Plasmonic Enhancement of Two-Photon Excited Luminescence of Gold Nanoclusters.

ABSTRACT: Plasmonic-enhanced luminescence of single molecules enables imaging and detection of low quantities of fluorophores, down to individual molecules. In this work, we present two-photon excited luminescence of single gold nanoclusters, Au18(SG)14, in close proximity to bare gold nanorods (AuNRs). We observed 25-times enhanced emission of gold nanoclusters (AuNCs) in near infrared region, which was mainly attributed to the resonant excitation of localized surface plasmon resonance (LSPR) of AuNRs and spectral overlap of LSPR band with photoluminescence of AuNCs. This work is an initial step in application of combined nanoparticles: gold nanorods and ultrasmall nanoclusters in a wide range of multiphoton imaging and biosensing applications.

Plasmonic Enhancement of Two-Photon-Excited Luminescence of Single Quantum Dots by Individual Gold Nanorods.

Project description:Plasmonic enhancement of two-photon-excited fluorescence is not only of fundamental interest but also appealing for many bioimaging and photonic applications. The high peak intensity required for two-photon excitation may cause shape changes in plasmonic nanostructures, as well as transient plasmon broadening. Yet, in this work, we report on strong enhancement of the two-photon-excited photoluminescence of single colloidal quantum dots close to isolated chemically synthesized gold nanorods. Upon resonant excitation of the localized surface plasmon resonance, a gold nanorod can enhance the photoluminescence of a single quantum dot more than 10 000-fold. This strong enhancement arises from the combined effect of local field amplification and the competition between radiative and nonradiative decay rate enhancements, as is confirmed by time-resolved fluorescence measurements and numerical simulations.

Unlocking multi-photon excited luminescence in pyrazolate trinuclear gold clusters for dynamic cell imaging.

Project description:The family of coinage-metal-based cyclic trinuclear complexes exhibits abundant photophysical properties, promising for diverse applications. However, their utility in biochemistry is often hindered by large particle size and strong hydrophobicity. Meanwhile, the investigation into multi-photon excited luminescence within this family remained undocumented, limiting their potential in bio-imaging. Herein, we unveil the multi-photon excited luminescent properties of pyrazolate-based trinuclear gold(I) clusters, facilitated by excimeric gold(I)···gold(I) interactions, revealing a nonlinear optical phenomenon within this family. Furthermore, to address issues of poor biocompatibility, we employ electrospinning coupled with hydroxypropyl-beta-cyclodextrin as the matrix to fabricate a flexible, durable, transparent, and red emissive film with a photoluminescence quantum yield as high as 88.3%. This strategy not only produces the film with sufficient hydrophilicity and stability, but also achieves the downsizing of trinuclear gold(I) clusters from microscale to nanoscale. Following the instantaneous dissolution of the film in the media, the released trinuclear gold(I) nanoparticles have illuminated cells and bacteria through a real-time, non-toxic, multi-photon bio-imaging approach. This achievement offers a fresh approach for utilizing coinage-metal-based cyclic trinuclear complexes in biochemical fields.

Efficient Generation of Two-Photon Excited Phosphorescence from Molecules in Plasmonic Nanocavities.

Project description:Nonlinear molecular interactions with optical fields produce intriguing optical phenomena and applications ranging from color generation to biomedical imaging and sensing. The nonlinear cross-section of dielectric materials is low and therefore for effective utilisation, the optical fields need to be amplified. Here, we demonstrate that two-photon absorption can be enhanced by 108 inside individual plasmonic nanocavities containing emitters sandwiched between a gold nanoparticle and a gold film. This enhancement results from the high field strengths confined in the nanogap, thus enhancing nonlinear interactions with the emitters. We further investigate the parameters that determine the enhancement including the cavity spectral position and excitation wavelength. Moreover, the Purcell effect drastically reduces the emission lifetime from 520 ns to <200 ps, turning inefficient phosphorescent emitters into an ultrafast light source. Our results provide an understanding of enhanced two-photon-excited emission, allowing for optimization of efficient nonlinear light-matter interactions at the nanoscale.

Two-Photon-Excited Single-Molecule Fluorescence Enhanced by Gold Nanorod Dimers.

Project description:We demonstrate two-photon-excited single-molecule fluorescence enhancement by single end-to-end self-assembled gold nanorod dimers. We employed biotinylated streptavidin as the molecular linker, which connected two gold nanorods in end-to-end fashion. The typical size of streptavidin of around 5 nm separates the gold nanorods with gaps suitable for the access of fresh dyes in aqueous solution, yet small enough to give very high two-photon fluorescence enhancement. Simulations show that enhancements of more than 7 orders of magnitude can be achieved for two-photon-excited fluorescence in the plasmonic hot spots. With such high enhancements, we successfully detect two-photon-excited fluorescence for a common organic dye (ATTO 610) at the single-molecule, single-nanoparticle level.

Gold nanocages as contrast agents for two-photon luminescence endomicroscopy imaging.

Two-photon luminescence imaging of cancer cells using molecularly targeted gold nanorods.

