The complete mitochondrial genome of Sarcophaga polystylata (Diptera: Sarcophagidae).

ABSTRACT: Sarcophaga polystylata (Ho, 1934) remains to be of medical and epidemiological significance. Here, we report the complete mitochondrial genome (mitogenome) of S. polystylata (GenBank accession no. MW592361). The length of this mitogenome was 15,233 bp with 39.4% A, 9.5% G, 14.3% C, and 36.8% T. The mitogenome of S. polystylata was composed of 13 protein-coding genes (PCGs), two ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), and a non-coding control region. Phylogenetic tree indicated that S. polystylata and Sarcophaga peregrina clustered together closely, both of which separated clearly from other species. This study provides the mitochondrial genetic data of S. polystylata for further understanding the phylogenetic relationship of sarcophagids species.

Project description:Sarcophaga brevicornis (Diptera: Sarcophagidae) was of utmost forensic importance due to their wide distribution, ubiquitous, and synanthropic nature. The complete mitochondrial genome (mitogenome) of S. brevicornis was first sequenced and assembled in this study. The length of circular mitogenome was 15,152 bp, which showed in a typical arthropod genome, including 13 protein-coding genes (13 PCGs), two ribosomal RNA (two rRNA) genes, 22 transfer RNA (22 tRNA) genes, and an AT-rich region. Its nucleotide composition was A 39.0%, C 12.7%, G 10.6%, and T 37.7%. Furthermore, phylogenetic relationships of S. brevicornis and the published sarcophagid species were evaluated based on 13 PCGs. The results indicated that S. brevicornis was clearly separated from the other sarcophagid species, but it was closed to the species of Sarcophaga similis. This study provided a significant database reference for genetic structure and phylogenetic analysis of Sarcophagidae.

The complete mitochondrial genome (mitogenome) of S. pterygota was first sequenced and annotated. The circle DNA is composed of 13 protein-coding genes (13 PCGs), 2 ribosomal RNA (2 rRNAs), 22 transfer RNA (22 tRNAs) and a AT-rich region. It shows that arrangement of the genes is similar with the classical metazoan. The size of mitogenome is 15,236 bp containing A (40.0%), G (9.3%), T (36.6%), and C (14.1%). Moreover, phylogenetic analysis reveals that the branch of S. pterygota is clustered separately. This study enriches the mitogenome database of flesh flies and represents progress for analyzing of phylogenetic relationships.

Project description:Sarcophaga (Liosarcophaga) angarosinica (Rohdendorf, 1937) (Diptera: Sarcophagidae) is a species of both medical and ecological significance. In this study, the complete mitochondrial genome (mitogenome) of S. angarosinica was sequenced and characterized. The mitogenome has a total length of 15,215 bp, including 13 protein-coding genes, two ribosomal RNAs, 22 transfer RNAs, and an adenine and thymine-rich region. This mitogenome comprises 39.5% adenine, 9.4% guanine, 14.4% cytosine, and 36.8% thymine. Phylogenetic analysis revealed that S. angarosinica is closely related to Sarcophaga similis. This study enriches the genetic data on S. angarosinica and will contribute to establishing the phylogenetic relationships among flesh flies.

In this study, we first report the complete mitochondrial genome (mitogenome) of S. gracilior. This mitogenome was 15,534 bp in length (GenBank No. MW531675), comprising 13 protein-coding genes (PCGs), two ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), and a non-coding control region. The arrangement of genes was identical to that of ancestral metazoan. Nucleotide composition revealed a strong A + T bias, accounting for 76.7% (A 39.6%, G 9.3%, C 14.0%, T 37.1%). The phylogenetic relationships indicated that the species of S. gracilior emerged as sister to Sarcophaga melanura. This study provides important mitochondrial data for further studying evolutionary relationships and species identification of flesh flies.

