Succession Pattern in Soil Micro-Ecology Under Tobacco (Nicotiana tabacum L.) Continuous Cropping Circumstances in Yunnan Province of Southwest China.

ABSTRACT: Continuous cropping obstacle (CCO) is a common phenomenon in agricultural production and extremely threatens the sustainable development of agriculture. To clarify the potential keystone factors causing tobacco (Nicotiana tabacum L.) CCO, tobacco plants, topsoil, and rhizosphere soil were sampled from the fields with no, slight, and severe tobacco disease in Dali and Yuxi of Yunnan province in China. The physicochemical properties of topsoil and rhizosphere soil, the phenolic acids (PAs) contents in rhizosphere soil, and elemental contents in topsoil, rhizosphere soil, and tobacco plants were analyzed. Microbial diversity in rhizosphere soil was determined by the metagenomic sequencing method. The results showed that soil pH, texture, cation exchange capacity, organic matter, TC, TN, and available K contents showed a significant difference (p < 0.05) in soil physicochemical properties. There was a deficiency of B, K, Mg, and Mn contents in soil and/or tobacco plants. The contents of PAs, especially syringic acid in rhizosphere soil, varied significantly among the three sampling groups (p < 0.05). Meanwhile, microbial communities and functional genes changed from beneficial to harmful, showing an intimate correlation with soil pH and syringic acid content. It can be concluded that tobacco CCO could be allocated to the imbalance of soil micro-ecology, which possessed a regional feature at the two sampling sites.

Project description:Yunnan Province, China is thought to be the original source of biovar Orientalis of Yersinia pestis, the causative agent of the third plague pandemic that has spread globally since the end of the 19th century. Although encompassing a large area of natural plague foci, Y. pestis strains have rarely been found in live rodents during surveillance in Yunnan, and most isolates are from rodent corpses and their fleas. In 2017, 10 Y. pestis strains were isolated from seven live rodents and three fleas in Heqing County (HQ) of Yunnan. These strains were supposed to have low virulence to local rodents Eothenomys miletus and Apodemus chevrieri because the rodents were healthy and no dead animals were found in surrounding areas, as had occurred in previous epizootic disease. We performed microscopic and biochemical examinations of the isolates,and compared their whole-genome sequences and transcriptome with those of 10 high virulence Y. pestis strains that were isolated from the adjacent city (Lijiang). We analyzed the phenotypic, genomic, and transcriptomic characteristics of live rodent isolates. The isolates formed a previously undefined monophyletic branch of Y. pestis that was named 1.IN5. Six SNPs, two indels, and one copy number variation were detected between live rodent isolates and the high virulence neighbors. No obvious functional consequence of these variations was found according to the known annotation information. Among the genes that were differentially expressed between the live rodent isolates and their high virulence neighbors, we detected five iron transfer-related genes that were significantly up-regulated in live rodent isolates compared with high virulence isolates (|log2 (FC) | >1, p.adjust <0.05), indicating these genes may be related to the low-virulence phenotype. The novel genotype of Y. pestis reported here provides further insights into the evolution and spread of plague as well as clues that may help to decipher the virulence mechanism of this notorious pathogen.

Project description:Currently, natural restoration has been widely proposed as the primary method of ecological restoration and has been studied for a long time. However, research on how to quantify the progress of natural restoration in different climate conditions, especially using long-term succession monitoring data combined with habitat quality data across various succession stages, has been scarce. Our study aims to address this issue in Yunnan, southwest China. To quantify the progress of natural restoration under different climates in Yunnan, we introduced an index, the Natural Succession Index. Utilizing topography and meteorological data, we divided the study area into different climate sub-areas using the Two-stage clustering algorithm. We then combined 1703 sets of 30-year succession monitoring data, each with six observations taken at five-year intervals from 1987 to 2017, with habitat quality data from different succession stages (grassland, shrub, and forest) to quantify the Natural Succession Index. Yunnan province was divided into 14 sub-areas, namely C(I to II), M(I to III), W(I to IV), and H(I to IV), each possessing a unique environment. The indices in each sub-area were calculated, with the results showing a specific order: H-I (0.7812) > H-IV (0.7739) > W-I (0.6498) > M-III (0.6356) > H-III (0.6316) > M-II (0.5735) > W-III (0.5644) > W-IV (0.5571) > C-II (0.4778) > W-II (0.3980) > M-I (0.3624) > H-II (0.3375) > C-I (0.2943). The times for natural succession to reach the forest stage vary from 5 to 19 years, which aligns with the order of indices. The stand volumes of vegetation in the forest stage range from 5 m³ to 110 m³, with a higher Natural Succession Index value corresponding to a higher stand volume of vegetation. In the future, the index could be utilized to reallocate investments in natural restoration projects for better returns. Constant vigilance is required in the first five years following the implementation of restoration actions to avoid failure due to calculation errors.
