In the Search of Marine Pestiviruses: First Case of Phocoena Pestivirus in a Belt Sea Harbour Porpoise.

ABSTRACT: Pestiviruses are widespread pathogens causing severe acute and chronic diseases among terrestrial mammals. Recently, Phocoena pestivirus (PhoPeV) was described in harbour porpoises (Phocoena phocoena) of the North Sea, expanding the host range to marine mammals. While the role of the virus is unknown, intrauterine infections with the most closely related pestiviruses- Bungowannah pestivirus (BuPV) and Linda virus (LindaV)-can cause increased rates of abortions and deaths in young piglets. Such diseases could severely impact already vulnerable harbour porpoise populations. Here, we investigated the presence of PhoPeV in 77 harbour porpoises, 277 harbour seals (Phoca vitulina), grey seals (Halichoerus grypus) and ringed seals (Pusa hispida) collected in the Baltic Sea region between 2002 and 2019. The full genome sequence of a pestivirus was obtained from a juvenile female porpoise collected along the coast of Zealand in Denmark in 2011. The comparative Bayesian phylogenetic analyses revealed a close relationship between the new PhoPeV sequence and previously published North Sea sequences with a recent divergence from genotype 1 sequences between 2005 and 2009. Our findings provide further insight into the circulation of PhoPeV and expand the distribution from the North Sea to the Baltic Sea region with possible implications for the vulnerable Belt Sea and endangered Baltic Proper harbour porpoise populations.

Project description:Marine traffic is threatening cetaceans on a local and global scale. The Istanbul Strait is one of the busiest waterways, with up to 2,500 vessels present daily. This is the first study to assess the magnitude of short- and long-term behavioural changes of the endangered Black Sea harbour porpoises (Phocoena phocoena relicta) in the presence of marine vessels within the Istanbul Strait. Markov chains were used to investigate the effect of vessel presence on the transition probability between behavioural states (diving, surface-feeding and travelling), and to quantify the effect on the behavioural budget and bout length (duration of time spent in a given state) of porpoises. Further, the changes on swimming directions of porpoises in relation to vessel speed and distance was investigated using generalized linear models. In vessel presence, porpoises were less likely to remain in a given behavioural state and instead more likely to switch to another state. Because of this, the bout length of all three behavioural states decreased significantly in the presence of vessels. The vessel effect was sufficiently large to alter the behavioural budget, with surface-feeding decreasing significantly in the presence of vessels. However, when taking into account the proportion of time that porpoises were exposed to vessels (i.e. 50%), the measured effect size was not large enough to significantly alter the animals' cumulative (diurnal) behavioural budget. Additionally, vessel speed and distance had a significant effect on the probability of porpoises showing a response in their swimming directions. The southern and middle sections of the Istanbul Strait, which have the heaviest marine traffic pressure, had the lowest porpoise sightings throughout the year. Conversely, northern sections that were exposed to a lesser degree of marine traffic hold the highest porpoise sightings. The effect shown in this study in combination with increasing human impacts within the northern sections should be considered carefully and species-specific conservation actions, including establishment of protected areas, should be put in place to prevent the long-term consequences of marine traffic on the Black Sea harbour porpoise population.

Project description:Harbour porpoises (Phocoena phocoena) are the only native cetacean species in the German North and Baltic Seas and the final host of Anisakis (A.) simplex, which infects their first and second gastric compartments and may cause chronic ulcerative gastritis. Anisakis simplex belongs to the family Anisakidae (Ascaridoidea, Rhabditida) as well as the phocine gastric nematode species Pseudoterranova (P.) decipiens and Contracaecum (C.) osculatum. These nematode species are the main causative agents for the zoonosis anisakidosis. The taxonomy of these genus with life cycles including crustaceans and commercially important fish is complex because of the formation of sibling species. Little is known about anisakid species infecting porpoises in the study area. Mature nematodes and larval stages are often identifiable only by molecular methods due to high morphological and genetic similarity. The restriction fragment length polymorphism (RFLP) method is an alternative to sequencing and was applied to identify anisakid nematodes found in harbour porpoises from the North Sea, Baltic Sea and North Atlantic to species level for the first time. In the study areas, five gastric nematodes from different harbour porpoise hosts were selected to be investigated with restriction enzymes HinfI, RsaI and HaeIII, which were able to differentiate several anisakid nematode species by characteristic banding patterns. Anisakis simplex s. s. was the dominant species found in the North Sea and Baltic porpoises, identified by all three restriction enzymes. Additionally, a hybrid of A. simplex s. s. and A. pegreffii was determined by HinfI in the North Sea samples. Within the North Atlantic specimens, A. simplex s. s., P. decipiens s. s. and Hysterothylacium (H.) aduncum were identified by all enzymes. This demonstrates the value of the RFLP method and the chosen restriction enzymes for the species identification of a broad variety of anisakid nematodes affecting the health of marine mammals.
