Photocatalytic defluoroalkylation and hydrodefluorination of trifluoromethyls using o-phosphinophenolate.

ABSTRACT: Under visible light irradiation, o-phosphinophenolate functions as an easily accessible photoredox catalyst to activate trifluoromethyl groups in trifluoroacetamides, trifluoroacetates, and trifluoromethyl (hetero)arenes to deliver corresponding difluoromethyl radicals. It works in relay with a thiol hydrogen atom transfer (HAT) catalyst to enable selective defluoroalkylation and hydrodefluorination. The reaction allows for the facile synthesis of a broad scope of difluoromethylene-incorporated carbonyl and (hetero)aromatic compounds, which are valuable fluorinated intermediates of interest in the pharmaceutical industry. The ortho-diphenylphosphino substituent, which is believed to facilitate photoinduced electron transfer, plays an essential role in the redox reactivity of phenolate. In addition to trifluoromethyl groups, pentafluoroethyl groups could also be selectively defluoroalkylated.

Hydrogen Bond Directed Photocatalytic Hydrodefluorination: Overcoming Electronic Control.

Project description:The photocatalytic C-F functionalization of highly fluorinated arenes is a powerful method for accessing functionalized multifluorinated arenes. The decisive step in the determining regioselectivity in fluorine functionalization is fluoride fragmentation from the radical anion of the multifluorinated arene. To date, the availability of regioisomers has been dictated by the innate electronics of the fluorinated arene, limiting the synthetic utility of the chemistry. This study investigates the remarkable ability of a strategically located hydrogen bond to transcend the normal regioselectivity of the C-F functionalization event. A significant rate acceleration is additionally observed for hydrodefluorination of fluorines that can undergo intramolecular hydrogen bonds that form 5-8-membered cycles with moderately acidic N-H's. The hydrogen bond is expected to facilitate the fragmentation not only by bending the C-F bond of the radical anion out of planarity but also by increasing the exothermicity of the fluoride extrusion step through protonation of the naked fluoride. Finally, the synthetic utility of the method is demonstrated in an expedited synthesis of the trifluorinated α-phenyl acetic acid derivative required for the commercial synthesis of Januvia, an antidiabetic drug. This represents the first synthesis of a commercially important multifluorinated arene via a defluorination strategy and is significantly shorter than the current strategy.

Molecular Sculpting: A Multipurpose Tool for Expedited Access to Various Fluorinated Arenes via Photocatalytic Hydrodefluorination of Benzoates.

Project description:Starting with highly fluorinated benzoates, we develop the directed photocatalytic hydrodefluorination (HDF) of fluorinated aryl benzoates and demonstrate its synergistic use with other HDF strategies, along with C-H arylation, decarboxylative coupling, and decarboxylative protonation, to access most fluorination patterns found in benzoate derivatives and by extension benzene derivatives via a molecular sculpting approach. Mild reaction conditions and excellent regioselectivity make the approach ideal for synthesis. This approach provides access to 16 benzoate derivatives with different fluorination patterns from just a couple of highly fluorinated, commercially available benzoic acids. We synthesize key intermediates or the active pharmaceutical ingredient for sitagliptin, diflunisal, and other pharmaceutically important molecules. Importantly, we provide key insights into relative rates of defluorination and strategies to alter these rates. We provide demonstrations of the synergistic use of HDF and related technologies to rapidly enhance the synthetic complexity of these simple commercially available perfluoroarenes to form complex partially fluorinated molecules.

Direct electrochemical hydrodefluorination of trifluoromethylketones enabled by non-protic conditions.

Project description:CF2H groups are unique due to the combination of their lipophilic and hydrogen bonding properties. The strength of H-bonding is determined by the group to which it is appended. Several functional groups have been explored in this context including O, S, SO and SO2 to tune the intermolecular interaction. Difluoromethyl ketones are under-studied in this context, without a broadly accessible method for their preparation. Herein, we describe the development of an electrochemical hydrodefluorination of readily accessible trifluoromethylketones. The single-step reaction at deeply reductive potentials is uniquely amenable to challenging electron-rich substrates and reductively sensitive functionality. Key to this success is the use of non-protic conditions enabled by an ammonium salt that serves as a reductively stable, masked proton source. Analysis of their H-bonding has revealed difluoromethyl ketones to be potentially highly useful dual H-bond donor/acceptor moieties.

Catalyst-Free Hydrodefluorination of Perfluoroarenes with NaBH<sub>4</sub>.

Project description:Presented is an economical means of removing fluorine from various highly fluorinated arenes using NaBH4. The procedure was adapted for different classes of perfluoroarenes. A novel isomer of an emerging class of organic dyes based on the carbazole phthalonitrile motif was succinctly synthesized in two steps from tetrafluorophthalonitrile, demonstrating the utility of the hydrodefluorination procedure. Initial exploration of the dye shows it to be photoactive and capable of facilitating contrathermodynamic styrenoid E/Z isomerization.

Organophotoredox Hydrodefluorination of Trifluoromethylarenes with Translational Applicability to Drug Discovery.

Project description:Molecular editing such as insertion, deletion, and single atom exchange in highly functionalized compounds is an aspirational goal for all chemists. Here, we disclose a photoredox protocol for the replacement of a single fluorine atom with hydrogen in electron-deficient trifluoromethylarenes including complex drug molecules. A robustness screening experiment shows that this reductive defluorination tolerates a range of functional groups and heterocycles commonly found in bioactive molecules. Preliminary studies allude to a catalytic cycle whereby the excited state of the organophotocatalyst is reductively quenched by the hydrogen atom donor, and returned in its original oxidation state by the trifluoromethylarene.

