Functional Investigation of Plant Growth Promoting Rhizobacterial Communities in Sugarcane.

ABSTRACT: Plant microbiota are of great importance for host nutrition and health. As a C₄ plant species with a high carbon fixation capacity, sugarcane also associates with beneficial microbes, though mechanisms underlying sugarcane root-associated community development remain unclear. Here, we identify microbes that are specifically enriched around sugarcane roots and report results of functional testing of potentially beneficial microbes propagating with sugarcane plants. First, we analyzed recruitment of microbes through analysis of 16S rDNA enrichment in greenhouse cultured sugarcane seedlings growing in field soil. Then, plant-associated microbes were isolated and assayed for beneficial activity, first in greenhouse experiments, followed by field trials for selected microbial strains. The promising beneficial microbe SRB-109, which quickly colonized both roots and shoots of sugarcane plants, significantly promoted sugarcane growth in field trials, nitrogen and potassium acquisition increasing by 35.68 and 28.35%, respectively. Taken together, this report demonstrates successful identification and utilization of beneficial plant-associated microbes in sugarcane production. Further development might facilitate incorporation of such growth-promoting microbial applications in large-scale sugarcane production, which may not only increase yields but also reduce fertilizer costs and runoff.

Project description:Maize (Zea mays L.), a key staple crop in Sub-Saharan Africa, is particularly vulnerable to concurrent drought and heat stress, which threatens crop yield and food security. Plant growth-promoting rhizobacteria (PGPR) have shown potential as biofertilizers to enhance plant resilience under such abiotic stresses. This study aimed to (1) identify PGPR isolates tolerant to drought and heat, (2) assess their capacity to mitigate the effects of these stresses on early maize growth, and (3) analyze maize gene expression changes associated with PGPR-induced tolerance. Rhizobacteria were isolated and screened for drought and heat tolerance, alongside key plant growth-promoting (PGP) traits, including phosphorus solubilization, nitrogen fixation, and indole acetic acid production. In vitro and pot trials evaluated the effects of selected isolates on maize growth under stress, using indicators such as shoot length, root and shoot biomass (wet and dry), and leaf water content. Quantitative reverse transcription PCR (qRT-PCR) was employed to profile maize stress response genes. The identified PGPR isolates included Bacillus cereus (11MN1), Bacillus pseudomycoides (21MN1B), Lelliottia amnigena (33MP1), and Leclercia adecarboxylata (36MP8). Greenhouse trials demonstrated that L. amnigena 33MP1, L. adecarboxylata 36MP8, and a mixed culture of isolates (11MN1, 21MN1B, 33MP1, 36MP8) effectively alleviated the adverse effects of concurrent drought and heat stress in maize. Notably, qRT-PCR analysis indicated that PGPR-induced tolerance may involve the modulation of stress response genes CAT2 (catalase 2) and DHN2 (dehydrin 2), which play roles in oxidative stress management and cellular protection. The PGPR isolates identified in this study represent promising bioinoculants for enhancing maize resilience under climate-induced stresses, offering a sustainable approach to improve maize productivity, conserve water, and reduce irrigation needs in drought-prone regions.

Project description:Efficient screening method is the prerequisite for getting plant growth-promoting (PGP) rhizobacteria (PGPR) which may play an important role in sustainable agriculture from the natural environment. Many current traditional preliminary screening criteria based on knowledge of PGP mechanisms do not always work well due to complex plant-microbe interactions and may lead to the low screening efficiency. More new screening criteria should be evaluated to establish a more effective screening system. However, the studies focused on this issue were not enough, and few new screening criteria had been proposed. The aim of this study was to analyze the correlation between the metabolic phenotypes of rhizobacterial isolates and their PGP ability. The feasibility of using these phenotypes as preliminary screening criteria for PGPR was also evaluated. Twenty-one rhizobacterial isolates were screened for their PGP ability, traditional PGP traits, and multiple metabolic phenotypes that are not directly related to PGP mechanisms, but are possibly related to rhizosphere colonization. Correlations between the PGP traits or metabolic phenotypes and increases in plant agronomic parameters were analyzed to find the indicators that are most closely related to PGP ability. The utilization of 11 nutrient substrates commonly found in root exudates, such as D-salicin, β-methyl-D-glucoside, and D-cellobiose, was significantly positively correlated with the PGP ability of the rhizobacterial isolates. The utilization of one amino acid and two organic acids, namely L-aspartic acid, α-keto-glutaric acid, and formic acid, was negatively correlated with PGP ability. There were no significant correlations between four PGP traits tested in this study and the PGP ability. The ability of rhizobacterial isolates to metabolize nutrient substrates that are identical or similar to root exudate components may act as better criteria than PGP traits for the primary screening of PGPR, because rhizosphere colonization is a prerequisite for PGPR to affect plants.
