Coffee Beverage: A New Strategy for the Synthesis of Polymethacrylates via ATRP.

ABSTRACT: Coffee, the most popular beverage in the 21st century society, was tested as a reaction environment for activators regenerated by electron transfer atom transfer radical polymerization (ARGET ATRP) without an additional reducing agent. Two blends were selected: pure Arabica beans and a proportional blend of Arabica and Robusta beans. The use of the solution received from the mixture with Robusta obtained a high molecular weight polymer product in a short time while maintaining a controlled structure of the synthesized product. Various monomers with hydrophilic characteristics, i.e., 2-(dimethylamino)ethyl methacrylate (DMAEMA), oligo(ethylene glycol) methyl ether methacrylate (OEGMA500), and glycidyl methacrylate (GMA), were polymerized. The proposed concept was carried out at different concentrations of coffee grounds, followed by the determination of the molar concentration of caffeine in applied beverages using DPV and HPLC techniques.

Fast Bulk Depolymerization of Polymethacrylates by ATRP.

Project description:Fast bulk depolymerization of poly(n-butyl methacrylate) and poly(methyl methacrylate), prepared by atom transfer radical polymerization (ATRP), is reported in the temperature range between 150 and 230 °C. Depolymerization of Cl-terminated polymethacrylates was catalyzed by a CuCl2/TPMA complex (0.022 or 0.22 equiv vs P-Cl) and was studied using TGA, also under isothermal conditions. Relatively rapid 5-20 min depolymerization was observed at 230 and 180 °C. The preparative scale reactions were carried out using a short-path distillation setup with up to 84% depolymerization within 15 min at 230 °C.

A Novel Strategy for the Synthesis of Amphiphilic and Thermoresponsive Poly(N-isopropylacrylamide)-<i>b</i>-Polystyrene Block Copolymers via ATRP.

Project description:A new synthetic approach is presented for the preparation of Poly(N-isopropylacrylamide-block-styrene) PNIPAM-b-PS via an Atom Transfer Radical Polymerization (ATRP) technique. The proposed method is based on application of 2-chloro-N-(2-hydroxyethyl)propanamide (NCPAE) as a bifunctional initiator, which enables ATRP of two monomers, differing in activity and polarity, into two stages. The synthesized copolymer molecules contain two well-defined polymer chains connected by a linker, which is a derivative of the proposed initiator. Using NCPAE led to PNIPAMs with well-planned molecular weight, low polydispersities (PDI=1.1÷1.3) and hydroxyl functionality. Activation of such blocks for initiation of styrene polymerization was performed using α-bromoisobutyryl bromide. After such a modification, the synthesized homopolymers acted as macroinitiators in ARGET ATRP and a well-defined polystyrene block, as the next one in the polymer chain was successfully formed. Both of the synthesized macromolecules, PNIPAM and PNIPAM-b-PS, exhibit a thermoresponsive behavior with explicit lower critical solution temperatures (LCST) in their aqueous solutions. The synthesized homopolymers and subsequently derived block copolymers were characterized using Size-Exclusion Chromatography, Differential Scanning Calorimetry, Dynamic Light Scattering, and NMR spectroscopy.

ATRP Synthesis of Sunflower Polymers using Cyclic Multimacroinitiators.

Project description:Polymers with advanced architectures can now be readily and reproducibly synthesized using controlled living polymerization. These materials are attractive as potential drug carriers due to their tunable size, versatile methods of drug incorporation and release, and ease of functionalization with targeting ligands. In this work, we report the design and development of macrocyclic brush, or "sunflower," polymers, synthesized by controlled radical polymerization of hydrophilic "petals" from a cyclic multimacroinitiator "core." These nanostructures can be synthesized with low polydispersity and controlled sizes depending on polymerization time. We further demonstrate that folate-functionalized sunflower polymers facilitate receptor-mediated uptake into cancer cells. These materials therefore show potential as drug carriers for anti-cancer therapies.

