Detecting Redox Potentials Using Porous Boron Nitride/ATP-DNA Aptamer/Methylene Blue Biosensor to Monitor Microbial Activities.

ABSTRACT: Microbial activity has gained attention because of its impact on the environment and the quality of people's lives. Most of today's methods, which include genome sequencing and electrochemistry, are costly and difficult to manage. Our group proposed a method using the redox potential change to detect microbial activity, which is rooted in the concept that metabolic activity can change the redox potential of a microbial community. The redox potential change was captured by a biosensor consisting of porous boron nitride, ATP-DNA aptamer, and methylene blue as the fluorophore. This assembly can switch on or off when there is a redox potential change, and this change leads to a fluorescence change that can be examined using a multipurpose microplate reader. The results show that this biosensor can detect microbial community changes when its composition is changed or toxic metals are ingested.

ZnO/Boron Nitride Quantum Dots Nanocomposites for the Enhanced Photocatalytic Degradation of Methylene Blue and Methyl Orange.

Project description:In this study, a heterostructure photocatalyst of ZnO nanoparticles decorated with boron nitride quantum dots (ZnO/BNQDs) was successfully synthesized by a simple solution procedure. The synthesized ZnO/BNQDs show that the BNQDs effectively suppress the recombination of photoinduced electrons and holes and the transfer of holes from ZnO nanoparticles by the formation of a heterojunction. The ZnO/BNQD nanocomposites thus demonstrate superior photocatalytic performances and excellent stability for the degradation of methylene blue (MB) and methyl orange (MO) under UV light irradiation. Based on the obtained results, the possible photocatalytic mechanism is proposed and discussed. Thus, the ZnO/BNQD nanocomposites demonstrate potential as an efficient low-cost photocatalyst for application in the photodegradation of organic dyes in wastewater for environmental remediation.

Sensitive Identification of Microcystin-LR via a Reagent-Free and Reusable Electrochemical Biosensor Using a Methylene Blue-Labeled Aptamer.

Project description:We report a methylene blue (MB)-modified electrochemical aptamer (E-AB) sensor for determining microcystin-LR (MC-LR). The signal transduction of the sensor was based on changes in conformation and position of MB induced by the binding between MC-LR and the modified aptamer probe. In the absence of MC-LR, an aptamer probe was considered partially folded. After combining aptamer and MC-LR, the configuration of the aptamer probe changed and facilitated the electron transfer between MB and the electrode surface. As a result, an increased current response was observed. We optimized the parameters and evaluated the electrochemical performance of the sensor using square wave voltammetry (SWV). MC-LR was measured from 1.0 to 750.0 ng/L with a detection limit of 0.53 ng/L. The reliability of the method was verified by the determination of MC-LR in environmental real samples, such as pond water and tap water. Moreover, we demonstrated that this reagent-less biosensor could be regenerated and reused after rinsing with deionized water with good accuracy and reproducibility. As a reusable and regenerable E-AB sensor, this rapid, reagent-free, and sensitive sensing platform will facilitate routine monitoring of MC-LR in actual samples.

Anti-Aging Potentials of Methylene Blue for Human Skin Longevity.

Project description:Oxidative stress is the major cause of skin aging that includes wrinkles, pigmentation, and weakened wound healing ability. Application of antioxidants in skin care is well accepted as an effective approach to delay the skin aging process. Methylene blue (MB), a traditional mitochondrial-targeting antioxidant, showed a potent ROS scavenging efficacy in cultured human skin fibroblasts derived from healthy donors and from patients with progeria, a genetic premature aging disease. In comparison with other widely used general and mitochondrial-targeting antioxidants, we found that MB was more effective in stimulating skin fibroblast proliferation and delaying cellular senescence. The skin irritation test, performed on an in vitro reconstructed 3D human skin model, indicated that MB was safe for long-term use, and did not cause irritation even at high concentrations. Application of MB to this 3D skin model further demonstrated that MB improved skin viability, promoted wound healing and increased skin hydration and dermis thickness. Gene expression analysis showed that MB treatment altered the expression of a subset of extracellular matrix proteins in the skin, including upregulation of elastin and collagen 2A1, two essential components for healthy skin. Altogether, our study suggests that MB has a great potential for skin care.

Porous ZnCl2-Activated Carbon from Shaddock Peel: Methylene Blue Adsorption Behavior.

Project description:It is of great interest and importance to resource utilization of waste biomass to produce porous carbon for environmental treatments. Pore structure and properties of the obtained carbon mainly relate to carbonization conditions and biomass types. In this work, a series of porous, biomass-activated carbons (AC) were prepared using shaddock peel, with ZnCl2 as a pore-forming agent. The effect of carbonization temperature and the mass ratio between ZnCl2 and shaddock peel were thoroughly investigated. The material composition, surface chemical properties, and surface structures of samples were carefully characterized. The specific surface area and adsorption capacity to methylene blue (MB) of adsorbents were changed with the carbonization temperature and the mass ratios between ZnCl2 and shaddock peel; when the temperature was at 1000 °C and the mass ratio was equal to 2:1, the resulting adsorbent had the largest specific surface area of 2398.74 m2/g and average pore size of 3.04 nm, which showed the highest adsorption capacity to MB to be 869.57 mg/g. The adsorption processes of biomass AC adsorbent matched the pseudo-second-order kinetic model and Langmuir isotherm model. This efficient and environmentally friendly biomass AC adsorbent from shaddock peel, activated by ZnCl2, is a promising candidate for the treatment of water pollution.

Sweet-Potato-Vine-Based High-Performance Porous Carbon for Methylene Blue Adsorption.