Sarcophaga caerulescens can be better correlated with migration and postmortem interval (PMI) inference in forensic practice. In this study, we reported the complete mitochondrial genome (mitogenome) of S. caerulescens. The length of this mitogenome was 15,720 bp in total (GenBank accession No. MW551788), containing 13 protein-coding genes (PCGs), 2 ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), and a non-coding control region. Its nucleotide composition was A (39.7%), C (14.1%), G (9.4%), and T (36.9%). The phylogenetic relationships indicated that the species of S. caerulescens was closely related to S. similis. This study provides the mitochondrial data of S. caerulescens for further study of mitochondrial genome and enriches our understanding of the phylogenetic relationship of sarcophagid flies.

The complete mitochondrial genome of Sarcophaga tuberosa (Diptera: Sarcophagidae).

Project description:Sarcophaga tuberosa has certain sanitary and epidemiological significance and takes proteins from decaying matter. In this study, we first sequenced and analyzed the complete mitochondrial genome (mitogenome) of S. tuberosa. The length of mitogenome was 15,173 bp, consisting of A (39.5%), G (9.4%), T (37.0%), and C (14.1%), and the order and orientation of genes were identical with that from the mittogenomes of flesh flies previously reported. Phylogenetic analyses showed that S. tuberosa was clustered separately, but which was closed to the species of Sarcophaga portschinskyi. This study provided better support for further understanding of phylogenetic relationships and enriched the mitogenome database of flesh flies.

In this study, we report the complete mitochondrial genome (mitogenome) of S. tsinanensis. The length of this mitogenome was 14,972 bp in total, containing 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a non-coding control region. The arrangement of genes was the same as that of ancestral metazoan. Sarcophaga tsinanensis has a high nucleotide bias in A/T accounting for 76.80% of total nucleotides (A 39.9%, G 9.2%, C 13.9%, and T 36.9%). The result of phylogenetic analysis revealed that S. tsinanensis cluster together with species from the same subgenus Heteronychia, showing a clear monophyletic relationship. Sarcophaga tsinanensis is closely related to its sister species Sarcophaga depressifrons, Sarcophaga plotnikovi, and Sarcophaga shnitnikovi. This study provides the mitochondrial data of S. tsinanensis for further research on evolutionary relationships and species identification of flesh flies.

Project description:Sarcophaga scopariiformis (Diptera: Sarcophagidae), a potential vector of pathogens as well as one of the important flesh fly specie in forensic entomology. We have firstly sequenced and assembled the whole mitogenome of S. scopariiformis in this study. The circular mitogenome is 15,325 bp in length, consisting of A (39.3%), G (9.4%), T (36.8%), and C (14.5%). It showed in a typical mitochondrial genome similar to other sarcophagids species, which is composed of 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNA), 2 ribosomal RNA genes (rRNA), and a non-coding AT-rich region. Moreover, the phylogenetic analysis based on 13 PCGs has been conducted. The topological structure of the phylogenetic tree clearly indicated that S. scopariiformis was more closed to the species of Sarcophaga peregrina in genetic distance, compared to the other sarcophagids species. This study provided a useful data reference for genetic structure and phylogenetic analysis of Sarcophagidae.

The whole mitochondrial genome of Sarcophaga kanoi (Diptera: Sarcophagidae).

Project description:Sarcophaga kanoi Park, 1962, a widely distributed flesh fly in Southeast Asia, is important in forensic entomology. Notably, its mitochondrial genome could provide the unique and accurate molecular information in species identification which facilitate forensic practices in estimation of postmortem interval especially in putrefied cadaver cases. Thus, we sequenced and characterized the whole mitochondrial genome (mitogenome) of S. kanoi for the first time, which was collected from Southern China in this study. The 15,319 bp mitogenome contained 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNA), 2 ribosomal RNA genes (rRNA), and a putative control region. The total nucleotide composition of this circular genome was 39.7% for A, 9.3% for G, 14.2% for C, and 36.8% for T. To better understand the genetic relationship, the phylogenetic analysis was constructed according to the 13 PCGs sequence of S. kanoi and other 11 species. The phylogenetic tree showed that the S. kanoi was placed in a sub-clade with S. similis. Our study updated the new genetic information for dipteran mitogenomes, which could broaden the background knowledge for forensic entomology, molecular genetics and developmental biology. It has the potential application on the species identification using genetic markers.