New Photoresponsive Poly(meth)acrylates Bearing Azobenzene Moieties Obtained via ATRP Polymerization Exhibiting Liquid-Crystalline Behavior.

Project description:Here, we report our studies on photoresponsive poly(meth)acrylates containing azobenzene groups connected to a polymer backbone via a short methylene linker. A series of side-chain azobenzene polymers was synthesized via the atom transfer radical polymerization (ATRP) technique using several catalytic systems. The polymers synthesized under the optimized conditions were characterized by a narrow polydispersity (Đ ≤ 1.35), and they underwent a reversible transformation of their structure under light illumination. The fabricated polymers can store and release energy accumulated during the UV-light illumination by the thermal cis-trans isomerization of the chromophore groups. The enthalpy of the process (determined from DSC) was relatively high and equaled 61.9 J∙g-1 (17 Wh∙kg-1), indicating a high potential of these materials in energy storage applications. The liquid-crystalline behavior of the synthesized poly(meth)acrylates was demonstrated by the birefringent optical textures as observed in thin-films and X-ray scattering studies.

New Coffee Plant-Infecting Xylella fastidiosa Variants Derived via Homologous Recombination.

Project description:Xylella fastidiosa is a xylem-limited phytopathogenic bacterium endemic to the Americas that has recently emerged in Asia and Europe. Although this bacterium is classified as a quarantine organism in the European Union, importation of plant material from contaminated areas and latent infection in asymptomatic plants have engendered its inevitable introduction. In 2012, four coffee plants (Coffea arabica and Coffea canephora) with leaf scorch symptoms growing in a confined greenhouse were detected and intercepted in France. After identification of the causal agent, this outbreak was eradicated. Three X. fastidiosa strains were isolated from these plants, confirming a preliminary identification based on immunology. The strains were characterized by multiplex PCR and by multilocus sequence analysis/typing (MLSA-MLST) based on seven housekeeping genes. One strain, CFBP 8073, isolated from C. canephora imported from Mexico, was assigned to X. fastidiosa subsp. fastidiosa/X. fastidiosa subsp. sandyi. This strain harbors a novel sequence type (ST) with novel alleles at two loci. The two other strains, CFBP 8072 and CFBP 8074, isolated from Coffea arabica imported from Ecuador, were allocated to X. fastidiosa subsp. pauca. These two strains shared a novel ST with novel alleles at two loci. These MLST profiles showed evidence of recombination events. We provide genome sequences for CFBP 8072 and CFBP 8073 strains. Comparative genomic analyses of these two genome sequences with publicly available X. fastidiosa genomes, including the Italian strain CoDiRO, confirmed these phylogenetic positions and provided candidate alleles for coffee plant adaptation. This study demonstrates the global diversity of X. fastidiosa and highlights the diversity of strains isolated from coffee plants.

Synthesis of naturally-derived macromolecules through simplified electrochemically mediated ATRP.

Project description:The flavonoid-based macroinitiator was received for the first time by the transesterification reaction of quercetin with 2-bromoisobutyryl bromide. In accordance with the "grafting from" strategy, a naturally-occurring star-like polymer with a polar 3,3',4',5,6-pentahydroxyflavone core and hydrophobic poly(tert-butyl acrylate) (PtBA) side arms was synthesized via a simplified electrochemically mediated ATRP (seATRP), utilizing only 78 ppm by weight (wt) of a catalytic CuII complex. To demonstrate the possibility of temporal control, seATRP was carried out utilizing a multiple-step potential electrolysis. The rate of the polymerizations was well-controlled by applying optimal potential values during preparative electrolysis to prevent the possibility of intermolecular coupling of the growing polymer arms. This appears to be the first report using on-demand seATRP for the synthesis of QC-(PtBA-Br)5pseudo-star polymers. The naturally-derived macromolecules showed narrow MWDs (Đ = 1.08-1.11). 1H NMR spectral results confirm the formation of quercetin-based polymers. These new flavonoid-based polymer materials may find applications as antifouling coatings and drug delivery systems.

Extraction of polyphenols and synthesis of new activated carbon from spent coffee grounds.