Copper-catalyzed highly switchable defluoroborylation and hydrodefluorination of 1-(trifluoromethyl)alkynes.

Project description:CF2-containing compounds hold significant potential in drug discovery, organic synthesis, and materials science. However, synthesizing various CF2-containing building blocks from a single compound remains challenging. Here, we present a Cu-catalyzed, switchable defluoroborylation and hydrodefluorination of trifluoromethylated alkynes, yielding four types of CF2-containing compounds. The chemo- and regio-selective sp2/sp3 1,2-diborylation and sp2 monoborylation of 1-(trifluoromethyl)alkynes are controlled by adjusting the solvent and ligand quantity. Additionally, altering the base allows selective generation of gem-difluoroalkenes or difluoromethylalkenes. Notably, our method prevents over-defluorination of the CF3 group on unsaturated C-C bonds during nucleophilic additions, preserving the pharmaceutically valuable CF2 group. Experimental data and density functional theory (DFT) calculations elucidate the regioselectivities of Cu-Bpin addition and the regulatory role of the ligand in selective deborylation processes.

Activation of pentafluoropropane isomers at a nanoscopic aluminum chlorofluoride: hydrodefluorination versus dehydrofluorination.

Project description:The hydrofluorocarbon 245 isomers, 1,1,1,3,3-pentafluoropropane, 1,1,1,2,2- pentafluoropropane, and 1,1,1,2,3-pentafluoropropane (HFC-245fa, HFC-245cb, and HFC-245eb) were activated through C-F bond activations using aluminium chlorofluoride (ACF) as a catalyst. The addition of the hydrogen source Et3SiH is necessary for the activation of the secondary and tertiary C-F bonds. Multiple C-F bond activations such as hydrodefluorinations and dehydrofluorinations were observed, followed by hydroarylation and Friedel-Crafts-type reactions under mild conditions.

Photocatalytic cyclohexane oxidation and epoxidation using hedgehog particles.

Project description:Inorganic particles are effective photocatalysts for the liquid-state production of organic precursors and monomers at ambient conditions. However, poor colloidal stability of inorganic micro- and nanoparticles in low-polarity solvents limits their utilization as heterogeneous catalysts and coating them with surfactants drastically reduces their catalytic activity. Here we show that effective photo-oxidation of liquid cyclohexane (CH) is possible using spiky particles from metal oxides with hierarchical structure combining micro- and nanoscale structural features engineered for enhanced dispersibility in CH. Nanoscale ZnO spikes are assembled radially on α-Fe2O3 microcube cores to produce complex 'hedgehog' particles (HPs). The 'halo' of stiff spikes reduces van der Waals attraction, preventing aggregation of the catalytic particles. Photocatalysis in Pickering emulsions formed by HPs with hydrogen peroxide provides a viable pathway to energy-efficient alkane oxidation in the liquid state. Additionally, HPs enable a direct chemical pathway from alkanes to epoxides at ambient conditions, specifically to cyclohexene oxide, indicating that the structure of HPs has a direct effect on the recombination of ion-radicals during the hydrocarbon oxidation. These findings demonstrate the potential of inorganic photocatalysts with complex architecture for 'green' catalysis.

Photocatalytic Air Purification Using Functional Polymeric Carbon Nitrides.

Project description:The techniques for the production of the environment have received attention because of the increasing air pollution, which results in a negative impact on the living environment of mankind. Over the decades, burgeoning interest in polymeric carbon nitride (PCN) based photocatalysts for heterogeneous catalysis of air pollutants has been witnessed, which is improved by harvesting visible light, layered/defective structures, functional groups, suitable/adjustable band positions, and existing Lewis basic sites. PCN-based photocatalytic air purification can reduce the negative impacts of the emission of air pollutants and convert the undesirable and harmful materials into value-added or nontoxic, or low-toxic chemicals. However, based on previous reports, the systematic summary and analysis of PCN-based photocatalysts in the catalytic elimination of air pollutants have not been reported. The research progress of functional PCN-based composite materials as photocatalysts for the removal of air pollutants is reviewed here. The working mechanisms of each enhancement modification are elucidated and discussed on structures (nanostructure, molecular structue, and composite) regarding their effects on light-absorption/utilization, reactant adsorption, intermediate/product desorption, charge kinetics, and reactive oxygen species production. Perspectives related to further challenges and directions as well as design strategies of PCN-based photocatalysts in the heterogeneous catalysis of air pollutants are also provided.

Chemoselective catalytic hydrodefluorination of trifluoromethylalkenes towards mono-/gem-di-fluoroalkenes under metal-free conditions.

Project description:Fluorine-containing moieties show significant effects in improving the properties of functional molecules. Consequently, efficient methods for installing them into target compounds are in great demand, especially those enabled by metal-free catalysis. Here we show a diazaphospholene-catalyzed hydrodefluorination of trifluoromethylalkenes to chemoselectively construct gem-difluoroalkenes and terminal monofluoroalkenes by simple adjustment of the reactant stoichiometry. This metal-free hydrodefluorination features mild reaction conditions, good group compatibility, and almost quantitative yields for both product types. Stoichiometric experiments indicated a stepwise mechanism: hydridic addition to fluoroalkenes and subsequent β-F elimination from hydrophosphination intermediates. Density functional theory calculations disclosed the origin of chemoselectivity, regioselectivity and stereoselectivity, suggesting an electron-donating effect of the alkene-terminal fluorine atom.
