N'-Dimethylaminoethyl Methacrylate) with a Star Topology and Their Antibacterial Activity.

Project description:In this paper, we focus on the synthesis and characterization of novel stable nanolayers made of star methacrylate polymers. The effect of nanolayer modification on its antibacterial properties was also studied. A covalent immobilization of star poly(N,N'-dimethylaminoethyl methacrylate) (PDMAEMA) to benzophenone functionalized glass or silicon supports was carried out via a "grafting to" approach using UV irradiation. To date, star polymer UV immobilization has never been used for this purpose. The thickness of the resulting nanolayers increased from 30 to 120 nm with the molar mass of the immobilized stars. The successful bonding of star PDMAEMA to the supports was confirmed by surface sensitive quantitative spectroscopic methods. Next, amino groups in the polymer layer were quaternized with bromoethane, and the influence of this modification on the antibacterial properties of the obtained materials was analyzed using a selected reference strain of bacteria. The resulting star nanolayer surfaces exhibited higher antimicrobial activity against Bacillus subtilis ATCC 6633 compared to that of the linear PDMAEMA analogues grafted onto a support. These promising results and the knowledge about the influence of the topology and modification of PDMAEMA layers on their properties may help in searching for new materials for antimicrobial applications in medicine.

Quaternized Poly(N,N′-dimethylaminoethyl methacrylate) Star Nanostructures in the Solution and on the Surface

Project description:Antibacterial polymeric materials are promising in the fight against resistant bacteria strains. Amongst them, cationic macromolecules with quaternary ammonium groups are one of intensively studied, as they interact with the bacterial membranes causing cell death. In this work, we propose to use nanostructures composed of polycations with star topology for the preparation of antibacterial materials. First, star polymers of N,N′-dimethylaminoethyl methacrylate and hydroxyl-bearing oligo(ethylene glycol) methacrylate P(DMAEMA-co-OEGMA-OH) were quaternized with various bromoalkanes and their solution behavior was studied. It was shown that in water two modes of star nanoparticles were observed, of diameters about 30 nm and up to 125 nm, independently of the quaternizing agent. Separately layers of P(DMAEMA-co-OEGMA-OH) stars were obtained. In this case, the chemical grafting of polymers to the silicon wafers modified with imidazole derivatives was applied, followed by the quaternization of the amino groups of polycations. A comparison of the quaternary reaction in solution and on the surface showed that in the solution it is influenced by the alkyl chain length of the quaternary agent, while on the surface such relationship is not observed. After physico-chemical characterization of the obtained nanolayers, their biocidal activity was tested against two strains of bacteria E. coli and B. subtilis. The best antibacterial properties exhibited layers quaternized with shorter alkyl bromide, where 100% growth inhibition of E. coli and B. subtilis after 24 h of contact was observed.

Thermoresponsive Poly(N-Isopropylacrylamide-co-Dimethylaminoethyl Methacrylate) Microgel Aqueous Dispersions with Potential Antimicrobial Properties.

Project description:The work herein describes the preparation of thermoresponsive microgels with potential antimicrobial properties. Most of the work performed so far regarding microgels with antimicrobial activity, deals with the ability of microgels to carry and release antibiotics or antimicrobial agents (antimicrobial peptides). The originality of this work lies in the possibility of developing intrinsic antimicrobial microgels by copolymerization of the well-known thermoresponsive monomer, N-isopropylacrylamide (NIPAM) with dimethylaminoethyl methacrylate (DMAEMA), a water-soluble monomer, to form microgels via precipitation polymerization (radical polymerization). Due to the presence of a tertiary amine in the DMAEMA comonomer, microgels can be modified by N-alkylation reaction with methyl and butyl iodide. This quaternization confers positive charges to the microgel surfaces and thus the potential antimicrobial activity. The effect of DMAEMA content and its quaternization with both, methyl and butyl iodide is evaluated in terms of thermal and surface charge properties, as well as in the microgel size and viscoelastic behavior. Finally, a preliminary study of the antimicrobial activity against different microorganisms is also performed in terms of minimum inhibitory concentration (MIC). From this study we determined that in contrast with butylated microgels, methylated ones show potential antimicrobial activity and good physical properties besides of maintaining microgel thermo-responsiveness.