Project description:A valorization process of spent coffee grounds (SCG) was studied. Thus, a two-stage process, the first stage of polyphenols extraction and synthesis of a carbonaceous precursor and a subsequent stage of obtaining activated carbon (AC) by means of a carbonization process from the precursor of the previous stage, was performed. The extraction was carried out with a hydro-alcoholic solution in a pressure reactor, modifying time, temperature and different mixtures EtOH:H2O. To optimize the polyphenols extraction, a two-level factorial experimental design with three replicates at the central point was used. The best results were obtained by using a temperature of 80 °C during 30 min with a mixture of EtOH:H2O 50:50 (v/v). Caffeine and chlorogenic acid were the most abundant compounds in the analysed extracts, ranging from 0.09 to 4.8 mg∙g-1 and 0.06 to 9.7 mg∙g-1, respectively. Similarly, an experimental design was realized in order to analyze the influence of different variables in the AC obtained process (reaction time, temperature and KOH:precursor ratio). The best results were 1 h, 850 °C, and a mixture of 2.5:1. The obtained activated carbons exhibit a great specific surface (between 1600 m2∙g-1 and 2330 m2∙g-1) with a microporous surface. Finally, the adsorption capacity of the activated carbons was evaluated by methylene blue adsorption.

Synthesis of pH-Sensitive and Self-Fluorescent Polymeric Micelles Derived From Rosin and Vegetable Oils <i>via</i> ATRP.

Project description:Preparation and application of sustainable polymers derived from renewable resources are of great significance. The aim of this study is to synthesize a kind of sustainable polymeric micelles from rosin and vegetable oils via atom transfer radical polymerization (ATRP) and to investigate the doxorubicin delivery properties of these micelles. Dehydroabietic acid-based poly lauryl methacrylate (DA-PLMA) with narrow PDI of 1.13 was prepared in a well-controlled process using rosin as an ATRP initiator. Thereafter, carboxylic groups were introduced to form poly methacrylic acid (PMAA) moieties in DA-PLMA polymer via acid hydrolysis. The resulted DA-PLMA-PMAA could self-assemble in water to form pH-dependent polymeric micelles with a diameter of ∼65 nm and PDI as low as 0.105. Owing to the existence of rosin, DA-PLMA-PMAA micelles also showed self-fluorescence properties. In addition, Dox-loaded micelles were prepared in aqueous solution with the drug-loading capacity as high as 16.0% and showed sustained-release characteristics. These results demonstrate great promise for designing polymeric micellar from rosin and vegetable oils.

New strategy for the synthesis of substituted morpholines.

Project description:A four-step synthesis of cis-3,5-disubstituted morpholines from enantiomerically pure amino alcohols is described. The key step in the synthesis is a Pd-catalyzed carboamination reaction between a substituted ethanolamine derivative and an aryl or alkenyl bromide. The morpholine products are generated as single stereoisomers in moderate to good yield. This strategy also provides access to fused bicyclic morpholines as well as 2,3- and 2,5-disubstituted products.

A new strategy for the synthesis of chiral β-alkynyl esters via sequential palladium and copper catalysis.

Project description:A new strategy for the synthesis of chiral β-alkynyl esters which relies on sequential Pd and Cu catalysis is reported. Terminal alkynes bearing aryl, alkyl, and silyl groups can be employed without prior activation yielding a wide range of important chiral building blocks. The reaction sequence utilizes a robust Pd(II)-catalyzed hydroalkynylation of ynoates with terminal alkynes providing geometrically pure ynenoates which are readily reduced by CuH. In contrast to previous reports, where additions to ynenoates proceed with marginal preference for the 1,6-pathway, this conjugate reduction occurs with high 1,4-selectivity yielding β-alkynyl esters with excellent levels of enantioselectivity. Importantly, the method tolerates a wide range of functionality, including allylic carbonates and carbamates, and thus allows for rapid elaboration of the β-alkynyl esters into a variety of chiral, substituted heterocycles.
