Microplastic pollution in urban Lake Phewa, Nepal: the first report on abundance and composition in surface water of lake in different seasons.

ABSTRACT: Microplastics are man-made pollutants which have been detected in surface water and groundwater. Research on microplastic concentration in aquatic environment is attracting scientists from developing countries, but in Nepal no information regarding microplastic in freshwater system is available. Therefore, this study investigates the presence and abundance of microplastic in lake surface water of Phewa Lake, the second largest lake in Nepal. The average concentration of microplastic for surface water was 2.96 ± 1.83 particles/L in winter (dry) season and 1.51 ± 0.62 particles/L in rainy (wet) season. Significant difference with t = 4.687 (p < 0.01) in microplastic concentration was observed in two different seasons. Fibers (93.04% for winter and 96.69% for rainy season) were the commonly found microplastic type in lake water and transparent as the dominant color for the two seasons. Almost all the detected microplastic were found to be < 1 mm in size. Due to the small size of microplastic and unavailability of micro-Fourier transform infrared spectroscopy (μ-FTIR) and Raman spectroscopy in Nepal, polymer identification was not done. The findings from this study can provide a valuable baseline data on microplastics for the first time in Nepal's freshwater lake environment.

Project description:BackgroundDue to the copious disposal of plastics, marine ecosystems receive a large part of this waste. Microplastics (MPs) are solid particles smaller than 5 millimeters in size. Among the plastic polymers, polystyrene (PS) is one of the most commonly used and discarded. Due to its density being greater than that of water, it accumulates in marine sediments, potentially affecting benthic communities. This study investigated the ingestion of MP and their effect on the meiofauna community of a sandy beach. Meiofauna are an important trophic link between the basal and higher trophic levels of sedimentary food webs and may therefore be substantially involved in trophic transfer of MP and their associated compounds.MethodsWe incubated microcosms without addition of MP (controls) and treatments contaminated with PS MP (1-µm) in marine sediments at three nominal concentrations (103, 105, 107particles/mL), for nine days, and sampled for meiofauna with collections every three days. At each sampling time, meiofauna were collected, quantified and identified to higher-taxon level, and ingestion of MP was quantified under an epifluorescence microscope.ResultsExcept for Tardigrada, all meiofauna taxa (Nematoda, turbellarians, Copepoda, Nauplii, Acari and Gastrotricha) ingested MP. Absorption was strongly dose dependent, being highest at 107 particles/mL, very low at 105 particles/mL and non-demonstrable at 103 particles/mL. Nematodes accumulated MP mainly in the intestine; MP abundance in the intestine increased with increasing incubation time. The total meiofauna density and species richness were significantly lower at the lowest MP concentration, while at the highest concentration these parameters were very similar to the control. In contrast, Shannon-Wiener diversity and evenness were greater in treatments with low MP concentration. However, these results should be interpreted with caution because of the low meiofauna abundances at the lower two MP concentrations.ConclusionAt the highest MP concentration, abundance, taxonomic diversity and community structure of a beach meiofauna community were not significantly affected, suggesting that MP effects on meiofauna are at most subtle. However, lower MP concentrations did cause substantial declines in abundance and diversity, in line with previous studies at the population and community level. While we can only speculate on the underlying mechanism(s) of this counterintuitive response, results suggest that further research is needed to better understand MP effects on marine benthic communities.

Project description:To mitigate harmful cyanobacterial blooms (HCBs), toxic algicides have been used, but alternative methods of HCB prevention are needed. Our goal was to test the prophylactic addition of glucose to inhibit HCB development, using Microcystis and the toxin microcystin as the HCB model. Water samples were collected weekly, from 4 June to 2 July, from Harsha Lake in southwestern Ohio during the 2021 algal bloom season. From each weekly sample, a 25 mL aliquot was frozen for a 16S rRNA gene sequencing analysis. Then, 200 mL of Harsha Lake water was added to each of the three culture flasks, and glucose was added to create concentrations of 0 mM (control), 1.39 mM, or 13.9 mM glucose, respectively. The microcystin concentration in each flask was measured after 1 and 2 weeks of incubation. The results showed an 80 to 90% reduction in microcystin concentrations in glucose-treated water compared to the control. At the end of the second week of incubation, a 25 mL sample was also obtained from each of the culture flasks for molecular analysis, including a 16S rRNA gene sequencing and qPCR-based quantification of Microcystis target genes. Based on these analyses, the glucose-treated water contained significantly lower Microcystis and microcystin producing gene (mcy) copy numbers than the control. The 16S rRNA sequencing analysis also revealed that Cyanobacteria and Proteobacteria were initially the most abundant bacterial phyla in the Harsha Lake water, but as the summer progressed, Cyanobacteria became the dominant phyla. However, in the glucose-treated water, the Cyanobacteria decreased and the Proteobacteria increased in weekly abundance compared to the control. This glucose-induced proteobacterial increase in abundance was driven primarily by increases in two distinct families of Proteobacteria: Devosiaceae and Rhizobiaceae. In conclusion, the prophylactic addition of glucose to Harsha Lake water samples reduced Cyanobacteria's relative abundance, Microcystis numbers and microcystin concentrations and increased the relative abundance of Proteobacteria compared to the control.

Project description:With the development of urbanisation and the increasing number of modern vehicles, traffic contamination has become an important source of environmental pollution. Most previous studies have focused on using roadside soil or plants to determine the spatial pattern of traffic pollutants along roads and the factors that influence this pattern, whereas few studies have reconstructed pollution histories caused by traffic using suitable methods. In this study, two gravity cores were obtained from Qianhu Lake, which is in the Zhongshan tourist area of Nanjing City and is distant from industrial areas. An accurate chronological framework covering the period from 1994 to 2014 was established using the correlation between the variation in grain size of the sediment cores and the variation in annual rainfall in Nanjing City. Moreover, magnetic and chemical parameters were also measured, and the results demonstrated that concentration-related magnetic parameters exhibited different correlations with different heavy metal concentrations. These correlations were significantly positive for Zn, Pb, and Co; weakly positive for Ni; absent for Cr; and negative for V. Combined with statistical data on industrial emissions and private cars in Nanjing City since 1994, the observed variations in magnetic susceptibility, anhysteretic remanent magnetisation, saturation isothermal remanent magnetisation, Zn, Pb, and Co, were controlled by traffic activities in the tourist area but not by industry. Therefore, the variations in these parameters record the traffic pollution history of the study area. Combined with the obtained chronological framework, the traffic-related pollution history could be divided into two stages: 1) from 1994 to 2003, when traffic-related pollution became increasingly serious because of the exponential increase in the number of private cars and the prosperity of tourism; 2) from 2003 to 2014, when traffic-related pollution continuously increased but at a much slower rate than in stage 1. This slower rate of increase was probably related to the maximum carrying capacity of the tourist area and technological innovations in automobile manufacturing, as well as improvements in fuels.