Project description:We demonstrate the use of gold nanorods as bright contrast agents for two-photon luminescence (TPL) imaging of cancer cells in a three-dimensional tissue phantom down to 75 mum deep. The TPL intensity from gold-nanorod-labeled cancer cells is 3 orders of magnitude brighter than the two-photon autofluorescence (TPAF) emission intensity from unlabeled cancer cells at 760 nm excitation light. Their strong signal, resistance to photobleaching, chemical stability, ease of synthesis, simplicity of conjugation chemistry, and biocompatibility make gold nanorods an attractive contrast agent for two-photon imaging of epithelial cancer.

Molecular saturation determines distinct plasmonic enhancement scenarios for two-photon absorption signal.

Project description:Two-photon absorption in molecules, of significance for high-resolution imaging applications, is typically characterised with low cross sections. To enhance the TPA signal, one effective approach exploits plasmonic enhancement. For this method to be efficient, it must meet several criteria, including broadband operational capability and a high fluorescence rate to ensure effective signal detection. In this context, we introduce a plus-shaped silver nanostructure designed to exploit the coupling of bright and dark plasmonic modes. This configuration considerably improves both the absorption and fluorescence of molecules across near-infrared and visible spectra. By fine-tuning the geometrical parameters of the nanostructure, we align the plasmonic resonances with the optical properties of specific TPA-active dyes, i.e., ATTO 700, Rhodamine 6G, and ATTO 610. The expected TPA signal enhancement is evaluated using classical estimations based on the assumption of independent enhancement of absorption and fluorescence. These results are then compared with outcomes obtained in a quantum-mechanical approach to evaluate the stationary photon emission rate. Our findings reveal the important role of molecular saturation determining the regimes where either absorption or fluorescence enhancement leads to an improved TPA signal intensity, considerably below the classical predictions. The proposed nanostructure design not only addresses these findings, but also might serve for their experimental verification, allowing for active polarization tuning of the plasmonic response targeting the absorption, fluorescence, or both. The insight into quantum-mechanical mechanisms of plasmonic signal enhancement provided in our work is a step forward in the more effective control of light-matter interactions at the nanoscale.

Two-Photon Time-Gated In Vivo Imaging of Dihydrolipoic-Acid-Decorated Gold Nanoclusters.

Project description:Due to the unique advantages of two-photon technology and time-resolved imaging technology in the biomedical field, attention has been paid to them. Gold clusters possess excellent physicochemical properties and low biotoxicity, which make them greatly advantageous in biological imaging, especially for in vivo animal imaging. A gold nanocluster was coupled with dihydrolipoic acid to obtain a functionalized nanoprobe; the material displayed significant features, including a large two-photon absorption cross-section (up to 1.59 × 105 GM) and prolonged fluorescence lifetime (>300 ns). The two-photon and time-resolution techniques were used to perform cell imaging and in vivo imaging.

In vitro and in vivo two-photon luminescence imaging of single gold nanorods.

Project description:Gold nanorods excited at 830 nm on a far-field laser-scanning microscope produced strong two-photon luminescence (TPL) intensities, with a cos(4) dependence on the incident polarization. The TPL excitation spectrum can be superimposed onto the longitudinal plasmon band, indicating a plasmon-enhanced two-photon absorption cross section. The TPL signal from a single nanorod is 58 times that of the two-photon fluorescence signal from a single rhodamine molecule. The application of gold nanorods as TPL imaging agents is demonstrated by in vivo imaging of single nanorods flowing in mouse ear blood vessels.

Gold Nanoclusters: Bridging Gold Complexes and Plasmonic Nanoparticles in Photophysical Properties.

Project description:Recent advances in the determination of crystal structures and studies of optical properties of gold nanoclusters in the size range from tens to hundreds of gold atoms have started to reveal the grand evolution from gold complexes to nanoclusters and further to plasmonic nanoparticles. However, a detailed comparison of their photophysical properties is still lacking. Here, we compared the excited state behaviors of gold complexes, nanolcusters, and plasmonic nanoparticles, as well as small organic molecules by choosing four typical examples including the Au10 complex, Au25 nanocluster (1 nm metal core), 13 diameter Au nanoparticles, and Rhodamine B. To compare their photophysical behaviors, we performed steady-state absorption, photoluminescence, and femtosecond transient absorption spectroscopic measurements. It was found that gold nanoclusters behave somewhat like small molecules, showing both rapid internal conversion (<1 ps) and long-lived excited state lifetime (about 100 ns). Unlike the nanocluster form in which metal-metal transitions dominate, gold complexes showed significant charge transfer between metal atoms and surface ligands. Plasmonic gold nanoparticles, on the other hand, had electrons being heated and cooled (~100 ps time scale) after photo-excitation, and the relaxation was dominated by electron-electron scattering, electron-phonon coupling, and energy dissipation. In both nanoclusters and plasmonic nanoparticles, one can observe coherent oscillations of the metal core, but with different fundamental origins. Overall, this work provides some benchmarking features for organic dye molecules, organometallic complexes, metal nanoclusters, and plasmonic nanoparticles.
