Micro-Nano Plastic in the Aquatic Environment: Methodological Problems and Challenges.

ABSTRACT: Microplastic research has become a buzz word. It is seen as one of the most pressing issues of Anthropocene contamination. There is certainly no doubt about the ubiquitous presence of microplastic (MP) in almost all environmental matrices. However, the validity of considering them as a vector for contaminants needs some reconsideration, there are other more potent pathways. Their effect on marine biota also calls for some realistic experiments with environmental concentrations of MP and nanoplastic (NP). It has been observed that in most published literature, polymer characterization is performed. Is it necessary to do, or will merely finding and confirming the particle as plastic suffice for environmental research? Harmonization of protocols is necessary, and there is likely a need for some inter-laboratory comparison exercises in order to produce comparable data and reliable assessments across regions. Samples collected from the same area using different techniques show an order of magnitude difference in MP concentration. The issue of nanoplastic is more contentious; are we technologically ready to identify NP in environmental samples?

Project description:Recent studies have shown that plastic particles can sorb antibiotics, and these sorption properties have been examined in various studies; however, the possible mechanism responsible for the interactions requires a deeper investigation in terms of further interaction with living systems. Moreover, the usage of disposable surgical masks and plastic bottles has increased the plastic pollution risk for living systems like humans. Therefore, this study aimed to examine the sorption characteristics between antibiotics (amoxicillin and spiramycin) and plastic particles from surgical masks and plastic bottles through batch sorption experiments. In the study, their surface interactions were characterized using multispectroscopic approaches including FTIR, Raman spectrometry, and SEM-EDX, and various surface indicators (e.g., surface oxidation, deformation, and biological potential) were examined. The sorption results showed that adsorption kinetics and the isotherm of amoxicillin and spiramycin on micro(nano)plastics from surgical masks and plastic bottles closely fit the pseudo-second-order kinetic model and Langmiur isotherm. These results indicated that the evidence for the antibiotic interaction with particles was changes in the surface functional group intensities and up-shifting, and this correlated with the sorption of antibiotics on micro(nano)-sized plastics. The C/N ratio of the plastic particles before and after antibiotic treatment was used as an indicator for the surface biological interaction, and the results showed that C/N ratios of surgical mask particles increased with both types of antibiotic sorption. However, the C/N of the particles from plastic bottles showed antibiotic type-dependence. The surface deformation indicators (e.g., O/C, C=O, C=C, and O-H indices) showed that the O/C ratios of micro(nano)plastics from surgical masks were higher with the amoxicillin and spiramycin sorption, and the C=O indices were positively linked with the amoxicillin sorption stages, whereas the C=C and O-H had a negative correlation with the amoxicillin sorption stages. Moreover, amoxicillin sorption influenced the O/C ratio and indices of O-H and C=C of micro(nano)plastics from plastic bottles in a limited manner. The C=O groups of the micro(nano)plastics from plastic bottles were positively influenced by the spiramycin sorption stages, whereas it was negatively linked with amoxicillin sorption stages. Overall, the findings from surface indicators indicated that the micro(nano)plastics from surgical masks can be more influenced with antibiotic sorption compared to plastic bottles.

Project description:Reaction time testing is widely used in online computerized concussion assessments, and most concussion studies utilizing the metric have demonstrated varying degrees of difference between concussed and non-concussed individuals. The problem with most of these online concussion assessments is that they predominantly rely on consumer grade technology. Typical administration of these reaction time tests involves presenting a visual stimulus on a computer monitor and prompting the test subject to respond as quickly as possible via keypad or computer mouse. However, inherent delays and variabilities are introduced to the reaction time measure by both computer and associated operating systems that the concussion assessment tool is installed on. The authors hypothesized systems that are typically used to collect concussion reaction time data would demonstrate significant errors in reaction time measurements. To remove human bias, a series of experiments was conducted robotically to assess timing errors introduced by reaction time tests under four different conditions. In the first condition, a visual reaction time test was conducted by flashing a visual stimulus on a computer monitor. Detection was via photodiode and mechanical response was delivered via computer mouse. The second condition employed a mobile device for the visual stimulus, and the mechanical response was delivered to the mobile device's touchscreen. The third condition simulated a tactile reaction time test, and mechanical response was delivered via computer mouse. The fourth condition also simulated a tactile reaction time test, but response was delivered to a dedicated device designed to store the interval between stimulus delivery and response, thus bypassing any problems hypothesized to be introduced by computer and/or computer software. There were significant differences in the range of responses recorded from the four different conditions with the reaction time collected from visual stimulus on a mobile device being the worst and the device with dedicated hardware designed for the task being the best. The results suggest that some of the commonly used visual tasks on consumer grade computers could be (and have been) introducing significant errors for reaction time testing and that dedicated hardware designed for the reaction time task is needed to minimize testing errors.

Project description:We developed a Target Plastic Model (TPM) to estimate the critical plastic burden of organic toxicants in five types of plastics, namely, polydimethylsiloxane (PDMS), polyoxymethylene (POM), polyacrylate (PA), low-density polyethylene (LDPE), and polyurethane ester (PU), following the Target Lipid Model (TLM) framework. By substituting the lipid-water partition coefficient in the TLM with plastic-water partition coefficients to create TPM, we demonstrated that the biomimetic nature of these plastic phases allows for the calculation of critical plastic burdens of toxicants, similar to the notion of critical lipid burdens in TLM. Following this approach, the critical plastic burdens of baseline (n = 115), less-inert (n = 73), and reactive (n = 75) toxicants ranged from 0.17 to 51.33, 0.04 to 26.62, and 1.00 × 10-6 to 6.78 × 10-4 mmol/kg of plastic, respectively. Our study showed that PDMS, PA, POM, PE, and PU are similar to biomembranes in mimicking the passive exchange of chemicals with the water phase. Using the TPM, median lethal concentration (LC50) values for fish exposed to baseline toxicants were predicted, and the results agreed with experimental values, with RMSE ranging from 0.311 to 0.538 log unit. Similarly, for the same data set of baseline toxicants, other widely used models, including the TLM (RMSE: 0.32-0.34), ECOSAR (RMSE: 0.35), and the Abraham Solvation Model (ASM; RMSE: 0.31), demonstrated comparable agreement between experimental and predicted values. For less inert chemicals, predictions were within a factor of 5 of experimental values. Comparatively, ASM and ECOSAR showed predictions within a factor of 2 and 3, respectively. The TLM based on phospholipid had predictions within a factor of 3 and octanol within a factor of 4, indicating that the TPM's performance for less inert chemicals is comparable to these established models. Unlike these methods, the TPM requires only the knowledge of plastic bound concentration for a given plastic phase to calculate baseline toxic units, bypassing the need for extensive LC50 and plastic-water partition coefficient data, which are often limited for emerging chemicals. Taken together, the TPM can provide valuable insights into the toxicities of chemicals associated with environmental plastic phases, assisting in selecting the best polymeric phase for passive sampling and designing better passive dosing techniques for toxicity experiments.
