Project description:An Infinium microarray platform (GPL28271, HorvathMammalMethylChip40) was used to generate DNA methylation data from blood samples from yellow-bellied marmots (Marmota flaviventris). DNA methylation data from n=159 blood samples. All samples were collected as part of a long-term study of a free-living population of yellow-bellied marmots in the Gunnison National Forest, Colorado (USA), where marmots were captured and blood samples collected biweekly during the their active season (May to August). Genomic DNA was extracted with Qiagen DNeasy blood and tissue kit.

Hibernation slows epigenetic ageing in yellow-bellied marmots.

Project description:Species that hibernate generally live longer than would be expected based solely on their body size. Hibernation is characterized by long periods of metabolic suppression (torpor) interspersed by short periods of increased metabolism (arousal). The torpor-arousal cycles occur multiple times during hibernation, and it has been suggested that processes controlling the transition between torpor and arousal states cause ageing suppression. Metabolic rate is also a known correlate of longevity; we thus proposed the 'hibernation-ageing hypothesis' whereby ageing is suspended during hibernation. We tested this hypothesis in a well-studied population of yellow-bellied marmots (Marmota flaviventer), which spend 7-8 months per year hibernating. We used two approaches to estimate epigenetic age: the epigenetic clock and the epigenetic pacemaker. Variation in epigenetic age of 149 samples collected throughout the life of 73 females was modelled using generalized additive mixed models (GAMM), where season (cyclic cubic spline) and chronological age (cubic spline) were fixed effects. As expected, the GAMM using epigenetic ages calculated from the epigenetic pacemaker was better able to detect nonlinear patterns in epigenetic ageing over time. We observed a logarithmic curve of epigenetic age with time, where the epigenetic age increased at a higher rate until females reached sexual maturity (two years old). With respect to circannual patterns, the epigenetic age increased during the active season and essentially stalled during the hibernation period. Taken together, our results are consistent with the hibernation-ageing hypothesis and may explain the enhanced longevity in hibernators.

Gene expression shifts in yellow-bellied marmots prior to natal dispersal.

Project description:The causes and consequences of vertebrate natal dispersal have been studied extensively, yet little is known about the molecular mechanisms involved. We used RNA-seq to quantify transcriptomic gene expression in blood of wild yellow-bellied marmots (Marmota flaviventer) prior to dispersing from or remaining philopatric to their natal colony. We tested 3 predictions. First, we hypothesized dispersers and residents will differentially express genes and gene networks since dispersal is physiologically demanding. Second, we expected differentially expressed genes to be involved in metabolism, circadian processes, and immune function. Finally, in dispersing individuals, we predicted differentially expressed genes would change as a function of sampling date relative to dispersal date. We detected 150 differentially expressed genes, including genes that have critical roles in lipid metabolism and antigen defense. Gene network analysis revealed a module of 126 coexpressed genes associated with dispersal that was enriched for extracellular immune function. Of the dispersal-associated genes, 22 altered expression as a function of days until dispersal, suggesting that dispersal-associated genes do not initiate transcription on the same time scale. Our results provide novel insights into the fundamental molecular changes required for dispersal and suggest evolutionary conservation of functional pathways during this behavioral process.

Relaxing life of the city? Allostatic load in yellow-bellied marmots along a rural-urban continuum.

Project description:Urban environments are expanding. As rural areas are urbanized, animals living in those environments must respond. Examinations of ecological responses to urbanization are abundant, but much less work has focused on the physiological responses driving those ecological patterns, particularly in mammals. Whether an animal interprets urbanized environments as stressful or not can help us understand, and even predict, the likelihood of individuals persisting in urbanized areas. Unpredictable events can cause stress and responses to such events can deplete limited stores of energy. Differences between required and available energy is termed allostatic load and is an indicator of stress. Allostatic load, and hence stress, is correlated with baseline levels of the metabolic hormones, glucocorticoids. We examined allostatic load in yellow-bellied marmots along a rural-urban gradient through analysis of fecal glucocorticoid metabolites (FGMs). We used GIS data and 'on-the-ground' measurements to quantify the degree of urbanization. We collected fecal samples from males and females of all age classes at six sites along this continuum. Female marmots had higher FGMs than males. All age groups of marmots exhibited a parabolic relationship between the degree of urbanization and FGM levels. In general, adult marmots had higher FGMs in more rural than urban environments, and both juveniles and yearlings had exhibited higher FGM levels in more urban environments.

Elevation and vegetation determine Cryptosporidium oocyst shedding by yellow-bellied marmots (Marmota flaviventris) in the Sierra Nevada Mountains.

Project description:Wildlife are increasingly recognized as important biological reservoirs of zoonotic species of Cryptosporidium that might contaminate water and cause human exposure to this protozoal parasite. The habitat range of the yellow-bellied marmot (Marmota flaviventris) overlaps extensively with the watershed boundaries of municipal water supplies for California communities along the foothills of the Sierra Nevada. We conducted a cross-sectional epidemiological study to estimate the fecal shedding of Cryptosporidium oocysts by yellow-bellied marmots and to quantify the environmental loading rate and determine risk factors for Cryptosporidium fecal shedding in this montane wildlife species. The observed proportion of Cryptosporidium positive fecal samples was 14.7% (33/224, positive number relative to total number samples) and the environmental loading rate was estimated to be 10,693 oocysts animal(-1) day(-1). Fecal shedding was associated with the elevation and vegetation status of their habitat. Based on a portion of the 18s rRNA gene sequence of 2 isolates, the Cryptosporidium found in Marmota flaviventris were 99.88%-100% match to multiple isolates of C. parvum in the GenBank.

The timing of reproduction is responding plastically, not genetically, to climate change in yellow-bellied marmots (<i>Marmota flaviventer</i>).

Project description:With global climates changing rapidly, animals must adapt to new environmental conditions with altered weather and phenology. The key to adapting to these new conditions is adjusting the timing of reproduction to maximize fitness. Using a long-term dataset on a wild population of yellow-bellied marmots (Marmota flaviventer) at the Rocky Mountain Biological Laboratory (RMBL), we investigated how the timing of reproduction changed with changing spring conditions over the past 50 years. Marmots are hibernators with a 4-month active season. It is thus crucial to reproduce early enough in the season to have time to prepare for hibernation, but not too early, as snow cover prevents access to food. Importantly, climate change in this area has, on average, increased spring temperatures by 5°C and decreased spring snowpack by 50 cm over the past 50 years. We evaluated how female marmots adjust the timing of their reproduction in response to changing conditions and estimated the importance of both microevolution and plasticity in the variation in this timing. We showed that, within a year, the timing of reproduction is not as tightly linked to the date a female emerges from hibernation as previously thought. We reported a positive effect of spring snowpack but not of spring temperature on the timing of reproduction. We found inter-individual variation in the timing of reproduction, including low heritability, but not in its response to changing spring conditions. There was directional selection for earlier reproduction since it increased the number and proportion of pups surviving their first winter. Taken together, the timing of marmot reproduction might evolve via natural selection; however, plastic changes will also be extremely important. Further, future studies on marmots should not operate under the assumption that females reproduce immediately following their emergence.
