Gammarinema scyllae sp. n. and Monhystrium mangrovi sp. n. (Nematoda: Monhysteridae) from land crabs from New Caledonia.

ABSTRACT: Two new species of nematodes associated with crabs are described from New Caledonia. Gammarinema scyllae sp. n. from the gill chambers of Scylla serrata (Forsskål) is characterised by 3-4 mm long body, small outer labial and cephalic sensilla, distinct ocelli, short straight spicules and sub-cylindrical tail. Monhystrium mangrovi sp. n. from the gill chambers and body cavity of mangrove crab Neosarmatium sp. is characterised by 1-1.4 mm long body; outer labial sensilla longer than cephalic sensilla, amphid located at level with posterior stoma chamber, denticles in posterior stoma chamber and five pairs of genital papilla on tail. Phylogenetic relationships of two new species and other nematodes from the family Monhysteridae are analysed based on 18S and partial 28S rDNA sequences.

Project description:A recent examination of newly obtained specimens of philometrid nematodes (Philometridae) parasitising carangid and serranid fishes off New Caledonia, South Pacific, revealed the presence of several nematodes of the genus Philometra Costa, 1845, including three new species: P. austropacifica n. sp. (males and females) from the ovary of Alepes vari (Carangidae), P. piscaria n. sp. (males) from the ovary of Epinephelus coioides (Serranidae), and P. selaris n. sp. (males) probably from the abdominal cavity (found in washings) of Selar crumenophthalmus (Carangidae). The new species are characterised mainly by the length and structure of the spicules and gubernaculum, body size, their location in the host and the type of host. Philometra austropacifica n. sp. is the first known nominal gonad-infecting species of Philometra parasitising a carangid fish. In addition, the gravid female of P. fasciati Moravec & Justine, 2008 from the ovary of Epinephelus fasciatus (Serranidae) is described for the first time. Carangid host fish were identified by both morphology and DNA barcoding.

Phasmarhabditis zhejiangensis sp. nov. (Nematoda: Rhabditidae), a new rhabditid nematode from Zhejiang, China.

Project description:A new nematode species of the genus Phasmarhabditis was isolated from the body surface of a slug (Philomycus bilineatus Benson, PB). Morphological and molecular analyses confirmed this nematode as a new species. The nematode was named Phasmarhabditis zhejiangensis sp. nov. (Nematoda: Rhabditidae) and is dioecious. In males, the open bursa with genital papillae is characterized by the formula 1-1-1-2-1-3, and the spicule length is 58μm. In female, the vulva is located approximately in the middle of the body. The nematode belongs to papillosa group because of its tail shape pointed with filiform tip. The phasmids are rod-shaped. The posterior anus is slightly swollen. P. zhejiangensis was further characterized by internal transcribed spacer (ITS), 18S rDNA and 28S rDNA sequences. After the sequencing results were compared with sequences available from the National Center for Biotechnology Information (NCBI), the maximum similarities of ITS, 18S and 28S sequences were 89.81%, 96.22% and 95.28%, respectively. Phylogenetic analyses placed Phasmarhabditis zhejiangensis sp. nov. in the genus Phasmarhabditis.

Project description:Recent examinations of spirurid nematodes (Spirurida) from deep-sea or coral reef marine fishes off New Caledonia, collected in the years 2006-2009, revealed the presence of the following five species: Ichthyofilaroides novaecaledoniensis (Moravec et Justine, 2009) n. gen., n. comb. (transferred from Ichthyofilaria Yamaguti, 1935) (females) (Guyanemidae) from the deep-sea fish Hoplichthys citrinus (Hoplichthyidae, Scorpaeniformes), Philometra sp. (male fourth-stage larva and mature female) (Philometridae) from Epinephelus maculatus (Serranidae, Perciformes), Ascarophis (Dentiascarophis) adioryx Machida, 1981 (female) (Cystidicolidae) from Sargocentron spiniferum (Holocentridae, Beryciformes), Ascarophis (Ascarophis) nasonis Machida, 1981 (males and females) from Naso lituratus and N. unicornis (Acanthuridae, Perciformes), and Ascarophisnema tridentatum Moravec et Justine, 2010 (female) from Gymnocranius grandoculis (Lethrinidae, Perciformes). Two species, I. novaecaledoniensis and A. nasonis, are redescribed based on light microscopical (LM) and scanning electron microscopical (SEM) examinations, the latter used in these species for the first time. Morphological data on the specimen of A. tridentatum from the new host species are provided. Philometra sp. (from E. maculatus) most probably represents a new gonad-infecting species of this genus. The newly established genus Ichthyofilaroides n. gen. is characterized mainly by the presence of a small buccal capsule and by the number and distribution of cephalic papillae in the female; it is the sixth genus in the Guyanemidae.

Neotobrilus nicsmolae n. sp. (Tobrilidae: Nematoda) and Chronogaster carolinensis n. sp. (Chronogasteridae: Nematoda) from Lake Phelps, North Carolina.

