Silver nanotriangle array based LSPR sensor for rapid coronavirus detection.

ABSTRACT: A rapid, portable, and cost-effective method to detect the infection of SARS-CoV-2 is fundamental toward mitigating the current COVID-19 pandemic. Herein, a human angiotensin-converting enzyme 2 protein (ACE2) functionalized silver nanotriangle (AgNT) array localized surface plasmon resonance (LSPR) sensor is developed for rapid coronavirus detection, which is validated by SARS-CoV-2 spike RBD protein and CoV NL63 virus with high sensitivity and specificity. A linear shift of the LSPR wavelength versus the logarithm of the concentration of the spike RBD protein and CoV NL63 is observed. The limits of detection for the spike RBD protein, CoV NL63 in buffer and untreated saliva are determined to be 0.83 pM, 391 PFU/mL, and 625 PFU/mL, respectively, while the detection time is found to be less than 20 min. Thus, the AgNT array optical sensor could serve as a potential rapid point-of-care COVID-19 diagnostic platform.

Detection of nitrite with a surface-enhanced Raman scattering sensor based on silver nanopyramid array.

Project description:Nutrient pollution is of worldwide environmental and health concerns due to extensive use of nitrogen fertilizers and release of livestock waste, which induces nitrite compounds in aquatic systems. Herein a surface-enhanced Raman scattering (SERS) sensor is developed for nitrite detection based on coupling between the plasmonic gold nanostars and the silver nanopyramid array. When nitrite is present in the assay, an azo group is formed between the 1-naphthylamine-functionalized silver nanopyramids and the 4-aminothiophenol-functionalized gold nanostars. This not only generates the SERS spectral fingerprint for selective detection, but also creates "hot spots" at the gap between the Au nanostars and the Ag nanopyramids where the azo group is located, amplifying SERS signals remarkably. Finite-difference time-domain (FDTD) simulation shows a SERS enhancement factor of 4 × 1010 at the "hot spots". As a result, the SERS sensor achieves a limit of detection of 0.6 pg/mL toward nitrite in water, and enables nitrite detection in real-world river water samples. In addition, this sensor eliminates the use of any Raman reporter and any expensive molecular recognition probe such as antibody and aptamer. This highly sensitive, selective and inexpensive SERS sensor has unique advantages over colorimetric, electrochemical and fluorescent devices for small molecule detection.

LSPR-Based Aptasensor for Rapid Urinary Detection of NT-proBNP.

Project description:N-terminal pro-brain natriuretic peptide (NT-proBNP) is a myocardial stress biomarker that can be found in serum or plasma, saliva, and urine in the context of cardiovascular disease. In this study, we developed a rapid (~25 min) and straightforward localized surface plasmon resonance (LSPR)-based assay for detecting NT-proBNP in urine. The assay employs citrate-capped gold nanoparticles (AuNPs) and an aptamer specific for NT-proBNP, which initially interacts with NT-proBNP. The remaining unbound aptamer then interacts with the AuNPs, and the addition of NaCl induces the aggregation of the unprotected AuNPs, resulting in a decrease in absorbance at the LSPR band (A521) and an increase in absorbance at 750 nm (A750). The concentration of NT-proBNP showed a linear correlation with the aggregation ratio (A521/A750), and the assay demonstrated a limit of detection (LOD) of 0.303 µg·L-1 and a detection range of 0.566-8 µg·L-1. However, the presence of sulfur-containing proteins in saliva and fetal bovine serum hindered the detection of NT-proBNP in these biofluids. Nevertheless, the assay successfully detected NT-proBNP in diluted urine with an LOD of 0.417 µg·L-1 and a detection range of 0.589-6 µg·L-1. The observed values in urine samples from preterm infants with cardiovascular disease fell within this range, indicating the potential clinical relevance of the assay. The recovery percentages ranged from 92.3 to 116.3%. Overall, our findings suggest that the LSPR-based assay for NT-proBNP detection in urine can be a valuable tool for the diagnosis and treatment of cardiovascular disease.

Silver Nanoparticle Sensor Array-Based Meat Freshness Inspection System.

