Bioactivity of Inhaled Methane and Interactions With Other Biological Gases.

ABSTRACT: A number of studies have demonstrated explicit bioactivity for exogenous methane (CH₄), even though it is conventionally considered as physiologically inert. Other reports cited in this review have demonstrated that inhaled, normoxic air-CH₄ mixtures can modulate the in vivo pathways involved in oxidative and nitrosative stress responses and key events of mitochondrial respiration and apoptosis. The overview is divided into two parts, the first being devoted to a brief review of the effects of biologically important gases in the context of hypoxia, while the second part deals with CH₄ bioactivity. Finally, the consequence of exogenous, normoxic CH₄ administration is discussed under experimental hypoxia- or ischaemia-linked conditions and in interactions between CH₄ and other biological gases, with a special emphasis on its versatile effects demonstrated in pulmonary pathologies.

Inhaled Gases for Neuroprotection of Neonates: A Review.

Project description:Importance: Hypoxic-ischemic encephalopathy (HIE) is a significant cause of morbidity and mortality in neonates. The incidence of HIE is 1-8 per 1,000 live births in developed countries. Whole-body hypothermia reduces the risk of disability or death, but 7 infants needed to be treated to prevent death or major neurodevelopmental disability. Inhalational gases may be promising synergistic agents due to their rapid onset and easy titratability. Objective: To review current data on different inhaled gases with neuroprotective properties that may serve as adjunct therapies to hypothermia. Evidence review: Literature review was performed using the PubMed database, google scholar, and ClinicalTrials.Gov. Results focused on articles published from January 1, 2005, through December 31, 2017. Articles published earlier than 2005 were included when appropriate for historical perspective. Our review emphasized preclinical and clinical studies relevant to the use of inhaled agents for neuroprotection. Findings: Based on the relevance to our topic, 111 articles were selected pertaining to the incidence of HIE, pathophysiology of HIE, therapeutic hypothermia, and emerging therapies for hypoxic-ischemic encephalopathy in preclinical and clinical settings. Supplemental tables summarizes highly relevant 49 publications that were included in this review. The selected publications emphasize the emergence of promising inhaled gases that may improve neurologic survival and alleviate neurodevelopmental disability when combined with therapeutic hypothermia in the future. Conclusions: Many inhaled agents have neuroprotective properties and could serve as an adjunct therapy to whole-body hypothermia. Inhaled agents are ideal due to their easy administration, titrability, and rapid onset and offset.

Lack of harmonisation of greenhouse gases reporting standards and the methane emissions gap.

Project description:Monitoring companies' contributions to climate dynamics and their exposure to transition risks requires accurate measurements of their non-carbon dioxide greenhouse gas emissions (non-CO2 GHG). However, carbon accounting standards are not harmonised and allow for some discretion when converting emissions of different GHGs into CO2 equivalent units, the currency in which carbon footprints are expressed. Focusing on methane, we build counterfactual harmonised standards using the latest IPCC Global Warming Potential (GWP) values over 100 years and estimate a cumulative gap in reported methane emissions of 170MtCO2e ( ~6Tg) over a sample of 2864 companies. Changing the counterfactual from GWP100to GWP20, as recently codified in certain jurisdictions and initiatives, increases the cumulative gap to 3300MtCO2e ( ~40Tg). The gap only covers direct emissions and hence understates the extent of potential under-reporting across value chains. Overall, our study underscores the importance of global harmonisation of CO2-equivalence standards to coherently track corporate GHG emissions and their exposure to transition risks.

The biological and toxicological activity of gases and vapors.

Project description:A large amount of data on the biological and toxicological activity of gases and vapors has been collected from the literature. Processes include sensory irritation thresholds, the Alarie mouse test, inhalation anesthesia, etc. It is shown that a single equation using only five descriptors (properties of the gases and vapors) plus a set of indicator variables for the given processes can correlate 643 biological and non-lethal toxicological activities of 'non-reactive' compounds with a standard deviation of 0.36 log unit. The equation is scaled to sensory irritation thresholds obtained by the procedure of Cometto-Muñiz, and Cain provides a general equation for the prediction of sensory irritation thresholds in man. It is suggested that differences in biological/toxicological activity arise primarily from transport from the gas phase to a receptor phase or area, except for odor detection thresholds where interaction with a receptor(s) is important.

Inhaled Gases as Therapies for Post-Cardiac Arrest Syndrome: A Narrative Review of Recent Developments.

Project description:Despite recent advances in the management of post-cardiac arrest syndrome (PCAS), the survival rate, without neurologic sequelae after resuscitation, remains very low. Whole-body ischemia, followed by reperfusion after cardiac arrest (CA), contributes to PCAS, for which established pharmaceutical interventions are still lacking. It has been shown that a number of different processes can ultimately lead to neuronal injury and cell death in the pathology of PCAS, including vasoconstriction, protein modification, impaired mitochondrial respiration, cell death signaling, inflammation, and excessive oxidative stress. Recently, the pathophysiological effects of inhaled gases including nitric oxide (NO), molecular hydrogen (H2), and xenon (Xe) have attracted much attention. Herein, we summarize recent literature on the application of NO, H2, and Xe for treating PCAS. Recent basic and clinical research has shown that these gases have cytoprotective effects against PCAS. Nevertheless, there are likely differences in the mechanisms by which these gases modulate reperfusion injury after CA. Further preclinical and clinical studies examining the combinations of standard post-CA care and inhaled gas treatment to prevent ischemia-reperfusion injury are warranted to improve outcomes in patients who are being failed by our current therapies.

Cough and expiration reflexes elicited by inhaled irritant gases are intensified in ovalbumin-sensitized mice.