Project description:Two new species, Neotobrilus nicsmolae n. sp. and Chronogaster carolinensis n. sp. are described from a small, acidic, temperate, natural lake in North Carolina. N. nicsmolae n. sp. comes close to three members of the genus reported from North America, N. filipjevi, N. longus, and N. hopei. However, N. nicsmolae is unique with in the genus in having a combination of characters: size smaller than 1,700 μm, shorter outer labial and cephalic setae, tail shorter than 250 μm, last ventromedian supplement close (about 5 μm) to cloacal opening, spicule length of 61 to 85 μm, flagelloid sperm, and possession of subterminal setae. Assessment of relationships among clades within the Triplonchida using DNA sequences of the D2D3 expansion segment of the LSU rDNA showed that the family Trichodoridae and the genus Tripyla were recovered as monophyletic. The genus Tobrilus was recovered as monophyletic in the neighbor-joining and maximum likelihood trees, but that was not so in the maximum-parsimony tree. The separation among genera of the Trichodoridae, i.e., Trichodorus and Paratrichodorus, was not clear-cut in all phylograms. Chronogaster carolinensis n. sp. in having one ventral mucro with no spine and vacuolated lateral glandular bodies comes close to C. typica and C. ethiopica but differs from all hitherto known species in a combination of characteristics: in having long cephalic setae, long stoma, crystalloid bodies, vacuolated lateral glandular bodies, and a tail terminus with blunt ventral mucro, and its lack of lateral line.

In this study, the complete genome of NC201 was sequenced. This genome consists of a chromosome (4.13 Mb) and a chromid (1.24 Mb). The genome contains gene clusters coding for antibacterial peptides and secondary metabolites.

New stiletto flies from New Caledonia (Therevidae, Agapophytinae).

Project description:Stiletto-flies (Diptera: Therevidae) are highly diverse and species-rich in Australia and New Zealand, yet relatively few species have been recorded from neighbouring Papua New Guinea, Indonesia and throughout the remainder of Oceania. Indeed, in New Caledonia only a single species of the widely distributed Australasian genus Anabarhynchus Macquart (Therevinae) is previously known. Herein we describe two new agapophytine genera (i.e., Jeanchazeauia gen. nov., Calophytus gen. nov.), together comprising nine charismatic new species; this represents a first record of the subfamily from New Caledonia. The new genera and species are described and figured.

Morphological and Molecular Characterization of a New Root-Knot Nematode, Meloidogyne thailandica n. sp. (Nematoda: Meloidogynidae), Parasitizing Ginger (Zingiber sp.).

Project description:A root-knot nematode Meloidogyne thailandica n. sp. was discovered on roots of ginger (Zingiber spp.) intercepted from Thailand in October 2002 by the U.S. Department of Agriculture Animal and Plant Health Inspection Service at the port of San Francisco. Comparison by light microscopy (LM) and scanning electron microscopy (SEM) to five other morphologically related species (M. incognita, M. arenaria, M. microcephala, M. megatyla, and M. enterolobii) revealed that the new species differs from these by one or more of the following: body, tail and hyaline tail length, shape of head, tail and tail terminus of second-stage juveniles; stylet length and shape of spicules in males; perineal pattern, stylet length and shape of knobs in females. The distinctive perineal pattern is oval to rectangular, with smooth to moderately wavy and coarse striae, and with characteristic radial structures present underneath the pattern area; the dorsal arch is high, sometimes round to rectangular, and striae in and around the anal area form a thick network-like pattern interrupted by lateral lines and large phasmids. Second-stage juveniles have a long, slender tail and long, gradually tapering hyaline tail region ending in a rounded terminus. Male spicules commonly have an acutely angled shaft with a bidentate terminus. Molecular data from the ribosomal large subunit D3 expansion segment revealed four haplotypes, two of which were unique and distinguish M. thailandica n. sp. from M. arenaria, M. incognita, and M. javanica.

Description of Prionchulus jonkershoekensis n. sp. (Nematoda: Mononchida), a new predatory species from South Africa.

Project description:Prionchulus jonkershoekensis n. sp. is described from South Africa and illustrated using morphological, morphometric, and molecular techniques. This species is characterized by its body length (1.78-2.14 mm); the size of buccal cavity (38-44 × 24-31 µm), lower dorsal tooth position in relation to buccal cavity base, the position of amphidial aperture just above dorsal tooth apex, pars proximalis vaginae with almost straight walls, and tail 144-158 µm long with sickle shaped posterior third part. Phylogenetic analyses based on 18 S rDNA and 28 S rDNA of P. jonkershoekensis n. sp. revealed close relationships of the new species with Prionchulus punctatus and Prionchulus muscorum. This is also an additional geographical record for the genus from South Africa.

Meloidogyne aberrans sp. nov. (Nematoda: Meloidogynidae), a new root-knot nematode parasitizing kiwifruit in China.

Project description:High infection rates of roots of wild kiwifruit (Actinidia chinensis Planch) and soil infestation by a root-knot nematode were found in Anshun, GuiZhou Province, China. Morphology, esterase phenotype and molecular analyses confirmed that this nematode was different from previously described root-knot nematodes. In this report, the species is described, illustrated and named Meloidogyne aberrans sp. nov. The new species has a unique combination of characters. A prominent posterior protuberance, round and faint perineal pattern and a medium-length stylet (13.6-15.5 μm) characterized the females. Second-stage juveniles (J2) were characterized by a smooth lip region with distinctly protruded medial lips and a depression in outline at the oral aperture, a relatively long stylet (15.9-16.8 μm), four incisures in the lateral field and a very short, even poorly defined, hyaline tail terminus (2.2-5.5 μm). More incisures (11-15) existed in the lateral field of males, and the stylet and spicules of males were 18.2-19.6 μm and 22.7-36.8 μm long respectively. Egg masses were typically produced within the roots of kiwifruit. The new species had a rare Est phenotype, S2. Phylogenetic trees inferred from SSU, LSU D2D3, ITS, and partial coxII-16S rRNA revealed that M. aberrans sp. nov. was within the Meloidogyne clade and was distinguished from all described root-knot nematodes. Moreover, from histopathological observations, M. aberrans sp. nov. induced the formation of multinucleate giant cells.