Synthesis and Postpolymerization Modification of Fluorine-End-Labeled Poly(Pentafluorophenyl Methacrylate) Obtained via RAFT Polymerization.

Project description:Chain-end-labeled polymers are interesting for a range of applications. In polymer nanomedicine, chain-end-labeled polymers are useful to study and help understand cellular internalization and intracellular trafficking processes. The recent advent of fluorescent label-free techniques, such as nanoscale secondary ion mass spectrometry (NanoSIMS), provides access to high-resolution intracellular mapping that can complement information obtained using fluorescent-labeled materials and confocal microscopy and flow cytometry. Using poly(N-(2-hydroxypropyl)methacrylamide) (PHPMA) as a prototypical polymer nanomedicine, this paper presents a synthetic strategy to polymers that contain trace element labels, such as fluorine, which can be used for NanoSIMS analysis. The strategy presented in this paper is based on reversible addition fragmentation chain transfer (RAFT) polymerization of pentafluorophenyl methacrylate (PFMA) mediated by two novel chain-transfer agents (CTAs), which contain either one (α) or two (α,ω) fluorine labels. In the first part of this study, via a number of polymerization experiments, the polymerization properties of the fluorinated RAFT CTAs were established. 19F NMR spectroscopy revealed that these fluorinated RAFT agents possess unique spectral signatures, which allow to directly monitor RAFT agent conversion and measure end-group fidelity. Comparison with 4-cyanopentanoic acid dithiobenzoate, which is a standard CTA for the RAFT polymerization of PFMA, revealed that the introduction of one or two fluorine labels does not significantly affect the polymerization properties of the CTA. In the last part of this paper, a proof-of-concept study is presented that demonstrates the feasibility of the fluorine-labeled poly(pentafluorophenyl methacrylate) polymers as platforms for the postpolymerization modification to generate PHPMA-based polymer nanomedicines.

Synthesis and Aqueous Solution Properties of an Amino Bisphosphonate Methacrylate Homopolymer via RAFT Polymerization.

Project description:The present contribution reports on the synthesis via reversible addition-fragmentation chain transfer (RAFT) polymerization of a methacrylate derivative bearing an aminobisphosponate moiety as a pendant group, namely, ethyl N,N-tetramethylbis(phosphonate)-bis(methylene) amine methacrylate (MAC₂NP₂). The polymerization was performed by the use of cyanoisopropyl dithiobenzoate as chain transfer agent at 70 °C in various solvents with different polarities including N,N-dimethylformamide, acetonitrile, tetrahydrofuran, and in bulk. Best results were obtained in N,N-dimethylformamide where higher conversions and polymerization rates were noticed. The successful hydrolysis of the phosphonate ester groups was performed using bromotrimethylsilane with excellent yields leading to the formation of highly water soluble and pH-responsive polymers. Finally, a preliminary solution behavior study was carried out by investigating the aqueous solution properties of synthesized amino bisphosphonate methacrylate homopolymers and their phosphonic acid analogs via potentiometric titration and zeta potential measurements.

Superhydrophilic Coating with Antibacterial and Oil-Repellent Properties via NaIO4-Triggered Polydopamine/Sulfobetaine Methacrylate Polymerization.

Project description:Superhydrophilic coatings have been widely used for the surface modification of membranes or biomedical devices owing to their excellent antifouling properties. However, simplifying the modification processes of such materials remains challenging. In this study, we developed a simple and rapid one-step co-deposition process using an oxidant trigger to fabricate superhydrophilic surfaces based on dopamine chemistry with sulfobetaine methacrylate (SBMA). We studied the effect of different oxidants and SBMA concentrations on surface modification in detail using UV-VIS spectrophotometry, dynamic light scattering, atomic force microscopy, X-ray photoelectron spectroscopy, and surface plasmon resonance. We found that NaIO4 could trigger the rate of polymerization and the optimum ratio of dopamine to SBMA is 1:25 by weight. This makes the surface superhydrophilic (water contact angle < 10°) and antifouling. The superhydrophilic coating, when introduced to polyester membranes, showed great potential for oil/water separation. Our study provides a complete description of the simple and fast preparation of superhydrophilic coatings for surface modification based on mussel-inspired chemistry.