Project description:This study was designed to determine the effect of active sensitization with ovalbumin (Ova) on cough responses to inhaled irritant gases in mice. Conscious mice moved freely in a recording chamber, while the pressure change in the chamber and audio and video signals of the mouse movements were recorded simultaneously to measure the frequencies of cough reflex (CR) and expiration reflex (ER). To further verify the accuracy of cough analysis, the intrapleural pressure was also recorded by a telemetry sensor surgically implanted in the intrapleural space in a subgroup of mice. During the irritant gas inhalation challenge, sulfur dioxide (SO2; 200 and 400 ppm) or ammonia (NH3; 0.1% and 0.2%) was drawn into the chamber at a constant flow rate for 8 min. Ova sensitization and sham sensitization with vehicle (Veh) were performed over a 25-day period in separate groups of mice. Our results showed that 1) both SO2 and NH3 inhalation challenges increased CR and ER frequencies in a concentration-dependent manner before Ova sensitization; 2) the baseline CR frequency was significantly elevated after Ova sensitization, accompanied by pronounced airway inflammation; and 3) Ova sensitization also markedly augmented the responses of CR and ER to both SO2 and NH3 inhalation challenges; in sharp contrast, the cough responses did not change after sham sensitization in the Veh group. In conclusion, Ova sensitization caused distinct and lingering increases in baseline cough frequency, and also intensified both CR and ER responses to inhaled irritant gases, which probably resulted from an allergic inflammation-induced hypersensitivity of airway sensory nerves.

A Sensor Array for the Detection and Discrimination of Methane and Other Environmental Pollutant Gases.

Project description:We address the sensitive detection and discrimination of gases impacting the environment, such as CH₄, NH₃, SO₂, and CO, using a sensor array and aided by principal component analysis (PCA). A 32-element chemiresistive sensor array consisting of nine different sensor materials including seven types of modified single-walled carbon nanotubes and two types of polymers has been constructed. PCA results demonstrate excellent discriminating ability of the chemiresistor sensor chip in the 1-30 ppm concentration range. The accuracy of the sensor was verified against data collected using cavity ring down spectroscopy. The sensor chip has also been integrated with a smartphone and has been shown to reproduce the sensing performance obtained with the laboratory measurement system.

Biochemistry of aerobic biological methane oxidation.

Project description:Methanotrophic bacteria represent a potential route to methane utilization and mitigation of methane emissions. In the first step of their metabolic pathway, aerobic methanotrophs use methane monooxygenases (MMOs) to activate methane, oxidizing it to methanol. There are two types of MMOs: a particulate, membrane-bound enzyme (pMMO) and a soluble, cytoplasmic enzyme (sMMO). The two MMOs are completely unrelated, with different architectures, metal cofactors, and mechanisms. The more prevalent of the two, pMMO, is copper-dependent, but the identity of its copper active site remains unclear. By contrast, sMMO uses a diiron active site, the catalytic cycle of which is well understood. Here we review the current state of knowledge for both MMOs, with an emphasis on recent developments and emerging hypotheses. In addition, we discuss obstacles to developing expression systems, which are needed to address outstanding questions and to facilitate future protein engineering efforts.

Ecological interactions affect the bioactivity of medicinal plants.

Project description:Essential oils produced by medicinal plants possess important bioactive properties (antibacterial, antioxidant) of high value for human society. Pollination and herbivory can modify the chemical defences of plants and therefore they may influence the bioactivity of essential oils. However, the effect of ecological interactions on plant bioactivity has not yet been evaluated. We tested the hypothesis that cross-pollination and simulated herbivory modify the chemical composition of essential oils, improving the bioactive properties of the medicinal plant Lepechinia floribunda (Lamiaceae). Through controlled experiments, we showed that essential oils from the outcrossed plant progeny had a higher relative abundance of oxygenated terpenes and it almost doubled the bacteriostatic effect on Staphylococcus aureus, compared to inbred progeny (i.e., progeny produced in absence of pollinators). Herbivory affected negatively and positively the production of rare compounds in inbred and outcrossed plants, respectively, but its effects on bioactivity still remain unknown. We show for the first time that by mediating cross-pollination (indirect ecosystem service), pollinators can improve ecosystem services linked to the biological activity of plant's essential oils. We stress the importance of the qualitative component of pollination (self, cross); an aspect usually neglected in studies of pollination services.

Biological effects of inhaled hydraulic fracturing sand dust in rats

Project description:The pulmonary inflammatory response to inhalation exposure to fracking sand dust (FSD) was investigated in a rat model. Adult male Sprague-Dawley rats were exposed by whole-body inhalation to air or an aerosol of FSD at concentrations of 10 or 30 mg/m3, 6 hours/day for 4 days. The control and FSD-exposed rats were euthanized at post-exposure time intervals of 1, 7 or 27days and pulmonary inflammatory, cytotoxic and oxidant responses were determined. Deposition of FSD particles was detected in the lungs of all the FSD-exposed rats. Analysis of bronchoalveolar lavage parameters of toxicity, oxidant generation, and inflammation did not reveal any significant persistent pulmonary toxicity in the FSD- exposed rats. Similarly, the lung histology of the FSD-exposed rats showed only minimal changes in influx of macrophages following the exposure. Determination of global gene expression profiles detected significant differential expressions of only six and five genes in the 10 mg/m3, 1 day post-exposure, and the 30 mg/m3, 7-day post-exposure FSD groups, respectively. Taken together, data obtained from the present study demonstrated that FSD inhalation exposure resulted in minimal/no toxicity or gene expression changes in the lungs of the rats.