Project description:The series of biochemical reactions, metabolic pathways, and regulatory interactions that occur during the storage of meat are the main causes of meat loss and waste. The volatile compounds produced by these reactions, such as hydrogen sulfide, acids, and amines, can directly indicate changes in the freshness of meat during storage and sales. In this study, a one-pot hydrothermal method based on a surface control strategy was used to develop nanoparticles of silver with different reactivities, which were further immobilized in agar powder to develop a colorimetric sensor array. Due to the different chemical interactions with various volatile compounds, the colorimetric sensor array exhibited distinct color changes. The study demonstrates significant differences between 12 different volatile compounds and provides a quantitative and visual method to reveal rich detection indicators. The colorimetric sensor array is an economical and practical multi-analyte identification method. It has many potential applications such as food packaging, anti-counterfeiting, health monitoring, environmental monitoring, and optical filters.

High-sensitivity qPCR detection method based on silver flower-like LSPR-active material.

Project description:Quantitative polymerase chain reaction (qPCR) technology offers advantages such as easy operation and high specificity, but also faces issues like lengthy detection time and low sensitivity. This paper proposes a method by using novel SERS-active silver flower-like material to enhance the fluorescence signal generated by qPCR instrument, so as to shorten the detection time and improve the detection sensitivity. In this method, silver flower-like material is first prepared on substrates using a lyotropic liquid crystal soft template combined with electrodeposition growth. Then, an ultrasonic device is used to peel the active material into aqueous solution, and the peeling efficiency reaches 90%. Using rhodamine 6G (R6G) as the qPCR amplification detection molecule, the fluorescence signal generated from the flower-like material-containing sample is increased by 20%. Finally, the qPCR amplification effect is detected with the commercially available new coronavirus nucleic acid detection kit. The results showed that the CT value decreased from 35 to 31 after adding the silver flower-like material, and the detection efficiency increased by 20%. Meanwhile, no non-specific amplification is observed. For COVID-19 samples at S7 concentration, the positive detection rate rises from 1/200 to 2/3 by our method.

Label-free detection of DNA hybridization with a compact LSPR-based fiber-optic sensor.

Project description:A miniaturized, robust, localized surface plasmon resonance (LSPR)-coupled fiber-optic (FO) nanoprobe providing an integrated and portable solution for detection of DNA hybridization and measurement of DNA concentrations has been demonstrated. The FO nanoprobe was created by constructing arrays of metallic nanostructures on the end facets of optical fibers utilizing nanofabrication technologies, including electron beam lithography and lift-off processes. The LSPR-FO nanoprobe device offers real-time, label-free, and low-sample-volume quantification of single-strand DNA in water with high sensitivity and selectivity, achieving a limit of detection around 10 fM. These results demonstrate the feasibility of the LSPR-FO nanoprobe device as a compact and low-cost biosensor for detection of short-strand DNA.

Conductive Silver/Carbon Fiber Films for Rapid Detection of Human Coronavirus.

Project description:Polymerase chain reaction has gained attention since the outbreak of novel coronavirus in 2019. Due to its high specificity and capability for early detection, it is considered a standard method for the diagnosis of infectious diseases. However, the conventional thermocyclers used for nucleic acid amplification are not suitable for point-of-care testing applications, as they require expensive instruments, high-power consumption, and a long turnaround time. To suppress the widespread of the pandemic, there is an urgent need for the development of a rapid, inexpensive, and portable thermal cycler. Therefore, in this paper, we present a conductive silver/carbon fiber film-based thermal cycler with low power consumption (<5 W), efficient heating (~4.5 °C/s), low cost (<USD 200), and handheld size (11.5 × 7.1 × 7.5 mm). The conductive film, which was used as a heating source of the thermal cycler, was fabricated by the electrochemical deposition method. The successful coating of Ag was characterized by a scanning electron microscope and confirmed by energy-dispersive X-ray spectroscopy. The film showed excellent electrical/thermal conductivity and durability. Using our thermal cycler, 35 cycles of amplification were accomplished within 10 min. We also successfully demonstrated the multiplexed detection of various human coronaviruses (e.g., OC43, 229E, and NL63) using our thermal cycler.

A Novel Minidumbbell DNA-Based Sensor for Silver Ion Detection.