All-Atom Molecular Dynamics Simulations of Grafted Poly(N,N-dimethylaminoethyl methacrylate) Brushes.

Project description:Modeling polymer brushes is essential for understanding their complex behavior at surfaces and interfaces, enabling the design of materials with tunable properties. We present a computational protocol to model polymer brushes composed of grafted, brush-like chains of the charged polymer poly(N,N-dimethylaminoethyl methacrylate) (p(DMAEMA)) using an all-atom representation that captures detailed molecular interactions and structural properties. The approach is flexible and non-grid-based and allows for randomized strand configurations and the incorporation of periodic boundary conditions, enabling the construction of asymmetric polymer brush setups. An atactic p(DMAEMA) configuration is demonstrated as an example, though the protocol can be readily adapted to construct other brush-like polymer systems with varying tacticities or compositions, depending on the pH environment. Furthermore, this can be extended to stimuli-responsive materials, which generate conformation or charge upon changes in pH value or other external triggers. Molecular dynamics simulations are then employed to gain insights into the conformational behavior of the grafted p(DMAEMA) brushes and their surrounding aqueous environment, as well as their response to temperature, protonation, and variations in grafting densities, in terms of the solvent-accessible surface area, radius of gyration, and radial distribution functions. This versatile protocol provides a robust tool for simulating and analyzing the properties of diverse polyelectrolyte polymer brush systems and also as composite materials.

Evolution of Physical, Thermal, and Mechanical Properties of Poly(methyl Methacrylate)-Based Elium Thermoplastic Polymer During Polymerization.

Project description:Elium-based thermoplastic composites are a key material for future use in the marine, wind energy, and automotive industries because of their recyclability and ease of manufacture. To optimize the processing of the Elium composites to yield optimal structural properties, computational process modeling can be used to relate processing parameters to residual stresses and material durability. The key ingredient for reliable and accurate process modeling is the evolution of physical, thermal, and mechanical properties during polymerization. The objective of this study is to use molecular dynamics to predict the mass density, bulk modulus, shear modulus, Young's modulus, Poisson's ratio, glass transition temperature, and coefficient of thermal expansion as a function of the extent of reaction of the polymer. The predicted properties compare favorably to the experimentally measured values in the fully polymerized state. This data set of properties provides needed input data for process modeling of Elium-based composites for process parameter optimization and improved durability and performance.

Dual pH- and temperature-responsive poly(dimethylaminoethyl methacrylate)-coated mesoporous silica nanoparticles as a smart drug delivery system.

Project description:A robust drug delivery system was created by grafting poly(dimethylaminoethyl methacrylate) (PDMAEMA) onto silica nanoparticles with two different lengths using an in situ atom transfer radical polymerization, resulting in the formation of a pH- and temperature-sensitive shell. The high molecular weight PDMAEMA demonstrated effective controlled drug release, and prevented drug release in healthy cells. Drug release occurred through polymer shell protonation at pH 5. The critical temperature of 41 °C facilitated rapid solvation of the shell polymers in the blood, preventing tissue accumulation and reducing toxicity compared to systems with lower critical solution temperatures. Field-emission scanning electron microscopy analysis and nitrogen adsorption/desorption analysis showed that the nanoparticles have a fine network, mesoporous structure, and a mean size of around 17 nm that show their excellent capacity for loading drugs. Fourier-transform infrared spectroscopy showed that all the modification steps and polymerization were successfully implemented. Thermogravimetric analysis showed PDMAEMA chains with two different lengths grafted onto the nanoparticles. Transmission electron microscopy analysis also showed grafted polymer chains on the hybrid nanoparticles. The release profile of model cancer drugs (doxorubicin and methotrexate) varied with pH and temperature, with high molecular weight PDMAEMA shells effectively preventing drug release at neutral pH. In vitro analysis using the HeLa cell line showed minimal toxicity in blank samples and significant release profile in acidic environment.