Project description:In this study, sweet-potato-vine-based porous carbon (SPVPC) was prepared using zinc chloride as an activating and pore-forming agent. The optimised SPVPC exhibited abundant porous structures with a high specific surface area of 1397.8 m2 g-1. Moreover, SPVPC exhibited excellent adsorption characteristics for removing methylene blue (MB) from aqueous solutions. The maximum adsorption capacity for MB reached 653.6 mg g-1, and the reusability was satisfactory. The adsorption kinetics and isotherm were in good agreement with the pseudo-second-order kinetics and Langmuir models, respectively. The adsorption mechanism was summarised as the synergistic effects of the hierarchically porous structures in SPVPC and various interactions between SPVPC and MB. Considering its low cost and excellent adsorption performance, the prepared porous carbon is a promising adsorbent candidate for dye wastewater treatment.

This study opens a new approach for the utilization of ginger straw waste, and the porous materials can be employed as great potential adsorbents for treating dye wastewater.

An ultrasensitive Fano resonance biosensor using two dimensional hexagonal boron nitride nanosheets: theoretical analysis.

Project description:This study proposed a novel Fano resonance (FR) biosensor with ultrahigh detection sensitivity by integrating two dimensional (2D) hexagonal boron nitride (h-BN) nanosheets with a plasmonic silver film-silicon hybrid nanostructure. Owing to its ultralow-loss in surface plasmon polaritons (SPPs), 2D h-BN nanosheets can act as a planar photon waveguide (PWG) for generating energy level splitting. Notably, both asymmetric FR sharp lines and plasmon induced transparency (PIT) can be produced by modulating the coupling strength between the planar PWG mode provided by h-BN nanosheets and the surface plasmon polariton (SPP) mode in the silver film-silicon hybrid nanostructure. Compared with conventional phase-modulation SPR biosensors, our proposed configuration based on Fano resonance can produce ultrahigh reflectivity of 0.934 and overcome the limitation of quasi-darkness reflectivity which is difficult for further phase extraction. More importantly, our proposed FR configuration can provide a promising phase detection sensitivity as high as 3.13 × 106 degree per RIU (refractive index unit, RIU), which is enhanced by almost 100 times compared with conventional phase-modulation SPR biosensors. In addition, our proposed configuration has also shown the characteristics of multiple-order Fano resonances, largely depending on the partial coupling between the SPP mode and the different-order PWG mode. Our proposed FR biosensor can provide a highly promising candidate for designing a multiple-order FR platform for performing ultrasensitive detection.

Europium (III) Organic Complexes in Porous Boron Nitride Microfibers: Efficient Hybrid Luminescent Material.

Project description:We report the design and synthesis of a novel kind of organic-inorganic hybrid material via the incorporation of europium (III) β-diketonate complexes (Eu(TTA)3, TTA = 2-thenoyltrifluoroacetone) into one-dimensional (1D) porous boron nitride (BN) microfibers. The developed Eu(TTA)3@BN hybrid composites with typical 1D fibrous morphology exhibit bright visible red-light emission on UV illumination. The confinement of Eu(TTA)3 within pores of BN microfibers not only decreases the aggregation-caused quenching in solid Eu(TTA)3, but also improves their thermal stabilities. Moreover, The strong interactions between Eu(TTA)3 and porous BN matrix result in an interesting energy transfer process from BN host to TTA ligand and TTA ligand to Eu3+ ions, leading to the remarkable increase of red emission. The synthetic approach should be a very promising strategy which can be easily expanded to other hybrid luminescent materials based on porous BN.

Piezoelectric Response of Porous Nanotubes Derived from Hexagonal Boron Nitride under Strain Influence.

Project description:A computational study via periodic density functional theory of porous nanotubes derived from single-layer surfaces of porous hexagonal boron nitride nanotubes (PBNNTs) and inorganic graphenylene-like boron nitride nanotubes (IGP-BNNTs) has been carried out with the main focus in its piezoelectric behavior. The simulations showed that the strain provides a meaningful improve in the piezoelectric response on the zigzag porous boron nitride nanotubes. Additionally, its stability, possible formation, elastic, and electronic properties were analyzed, and for comparison purpose, the porous graphene and graphenylene nanotubes were studied. From the elastic properties study, it was found that IGP-BNNTs exhibited a higher rigidity because of the influence of the superficial porous area, as compared to PBNNTs. The present study provides evidence that the strain is a way to maximize the piezoelectric response and make this material a good candidate for electromechanical devices.

Ultraviolet radiation protection potentials of Methylene Blue for human skin and coral reef health.

Project description:Methylene blue (MB) is a century-old medicine, a laboratory dye, and recently shown as a premier antioxidant that combats ROS-induced cellular aging in human skins. Given MB's molecular structure and light absorption properties, we hypothesize that MB has the potential to be considered as a sunscreen active for UV radiation protection. In this study, we tested the effects of MB on UVB ray-induced DNA double-strand breaks in primary human keratinocytes. We found that MB treatment reduced DNA damages caused by UVB irradiation and subsequent cell death. Next, we compared MB with Oxybenzone, which is the most commonly used chemical active ingredient in sunscreens but recently proven to be hazardous to aquatic ecosystems, in particular to coral reefs. At the same concentrations, MB showed more effective UVB absorption ability than Oxybenzone and significantly outperformed Oxybenzone in the prevention of UVB-induced DNA damage and the clearance of UVA-induced cellular ROS. Furthermore, unlike Oxybenzone, MB-containing seawater did not affect the growth of the coral species Xenia umbellata. Altogether, our study suggests that MB has the potential to be a coral reef-friendly sunscreen active ingredient that can provide broad-spectrum protection against UVA and UVB.