Project description:Silver ion (Ag+) is one of the most common heavy metal ions that cause environmental pollution and affect human health, and therefore, its detection is of great importance in the field of analytical chemistry. Here, we report an 8-nucleotide (nt) minidumbbell DNA-based sensor (M-DNA) for Ag+ detection. The minidumbbell contained a unique reverse wobble C·C mispair in the minor groove, which served as the binding site for Ag+. The M-DNA sensor could achieve a detection limit of 2.1 nM and sense Ag+ in real environmental samples with high accuracy. More importantly, the M-DNA sensor exhibited advantages of fast kinetics and easy operation owing to the usage of an ultrashort oligonucleotide. The minidumbbell represents a new and minimal non-B DNA structural motif for Ag+ sensing, allowing for the further development of on-site environmental Ag+ detection devices.

Cuvette-Type LSPR Sensor for Highly Sensitive Detection of Melamine in Infant Formulas.

Project description:The globalization of food distribution has made necessary to secure safe products to the general consumers through the rapid detection of harmful additives on the field. For this purpose, we developed a cuvette-type localized surface plasmon resonance (LSPR) sensor that can be easily used by consumers with conventional ultraviolet-visible light spectrophotometer for in-situ measurements. Gold nanoparticles were uniformly deposited on a transparent substrate via a self-assembly method to obtain a plasmonically active chip, and the chemical receptor p-nitroaniline (p-NA) was functionalized to stabilize the device sensitivity under external temperature and pH conditions. The fabricated chip was fixed onto a support and combined with a cuvette-type LSPR sensor. To evaluate the applicability of this sensor on the field, sensitivity and quantitative analysis experiments were conducted onto melamine as a model sample from harmful food additives. Under optimal reaction condition (2 mM p-NA for 20 min), we achieved an excellent detection limit (0.01 ppb) and a dynamic range allowing quantitative analysis over a wide concentration range (0.1-1000 ppb) from commercially available milk powder samples.

Silver Nanoparticle-Based Sensor for the Selective Detection of Nickel Ions.

Project description:Silver nanoparticles (AgNPs) can be used as a surface plasmon resonance (SPR) colorimetric sensor; the correlation between the SPR phenomenon and the aggregation state of nanoparticle allows the real-time detection of a target molecule. Surface functionalization of NPs with proper molecular baits is often performed to establish the selectivity of the sensor. This work reports on the synthesis of AgNPs under reducing conditions and on the functionalization thereof with mercaptoundecanoic acid (11-MUA). UV-VIS Spectroscopy confirmed the formation of AgNPs, eliciting a surface plasmon absorption band (SPAB) at 393 nm that shifted to 417 nm upon surface coating. Dynamic light scattering was used to investigate the surface coatings; moreover, pelleted AgNPs@11MUA nanoparticles were characterized by scanning electron microscopy (SEM), energy dispersive X-ray analyzers (EDX), and infrared spectroscopy to corroborate the presence of 11MUA on the surface. Most interestingly, the resulting AgNPs@11MUA selectively detected micromolar levels of Ni2+, also in the presence of other cations such as Mn2+, Co2+, Cd2+, Cu2+, Zn2+, Fe2+, Hg2+, Pb2+, and Cr3+.

Metal Oxide Nanorods-Based Sensor Array for Selective Detection of Biomarker Gases.

Project description:The breath gas analysis through gas phase chemical analysis draws attention in terms of non-invasive and real time monitoring. The array-type sensors are one of the diagnostic methods with high sensitivity and selectivity towards the target gases. Herein, we presented a 2 × 4 sensor array with a micro-heater and ceramic chip. The device is designed in a small size for portability, including the internal eight-channel sensor array. In2O3 NRs and WO3 NRs manufactured through the E-beam evaporator's glancing angle method were used as sensing materials. Pt, Pd, and Au metal catalysts were decorated for each channel to enhance functionality. The sensor array was measured for the exhaled gas biomarkers CH3COCH3, NO2, and H2S to confirm the respiratory diagnostic performance. Through this operation, the theoretical detection limit was calculated as 1.48 ppb for CH3COCH3, 1.9 ppt for NO2, and 2.47 ppb for H2S. This excellent detection performance indicates that our sensor array detected the CH3COCH3, NO2, and H2S as biomarkers, applying to the breath gas analysis. Our results showed the high potential of the gas sensor array as a non-invasive diagnostic tool that enables real-time monitoring.
