Complete Genome Sequence of Streptomyces Phage ϕRKBJ001 Isolated from Prince Edward Island, Canada.

ABSTRACT: We reported here the complete genome sequence of Streptomyces phage ϕRKBJ001 that was isolated from a saltwater marsh on Prince Edward Island, Canada, using the Streptomyces sp. strain RKBHB0173. Based on electron microscopy and genomic analysis, this phage belongs to the Siphoviridae family and the BN Streptomyces phage cluster.

Project description:Streptomyces sp. strain RKND-216 was isolated from marine sediment collected in Prince Edward Island, Canada, and produces a putatively novel bioactive natural product with antitubercular activity. The genome assembly consists of two contigs covering 5.61 Mb. Genome annotation identified 4,618 predicted protein-coding sequences and 19 predicted natural product biosynthetic gene clusters.

Molecular diversity of methanogens in feedlot cattle from Ontario and Prince Edward Island, Canada.

Project description:The molecular diversity of rumen methanogens in feedlot cattle and the composition of the methanogen populations in these animals from two geographic locations were investigated using 16S rRNA gene libraries prepared from pooled PCR products from 10 animals in Ontario (127 clones) and 10 animals from Prince Edward Island (114 clones). A total of 241 clones were examined, with Methanobrevibacter ruminantium accounting for more than one-third (85 clones) of the clones identified. From these 241 clones, 23 different 16S rRNA phylotypes were identified. Feedlot cattle from Ontario, which were fed a corn-based diet, revealed 11 phylotypes (38 clones) not found in feedlot cattle from Prince Edward Island, whereas the Prince Edward Island cattle, which were fed potato by-products as a finishing diet, had 7 phylotypes (42 clones) not found in cattle from Ontario. Five sequences, representing the remaining 161 clones (67% of the clones), were common in both herds. Of the 23 different sequences, 10 sequences (136 clones) were 89.8 to 100% similar to those from cultivated methanogens belonging to the orders Methanobacteriales, Methanomicrobiales, and Methanosarcinales, and the remaining 13 sequences (105 clones) were 74.1 to 75.8% similar to those from Thermoplasma volcanium and Thermoplasma acidophilum. Overall, nine possible new species were identified from the two clone libraries, including two new species belonging to the order Methanobacteriales and a new genus/species within the order Methanosarcinales. From the present survey, it is difficult to conclude whether the geographical isolation between these two herds or differences between the two finishing diets directly influenced community structure in the rumen. Further studies are warranted to properly assess the differences between these two finishing diets.

Shaeky has very low sequence identity to other phages, with phage phiC31 being the most closely related in the NCBI database. The Shaeky genome is 45,617 bp with 77 protein-coding genes and 16 tRNAs.

Terrosamycins A and B, Bioactive Polyether Ionophores from Streptomyces sp. RKND004 from Prince Edward Island Sediment.

Project description:Terrosamycins A (1) and B (2), two polycyclic polyether natural products, were purified from the fermentation broth of Streptomyces sp. RKND004 isolated from Prince Edward Island sediment. The one strain-many compounds (OSMAC) approach coupled with UPLC-HRMS-based metabolomics screening led to the identification of these compounds. The structure of 1 was determined from analysis of NMR, HRMS, and X-ray diffraction data. NMR experiments performed on 2 revealed the presence of two methoxy groups replacing two hydroxy groups in 1. Like other polyether ionophores, 1 and 2 exhibited excellent antibiotic activity against Gram-positive pathogens. Interestingly, the terrosamycins also exhibited activity against two breast cancer cell lines.

Assessing Future Climate Change Impacts on Potato Yields - A Case Study for Prince Edward Island, Canada.

Project description:Crop yields are adversely affected by climate change; therefore, it is crucial to develop climate adaptation strategies to mitigate the impacts of increasing climate variability on the agriculture system to ensure food security. As one of the largest potato-producing provinces in Canada, Prince Edward Island (PEI) has recently experienced significant instability in potato production. PEI's local farmers and stakeholders are extremely concerned about the prospects for the future of potato farming industries in the context of climate change. This study aims to use the Decision Support System for Agrotechnology Transfer (DSSAT) potato model to simulate future potato yields under the Coupled Model Intercomparison Project Phase 6 (CMIP6) climate scenarios (including SSP1-1.9, SSP1-2.6, SSP2-4.5, SSP3-7.0, and SSP5-8.5). The study evaluates the combined effects of changing climatic conditions at local scales (i.e., warming temperature and changing precipitation patterns) and increasing carbon dioxide (CO2) concentration in the atmosphere. The results indicate future significant declines in potato yield in PEI under the current farming practices. In particular, under the high-emission scenarios (e.g., SSP3-7.0 and SSP5-8.5), the potato yield in PEI would decline by 48% and 60% in the 2070s and by 63% and 80% by 2090s; even under the low-emission scenarios (i.e., SSP1-1.9 and SSP1-2.6), the potato yield in PEI would still decline by 6-10%. This implies that it is important to develop effective climate adaptation measures (e.g., adjusting farming practices and introducing supplemental irrigation plans) to ensure the long-term sustainability of potato production in PEI.
