Clinical Tick-Borne Encephalitis in a Roe Deer (Capreolus capreolus L.).

ABSTRACT: Tick-borne encephalitis virus (TBEV) is the causative agent of tick-borne encephalitis (TBE), a severe zoonosis occurring in the Palearctic region mainly transmitted through Ixodes ticks. In Italy, TBEV is restricted to the north-eastern part of the country. This report describes for the first time a case of clinical TBE in a roe deer (Capreolus capreolus L.). The case occurred in the Belluno province, Veneto region, an area endemic for TBEV. The affected roe deer showed ataxia, staggering movements, muscle tremors, wide-base stance of the front limbs, repetitive movements of the head, persistent teeth grinding, hypersalivation and prolonged recumbency. An autopsy revealed no significant lesions to explain the neurological signs. TBEV RNA was detected in the brain by real-time RT-PCR, and the nearly complete viral genome (10,897 nucleotides) was sequenced. Phylogenetic analysis of the gene encoding the envelope protein revealed a close relationship to TBEV of the European subtype, and 100% similarity with a partial sequence (520 nucleotides) of a TBEV found in ticks in the bordering Trento province. The histological examination of the midbrain revealed lymphohistiocytic encephalitis, satellitosis and microgliosis, consistent with a viral etiology. Other viral etiologies were ruled out by metagenomic analysis of the brain. This report underlines, for the first time, the occurrence of clinical encephalitic manifestations due to TBEV in a roe deer, suggesting that this pathogen should be included in the frame of differential diagnoses in roe deer with neurologic disease.

Project description:BackgroundSmall ruminants are important hosts for various tick species and tick-associated organisms, many of which are zoonotic. The aim of the present study was to determine the presence of tick-borne protozoans and bacteria of public health and veterinary significance in goats and wild Siberian roe deer (Capreolus pygargus) from Heilongjiang Province, northeastern China.MethodsThe occurrence of piroplasms, Anaplasma phagocytophilum, A. bovis, A. marginale, A. capra, A. ovis, Ehrlichia spp. and spotted fever group rickettsiae was molecularly investigated and analyzed in 134 goats and 9 free ranging C. pygargus living in close proximity.ResultsPiroplasm DNA was detected in 16 (11.9%) goats and 5 C. pygargus. Sequence analysis of 18S rRNA sequences identified 3 Theileria species (T. luwenshuni, T. capreoli and T. cervi). Four Anaplasma species (A. ovis, A. phagocytophilum, A. bovis and A. capra) were identified in goats and C. pygargus. Anaplasma ovis and A. bovis were detected in 11 (8.2%) and 6 (4.5%) goats, respectively; A. phagocytophilum, A. bovis and A. capra were found in 3, 7 and 3 C. pygargus, respectively. Sequence analysis of 16S rRNA sequences revealed the presence of 5 different genetic variants of A. bovis in goats and C. pygargus, while the analysis of 16S rRNA and gltA sequence data showed that A. capra isolates identified from C. pygargus were closely related to the genotype identified from sheep and Haemaphysalis qinghaiensis, but differed with the genotype from humans. Anaplasma/Theileria mixed infection was observed in 2 (1.5%) goats and 5 C. pygargus, and co-existence involving potential zoonotic organisms (A. phagocytophilum and A. capra) was found in 2 C. pygargus. All samples were negative for A. marginale, Ehrlichia spp. and SFG rickettsiae.ConclusionsThese findings report the tick-borne pathogens in goats and C. pygargus, and a greater diversity of these pathogens were observed in wild animals. Three Theileria (T. luwenshuni, T. capreoli and T. cervi) and four Anaplasma species (A. ovis, A. phagocytophilum, A. bovis and A. capra) with veterinary and medical significance were identified in small domestic and wild ruminants. The contact between wild and domestic animals may increase the potential risk of spread and transmission of tick-borne diseases.

Project description:Background. Understanding species distribution, especially areas of overlapping habitat between sympatric species, is essential for informing conservation through natural habitat protection. New protection strategies should simultaneously consider conservation efforts for multiple species that exist within the same landscape, which requires studies that include habitat overlap analysis. Methods. We estimated the potential habitat of cervids, which are typical ungulates in northern China, using the present locations of red deer (Cervus elaphus; N = 90) and roe deer (Capreolus capreolus; N = 106) in a Maximum Entropy (MaxEnt) model. Our study area was a human-dominated landscape in the Tieli Forestry Bureau located at the southern slope of the Lesser Xing'an Mountains. We grouped 17 environmental predictor variables into five predictor classes (terrain, habitat accessibility, land cover, vegetation feature, and interference), which were used to build habitat suitability models. Results. Habitat accessibility and human interferences were found to have the strongest influence on habitat suitability among the five variable classes. Among the environmental factors, distance to farmland (26.8%), distance to bush-grass land (14.6%), elevation (13.5%), and distance to water source (12.2%) were most important for red deer, distance to farmland (22.9%), distance to settlement (21.4%), elevation (11.6%), and coverage of shrub-grass (8%) were most important for roe deer. Model accuracy was high for both species (mean area under the curve (AUC) = 0.936 for red deer and 0.924 for roe deer). The overlapping habitat comprised 89.93 km(2) within the study area, which occupied 94% of potentially suitable habitat for red deer and 27% for roe deer. Conclusions. In terms of habitat suitability, roe deer showed greater selectivity than red deer. The overlapping habitat was mostly located in the eastern mountains. The southwestern plain was not a suitable habitat for deer because it was close to Tieli City. Regarding management measures, we suggest that priority protection should be given to the potential areas of overlapping deer habitats found in this study.
